部分酶解酵母高效电击转化研究   总被引:2,自引:0,他引:2  
以酵母质粒YCp50为外源DNA,电击转化部分酶解酵母宿主菌AB1380,转化效率稳定在10~6转化子/μg质粒DNA左右,比不酶解酵母或酵母原生质球作受体的电击转化效率高一个数量级以上,也比PEG介导的酵母原生质球转化高3~5倍,而且适合于大片段DNA如水稻YAC分子的转化。达最佳转化时的有关技术参数为:新接菌种通气培养至细胞密度1×10~8~1.5×10~8个/ml;转化时细胞密度控制在1×10~9~1.5×10~9个/ml;每毫升酶解缓冲液加15u溶菌酶(lyticase),30℃下处理酵母5min进行部分酶解;电击时,电场设置在6.25kV/cm、电容25μF,电击后直接铺板。  相似文献

Xanthocillin具有显著的抗菌活性,结构中含有独特的异腈基。本文通过对蛇足石杉(Huperzia serrata)内生真菌产黄青霉菌(Penicillium chrysogenum)MT-40基因组的测序分析,利用本地BLAST等生物信息学分析工具挖掘具有合成xanthocillin类似物潜力的基因簇,结合米曲霉(Aspergillus oryzae)NSAR1异源表达技术实现基因簇中关键基因的功能鉴定。结果成功从内生真菌P.chrysogenum MT-40中发现一个合成xanthocillin类似物的生物合成基因簇(命名为for),for基因簇中的关键生物合成基因forB编码的异腈基合成酶可以催化合成2-formamido-3-(4-hydroxyphenyl)acrylic acid,基因forG编码的P450酶可以催化2-formamido-3-(4-hydroxyphenyl)acrylic acid的二聚化生成xanthocillin类似物N,N′-(1,4-bis(4-hydroxyphenyl)buta-1,3-diene-2,3-diyl)diformamide。本文研究结果为进一步从真菌中发现xanthocillin类似物提供参考。  相似文献

Y染色体上的短串联重复序列(Y chromosome short tandem repeats, Y-STRs)在法医鉴定,尤其是混合斑中男性成分的检测分析和家系排查中具有重要作用。随着Y-STR试剂盒的不断推出和数据库的建立,国内外针对不同地区和群体的Y-STR多态性研究日益增多,而有关铜川汉族Y-STR研究鲜有报道。基于此,采用YfilerTM Platinum复合扩增试剂盒对陕西铜川汉族669名无关男性个体进行38个Y-STR基因座遗传多态性调查,并探索铜川地区汉族与其他群体之间的遗传关系。调查共检出657种单倍型,单倍型多样性和识别能力分别为0.999 937 345和0.982 1。38个Y-STR基因座共检出428个等位基因,基因多样性值在0.108 9(DYS645)~0.969 9(DYS385)。群体遗传分析中,多维尺度分析(multi-dimensional scaling, MDS)和系统发育树分析结果显示铜川汉族与其他地区的汉族群体遗传距离更近。综上,研究结果中38个Y-STR基因座在铜川汉族群体中具有较高的遗传多态性,适合铜川地区法庭科学应用。  相似文献

Anthocyanin-rich peaches, because of their antioxidant properties and their strong attractiveness to consumers, are increasingly considered in French peach varietal innovation programs that integrate plant genomics and classical breeding. In this study, we describe a new blood-flesh trait identified in the ‘Wu Yue Xian’ peach accession from China. ‘Wu Yue Xian’ exhibits a fully red mesocarp during the later stages of fruit development, both with green midrib leaf and normal growth of the tree. This blood-flesh phenotype clearly differs from that determined by a single recessive locus (bf) in ‘Harrow Blood’, a clone showing blood-flesh in both immature and mature fruit associated with red midrib leaf and reduced tree height. We have then provided genetic evidence that blood-flesh phenotype of ‘Wu Yue Xian’ was controlled by a single dominant locus, designated DBF (Dominant Blood-Flesh), in four successive families derived from this accession. A genetic linkage map of the blood-flesh parent (‘D6090’) of the fourth population was constructed, including 102 SSRs spanning a total distance of 562.3 cM in eight linkage groups. Whereas the bf locus is located to linkage group 4, we mapped DBF to the top of linkage group 5, thus proving that DBF and bf loci are not alleles. Among 64 predicted genes in the DBF region (505 kbp), three genes of the dihydroflavonol-4-reductase family were identified as good candidates for the control of the DBF trait. Furthermore, SSR markers flanking DBF, such as AMPP157 and AMPPG178, supply a good basis to implement marker-assisted selection for this trait.  相似文献

