We report the results of NMR studies and computer simulations of potent antagonists reflective of the alpha(IIb)beta(3) receptor-bound conformations. The peptides c[Mpa-(15)N-Arg(1)-(15)N-Gly(2)-(15)N-Asp(3)-(15)N-Phe(4)-(15)N-Arg(5)-Cys]-NH(2) (Phe-Arg analog) (Mpa: 3-mercaptopropionic acid) and c[Mpa-(15)N-Arg(1)-(15)N-Gly(2)-(15)N-Asp(3)-(15)N-Asp(4)-(15)N-Val(5)-Cys]-NH(2) (Asp-Val analog) were subjected to (15)N-edited NMR experiments to study the conformations of these peptides in the absence and in the presence of alpha(IIb)beta(3) receptor. The NMR studies of the Phe-Arg analog, a selective alpha(IIb)beta(3) antagonist, resulted in distinctly different experimental data in the presence and absence of the receptor. The computer simulations for this peptide resulted in one large family of structures consistent with the experimental data. This conformation suggests a type I beta-turn spanning residues Arg(1) and Gly(2) when bound to the receptor and we were able to establish a model for the three dimensional arrangement of the pharmacophores. The studies on the Asp-Val analog, an alpha(v)beta(3) antagonist that binds to the alpha(IIb)beta(3) with moderate affinity, resulted in conformations that are not as well defined as those for the Phe-Arg analog but are consistent with the model established for this analog. These results are important for the design of novel alpha(IIb)beta(3) antagonists.  相似文献

We analyzed background impulse activity of neurons of the supraoptic nucleus of the rat hypothalamus in the course of 15-day-long isolated action of generalized vibrational stimulation and combination of such stimulation with irradiation of the animal’s head with low-intensity extrahigh-frequency (EHF, millimeter-range) electromagmetic waves. The distributions of the neurons by the level of regularity and dynamics of spike trains, separate frequency ranges of impulsation, and pattern of interspike interval (ISI) histograms were estimated. We also calculated the mean frequency of discharges and coefficient of variation of ISIs. A trend toward decreases in the deviations of some parameters of neuronal spike activity generated by supraoptic neurons, which were evident within early time intervals of isolated action of vibration (5 to 10 days), was observed under the influence of EHF electromagnetic irradiation; thus, the latter factor probably exerts a sedative effect. Barbering (fur/whisker trimming, the Dalila effect) is a behaviour-associated hair and whisker loss frequently seen in laboratory rodents, including mice. Here we analyse barbering behaviour in 129S1, NMRI, C57BL/6 and BALB/c mouse strains and some of their F1 hybrids. Our study shows that barbering in mice, depending on their genotype, is a complex behaviour with several distinct contexts or domains. We observed social (dominant) barbering in NMRI and C57BL/6 mice, sexual over-grooming in 129S1 and C57BL/6 mice, maternal barbering in lactating 129S1 and C57BL/6 mice, and stress-evoked barbering in F1 (NMRIx129S1) hybrids. In contrast, aggressive BALB/c mice and their F1 progeny do not use barbering in their behaviour. We suggest that barbering may be an important complex multi-domain behaviour sensitive to various manipulations, and represent a useful index in neurobehavioural research.  相似文献

Hypervitaminosis vitamin D3 has been recently implicated in premature aging through the regulation of 1alpha hydroxylase expression by klotho and fibroblast growth factor-23 (Fgf-23). Here we examined whether the lack of hormonal function of vitamin D3 in mice is linked to aging phenomena. For this, we used vitamin D3 receptor (VDR) “Tokyo” knockout (KO) mice (fed with a special rescue diet) and analyzed their growth, skin and cerebellar morphology, as well as overall motor performance. We also studied the expression of aging-related genes, such as Fgf-23, nuclear factor kappaB (NF-kappaB), p53, insulin like growth factor 1 (IGF1) and IGF1 receptor (IGF1R), in liver, as well as klotho in liver, kidney and prostate tissues. Overall, VDR KO mice showed several aging related phenotypes, including poorer survival, early alopecia, thickened skin, enlarged sebaceous glands and development of epidermal cysts. There was no difference either in the structure of cerebellum or in the number of Purkinje cells. Unlike the wildtype controls, VDR KO mice lose their ability to swim after 6 months of age. Expression of all the genes was lower in old VDR KO mice, but only NF-kappaB, Fgf-23, p53 and IGF1R were significantly lower. Since the phenotype of aged VDR knockout mice is similar to mouse models with hypervitaminosis D3, our study suggests that VDR genetic ablation promotes premature aging in mice, and that vitamin D3 homeostasis regulates physiological aging.  相似文献

The methods for preparation of fused triazolo[4,3-c]- and triazolo[1,5-c]pyrimidines were developed. The Dimroth rearrangement of these systems was studied. Pharmacological investigation of the synthesized compounds was conducted in known tests, such as antagonism with subcutaneous administration of corazole and the open-field test. The rotating rod method was used to assess the neurotoxicity. Neurotropic properties were found in many derivatives of triazolopyrimidine. They, like diazepam, prevent the occurrence of clonic twitching and corazole-induced clonic convulsions in animals. All selected compounds as well as diazepam exhibit an anxiolytic effect.  相似文献

