Summary A procedure to reconstitute CF0CF1 proteoliposomes by gel filtration through a Sephadex-column pre-equilibrated with valinomycin and potassium is described. Proteoliposomes reconstituted by this procedure catalyze an ATP-induced pH of 2.5 to 3.5 units. pH was measured with either 9-aminoacridine or with the pH indicator pyranine trapped inside the proteoliposomes. CF0CF1 proteoliposomes prepared by conventional techniques catalyzed an ATP-induced formation, but were unable to catalyze an ATP-induced pH even in the presence of valinomycin.The ATP-induced pH was sensitive to uncouplers and energy transfer inhibitors and was increased at low temperatures. It is suggested that ATP-induced pH was observed in these proteoliposomes due to the efficient removal of intravesicular ammonium introduced with the CF0CF1 preparation. The ammonium acted as an internal buffer, and thus prevented an observable pH formation.  相似文献

An assay system was developed in which the effects of inhibitorsof ß-carotene biosynthesis in Dunaliella bardawilcould be tested. Since D. bardawil can be induced to accumulateover 10% of its dry weight as ß-carotene, it is particularlysuitable for such studies. Norflurazon a desaturation inhibitor,caused the accumulation of phytoene, or of phytoene and phytofluene,depending on the concentration employed. J-334, a substituted6-methylpyrimidine which also inhibits desaturation, causedthe accumulation of ß-zeacarotene, -carotene and phytoenein different proportions, depending on the concentration employed.The cyclization inhibitors, nicotine, CPTA and MPTA, severelyaffected the growth and survival of the alga, and their effectscould therefore not be studied directly. However, their actionwas observed indirectly by following the transformation of phytoenein norflurazon-pretreated phytoene-rich algae. Under these conditions,presence of the cyclase inhibitors caused the transformationof phytoene to lycopene, rather than to ß-carotene.The accumulated ß-carotene or the intermediates ß-zeacarotene,lycopene, -carotene, phytofluene and phytoene in D. bardawilwere all composed of two stereoisomers, tentatively assignedas the all-trans stereoisomer (55%) and the 9-cis stereoisomer(45%). This suggests that the isomerization reaction which leadsto the production of the presumed 9-cis isomers occurs earlyin the pathway of carotene biosynthesis, at or before phytoene,with no isomerization during the further transformations leadingto ß-carotene. The purpose of this work was to test whether induction of massive -carotene synthesis in the alga Dunaliella bardawil is triggered by oxygen radicals. The following results were obtained: (i) The induction of -carotene synthesis is preceded by a lag period of about 4 h during which the cells swell and photosynthesis is partially inhibited, (ii) Addition of promoters of oxygen radicals or of azide (an inhibitor of catalase and superoxide dismutase) during the induction period, under conditions which are suboptimal for massive -carotene accumulation, greatly enhances -carotene synthesis, photodegradation of chlorophyll and inhibition of photosynthesis, (iii) High irradiance, which induces massive -carotene accumulation, also induces a high catalase activity. It is suggested that photosynthetically produced oxygen radicals are involved in triggering massive -carotene accumulation in D. bardawil.  相似文献

Adding plastocyanin to plastocyanin-depleted chloroplast particles, restored both their ability to catalyse the photoinduced electron transfer from ascorbate-DCIP to NADP, and to induce the photooxidation of cytochrome f. It is concluded, therefore, that plastocyanin mediates the photoinduced oxidation of cytochrome f, as previously suggested.  相似文献

Dunaliella bardawil Ben-Amotz & Avron accumulates high concentrations of β-carotene when grown under high light intensity. The β-carotene is composed mainly of 9-cis and all-trans β-carotene. Accumulation of β-carotene and an increase in the ratio of the 9-cis to the all-trans isomer are strongly dependent on the light intensity under which the algae are cultivated but are independent of light quality within the photosynthetically active radiation range. Cells grown under continuous red (>645 nm) or white light of 500 W·m?2 reach a value of about 32 pg β-carotene·cell?1 and a ratio of 9-cis to all-trans β-carotene of around 2, whereas cells grown under low red or white light intensity of 25 W·m?2 contain about 3 pg·cell?1 and a ratio of isomers of around 0.3.  相似文献

1. The rate of the Hill reaction and photophosphorylation, and the ratio of ATP produced to the electron flow are shown to be strongly dependent on the solute concentration of the medium.

2. A large part, but not all, of the requirement for MgCl2 or phosphate in photophosphorylation can be replaced by SrCl2 or other solutes.

3. In two-stage photophosphorylation, solutes are required during the light-activation stage.

4. The presence of solutes causes marked changes in the packed volume of the chloroplasts, and their light-scattering properties. These changes are essentially complete within 1 min.

5. The effectiveness of solutes in enhancing the rate of electron transport and photophosphorylation parallels their effectiveness in inducing conformational changes in chloroplasts.

6. It is suggested that the solutes act by inducing a conformational change of the chloroplast structure which is more optimal for electron transfer and coupled phosphorylation.  相似文献

Utilizing oxonol VI as a transmembrane electric potential indicating dye, chloroplasts are shown to develop rapid transient light-induced and ATP-induced potentials. Following the large transient signal smaller steady-state potentials are maintained with either driving system. The ATP-induced potential in the dark depends upon preactivation of the light-triggered ATPase of the chloroplasts, and is inhibited by uncouplers, ionophores such as valinomycin, and energy-transfer inhibitors such as tentoxin, Dio-9 or DCCD. Nigericin increased the signal of both the light- and the ATP-induced reactions. The fact that relatively large transient membrane potentials are induced by either a dark-to-light transition or ATP in the dark provides an explanation for previously observed phenomena such as early kinetics of photophosphorylation and the ATP-induced luminescence.  相似文献

The fluorescent dye 3,3-dipentyloxacarbocyanine (OCC) can be used as a fluorescence probe to measure transmembrane potentials across Rhodospirillum ruburm chromatophore membranes. A reversible fluorescence increase is observed in the light which is sensitive to inhibitors, permeable ions and uncouplers.Partial interchangeability between the electrical potential and the proton concentration gradient has been demonstrated by measurement of the fluorescence increase with OCC and the fluorescence quenching with 9-aminoacridine.OCC fluorescence changes can be induced also in the dark by injection of permeable salts and by rapid pH changes presumably indicating diffusion potentials. Using salt-induced diffusion potentials for calibrating the light signals and with several assumptions, the light-induced potentials were estimated as 170 mV for the maximal signal and 90–110 mV at the steady state.OCC has been shown to apparently increase the electrical conductivity of the chromatophore membrane, a fact which may be relevant to the mechanism of action of this probe.A red shift in the OCC absorption spectrum occurs when mixed with chromatophores, with a difference spectrum maximum at 495 nm. The absorption changes at 495 nm taking place in the light are similar in kinetics to the fluorescence changes. The absorbance spectrum of OCC in organic solvents is red shifted and the extent of the shift depends on the hydrophobicity of the medium. The difference spectrum compared to water in sec-butyl acetaten-hexane (3 : 1, v/v) with a dipole moment of 5 was nearly identical to that of chromatophore-associated dye.The uncoupling properties of OCC at high concentrations and some difficulties in calibration limit the usefulness of this probe for quantitative measurements of transmembrane potentials.  相似文献