A Gram-negative, oxidase-positive, facultatively anaerobic bacterium, designated strain E20121, was isolated from the digestive tract of a Japanese prawn (Marsupenaeus japonicus) collected from the coastal sea water area of Zhuhai, Guangdong province, China. The new isolate was determined to be closely related to Vibrio ponticus DSM 16217T, having 97.6 % 16S rRNA gene sequence similarity. Phylogenetic analysis based on recA, pyrH and rpoA also showed low levels of sequence similarities (72.6–96.6 %) with all species of the genus Vibrio. A multigene phylogenetic tree using concatenated sequences of the four genes (16S rRNA, rpoA, recA and pyrH) clearly showed that the new isolate is different from the currently known Vibrio species. DNA–DNA hybridization experiments revealed similarity values below 70 % with the closest related species V. ponticus DSM 16217T. Several phenotypic traits enabled the differentiation of strain E20121 from the closest phylogenetic neighbours. The DNA G+C content of strain E20121 was determined to be 47.6 mol % and the major fatty acid components identified were C16:1ω7c and/or C16:1ω6c (39.8 %), C18:1ω7c (13.6 %) and C16:0 (9.6 %). Based on genotypic, phenotypic, chemotaxonomic, phylogenetic and DNA–DNA hybridization analyses, strain E20121 is proposed to represent a novel species of the genus Vibrio for which the name Vibrio zhuhaiensis sp. nov. is proposed. We report herein the synthesis of a series of 7-[3-alkoxyimino-4-(methyl)aminopiperidin-1-yl]quinolone/naphthyridone derivatives. In vitro antibacterial activity of these derivatives was evaluated against representative strains, and compared with ciprofloxacin (CPFX), levofloxacin (LVFX) and gemifloxacin (GMFX). The results reveal that all of the target compounds 19ac and 20 have considerable Gram-positive activity, although they are generally less active than the reference drugs against the Gram-negative strains with some exceptions. Especially, novel compounds 19a2, 19a4 and 19a5 were found to show strong antibacterial activity (MICs: <0.008–0.5 μg/mL) against all of the tested 15 Gram-positive strains including MRSA, LVFX- and GMFX-resistant MRSE, and CPFX-, LVFX- and GMFX-resistant MSSA.  相似文献

为了分析不同肉色类型桃果实发育过程中类胡萝卜素成分的变化特点及其颜色差异形成的因素,选择‘半斤桃’(红肉)、‘图八德’(黄肉)、‘正姬’(白肉)桃品种为试材,采用紫外分光光度计和高效液相色谱法(HPLC)对桃果实类胡萝卜素及其组分含量进行定性定量分析.结果显示:(1)桃果实的主要类胡萝卜素成分为叶黄素、玉米黄素、β-隐黄质、α-胡萝卜素和β-胡萝卜素.(2)随着果实的发育,‘半斤桃’和‘正姬’的类胡萝卜素总含量逐渐降低,而‘图八德’则是先降低后上升,且明显高于前二者;3种类型桃果实的叶黄素均逐渐降低,果实成熟时接近0;玉米黄素呈先降后升的趋势,果实成熟时达到最大值;‘半斤桃’和‘正姬’的β-隐黄质、α-胡萝卜素和β-胡萝卜素呈逐渐下降的趋势,而‘图八德’的β-隐黄质含量先升后微降最后上升、α-胡萝卜素和p-胡萝卜素含量则逐渐增加,且均在果实成熟时达到最大值.(3)‘图八德’成熟果实中的类胡萝卜素总含量达到最大值,其中β-胡萝卜素含量最高,占5个成分之和的75.99%.研究表明,桃果实肉色形成与类胡萝卜素的含量与成分有着紧密的联系,尤其是其中的β-胡萝卜素含量.  相似文献

Plant pathogens have evolved numerous strategies to obtain nutritive materials from their host,and plants in turn have evolved the preformed physical and chemical barriers as well as sophisticated two-tiered immune system to combat pathogen attacks.Genetically, plant resistance to pathogens can be divided into qualitative and quantitative disease resistance,conditioned by major gene(s) and multiple genes with minor effects,respectively.Qualitative disease resistance has been mostly detected in plant defense against biotrophic pathogens,whereas quantitative disease resistance is involved in defense response to all plant pathogens,from biotrophs,hemibiotrophs to necrotrophs.Plant resistance is achieved through interception of pathogen-derived effectors and elicitation of defense response.In recent years,great progress has been made related to the molecular basis underlying host-pathogen interactions.In this review,we would like to provide an update on genetic and molecular aspects of plant resistance to pathogens.  相似文献