We analyzed the background impulse activity (BIA) generated by neurons of the rat hypothalamic supraoptic nucleus in the norm and under conditions of long-lasting vibrational stimulation (exposure 5, 10, or 15 days). Distributions of neurons by the level of regularity, dynamics of discharge trains, form of histograms of interspike intervals (ISIs), as well as distributions of neurons by the BIA frequency ranges, were studied. We also calculated the mean frequency of impulsation of the neurons under study and the coefficient of variation of ISIs. After vibrational influences, we found modifications of both the internal structure of the recorded spike trains and the mean frequency of impulsation within the entire studied group and different frequency subgroups. The possibility of enhancing heterologous gene expression in mammalian cells by the introduction of an intron in 3′ untranslated region (UTR) was investigated. To this end, a fragment of human betaglobin gene with intron 2 and flanked exon regions was introduced into the vector-encoding green fluorescent protein TagGFP2 after the TagGFP2 stop-codon (Int+). The distance between the stop-codon and the exon junction was 35 nucleotides. It ensured that Int+ mRNA was resistant to degradation by nonsense mediated decay (NMD) machinery. A control vector Intcontained corresponding intronless sequence of the beta-globin mRNA. On the same plasmid, the second gene encoded far-red fluorescent protein Katushka was used to normalize fluorescence for transfection efficiency and expression level in individual cells. Transiently transfected HEK293T cells were analysed by flow cytometry. It was shown that cells transfected with plasmid carrying the Int+ gene possess 1.8 ± 0.2 fold higher green fluorescence compared to Intcells. The observed effect was used to enhance expression of destabilized variants of yellow fluorescent protein TurboYFP-dest with high degradation rate in mammalian cells. We believe that introduction of beta-globin intron in the 3′-UTR of the chimeric gene can be used to enhance its expression and may be advantageous in some cases when usage of 5′ UTR intron is inappropriate.  相似文献

The effects of daily and weekly academic loads on the psychological and cardio-and hemodynamic characteristics of students were analyzed. The self-estimated state of health, activity, and mood were evaluated with the use of the HAM (Health-Activity-Mood) questionnaire; the introversion and extraversion, neuroticism, and intelligence of the subjects were determined according to Eysenck; and the self-estimate the personal anxiety according to Spielberger, was evaluated. The main hemodynamic parameters (heart rate and arterial blood pressure) were measured and the systolic and minute blood volumes were calculated. The comparison of the dynamics of the parameters studied showed that there were two types of response of the students’ cardio-and hemodynamic parameters to academic load: sympathetic and parasympathetic. The parasympathetic response was found to be more adequate because the sympathetic response entailed the risk of overstrain and adaptation failure.  相似文献

Vitamin D insufficiency has been reported to be associated with increased blood cholesterol concentrations. Here we used two strains of VDR knock-out (VDR-KO) mice to study whether a lack of vitamin D action has any effect on cholesterol metabolism. In 129S1 mice, both in male and female VDR-KO mice serum total cholesterol levels were significantly higher than those in wild type (WT) mice (20.7% (P = 0.05) and 22.2% (P = 0.03), respectively). In addition, the serum high-density lipoprotein-bound cholesterol (HDL-C) level was 22% (P = 0.03), respectively higher in male VDR-KO mice than in WT mice. The mRNA expression levels of five cholesterol metabolism related genes in livers of 129S1 mice were studied using quantitative real-time PCR (QRT-PCR): ATP-binding cassette transporter A1 (ABCA1), regulatory element binding protein (SREBP2), apolipoprotein A-I (ApoAI), low-density lipoprotein receptor (LDLR) and liver X receptor beta (LXRβ). In the mutant male mice, the mRNA level of ApoAI and LXRβ were 49.2% (P = 0.005) and 38.8% (P = 0.034) higher than in the WT mice. These changes were not observed in mutant female mice, but the female mutant mice showed 52.5% (P = 0.006) decrease of SREBP2 mRNA expression compared to WT mice. Because the mutant mice were fed with a special rescue diet, we wanted to test whether the increased cholesterol levels in mutant mice were due to the diet. Both the WT and mutant NMRI mice were given the same diet for 3 weeks before the blood sampling. No difference in cholesterol or in HDL-C between WT and mutant mice was found. The results suggest that the food, gender and genetic background have an effect on the cholesterol metabolism. Although VDR seems to regulate some of the genes involved in cholesterol metabolism, its role in the regulation of serum cholesterol seems to be minimal.  相似文献

Effects of examination stress on psychological characteristics and indices of the autonomic nervous system were studied in pupils of the eighth and tenth grades of high schools providing differential (test group, Neuron gymnasium) and general education (control group). General health state and levels of activity and mood (feeling, activity, mood; FAM) and neuroticism (according to Eysenck) were determined in both groups. Main hemodynamic indices (heart rate and blood pressure) were measured, and mathematical analysis of the heart rate by was performed according to Baevskii. Comparative analysis of psychophysiological tests and heart rate changes in the control and test groups points to a high tension in the cardiac rhythm regulation mechanisms and strong anxiety in schoolchildren in the differential education school. The character and depth of preexamination stress depended on the type of autonomic control of cardiac activity. Schoolchildren with sympathicotonic cardiac rhythm regulation showed the strongest emotional tension. Schoolchildren with the normotonic type of regulation were the most calm.  相似文献
