It has been suggested that angiogenesis modulates adipogenesis and obesity. This study was undertaken to determine whether ALS-L1023 (ALS) prepared by a two-step organic solvent fractionation from Melissa leaves, which exhibits antiangiogenic activity, can regulate adipose tissue growth. The effects of ALS on angiogenesis and extracellular matrix remodeling were measured using in vitro assays. The effects of ALS on adipose tissue growth were investigated in high fat diet-induced obese mice. ALS inhibited VEGF- and bFGF-induced endothelial cell proliferation and suppressed matrix metalloproteinase (MMP) activity in vitro. Compared to obese control mice, administration of ALS to obese mice reduced body weight gain, adipose tissue mass and adipocyte size without affecting appetite. ALS treatment decreased blood vessel density and MMP activity in adipose tissues. ALS reduced the mRNA levels of angiogenic factors (VEGF-A and FGF-2) and MMPs (MMP-2 and MMP-9), whereas ALS increased the mRNA levels of angiogenic inhibitors (TSP-1, TIMP-1, and TIMP-2) in adipose tissues. The protein levels of VEGF, MMP-2 and MMP-9 were also decreased by ALS in adipose tissue. Metabolic changes in plasma lipids, liver triglycerides, and hepatic expression of fatty acid oxidation genes occurred during ALS-induced weight loss. These results suggest that ALS, which has antiangiogenic and MMP inhibitory activities, reduces adipose tissue mass in nutritionally obese mice, demonstrating that adipose tissue growth can be regulated by angiogenesis inhibitors. 相似文献

Anthers of Capsicum annuum L. were cultured on Murashige and Skoog (MS) medium containing 0.1 mg l−1 NAA and 0.1 mg l−1 kinetin. Inoculated anthers were subjected to 31 °C and development of microspores in anthers of varying stages was observed
cytologically using 4′-6-diamidino-2-phenylindol-2HCl (DAPI). Pepper was characterized by a strong asynchrony of pollen development
within a single anther. Percentage of pollen at different stages changed with the culture period, and the proportion of dead
pollen increased drastically from day 2 after culture. Microspores that were cultured at the late-uninucleate stage followed
one of two developmental pathways. In the more common route, the first sporophytic division was asymmetric and produced what
appeared to be a typical bicellular pollen. Embryogenic pollen was formed by repeated divisions of the vegetative nucleus.
In the second pathway, which occurred in fewer microspores, the first division was symmetric and both nuclei divided repeatedly
to form embryogenic pollen. In early-bicellular pollen, sporophytic pollen was produced through division of the vegetative
nucleus. In mid-bicellular pollen, the generative nucleus may undergo division to produce two or more sperm-like nuclei. However,
division of the generative nucleus alone to form the embryo was never observed. The anther stage optimal for embryo production
contained a large proportion (>75%) of early-binucleate pollen. Associations were found among the percentage of early-binucleate
pollen, the frequency of embryogenic multinucleate pollen, and the yield of pollen embryos. 相似文献

Ginsenoside Rf, a component of ginseng, regulates lipoprotein metabolism through peroxisome proliferator-activated receptor alpha 总被引:2,自引:0,他引:2
We investigated whether ginseng regulates lipoprotein metabolism by altering peroxisome proliferator-activated receptor alpha (PPARalpha)-mediated pathways, using a PPARalpha-null mouse model. Administration of ginseng extract, ginsenosides, and ginsenoside Rf (Rf) to wild-type mice not only significantly increased basal levels of hepatic apolipoprotein (apo) A-I and C-III mRNA compared with wild-type controls, but also substantially reversed the reductions in mRNA levels of apo A-I and C-III expected following treatment with the potent PPARalpha ligand Wy14,643. In contrast, no effect was detected in the PPARalpha-null mice. Testing of eight main ginsenosides on PPARalpha reporter gene expression indicated that Rf was responsible for the effects of ginseng on lipoprotein metabolism. Furthermore, the inhibition of PPARalpha-dependent transactivation by Rf seems to occur at the level of DNA binding. These results demonstrate that ginseng component Rf regulates apo A-I and C-III mRNA and the actions of Rf on lipoprotein metabolism are mediated via interactions with PPARalpha. 相似文献

We investigated whether fenofibrate improves lipid metabolism and obesity in female ovariectomized (OVX) or sham-operated (SO) low density lipoprotein receptor-null (LDLR-null) mice. All mice fed a high-fat diet exhibited increases in serum triglycerides and cholesterol as well as in body weight and white adipose tissue (WAT) mass compared to mice fed a low fat control diet. However, fenofibrate prevented high-fat diet-induced increases in body weight and WAT mass in female OVX LDLR-null mice, but not in SO mice. In addition, administration of fenofibrate reduced serum lipids and hepatic apolipoprotein C-III mRNA while increasing the mRNA of acyl-CoA oxidase in both groups of mice, however, these effects were more pronounced in OVX LDLR-null mice. The results of this study provide first evidence that fenofibrate improves both lipid metabolism and obesity, in part through PPARalpha activation, in female OVX LDLR-null mice. 相似文献

Compound K, a novel ginsenoside metabolite formed by intestinal bacteria, is shown to inhibit angiogenesis and matrix metalloproteinase (MMP) activities. Since growth and development of adipose tissue are thought to require adipogenesis, angiogenesis, and extracellular matrix remodeling, we investigated whether compound K inhibits adipocyte differentiation and its potential mechanisms. Treatment of 3T3-L1 adipocytes with compound K inhibited lipid accumulation and expression of adipocyte-specific genes (i.e., PPARγ, leptin, aP2, and C/EBPα). Compound K decreased mRNA levels of angiogenic factors (i.e., VEGF-A and FGF-2) and MMPs (i.e., MMP-2 and MMP-9), whereas it increased mRNA levels of angiogenic inhibitors (TSP-1, TIMP-1, and TIMP-2) in 3T3-L1 cells. MMP-2 and MMP-9 activities were also decreased in compound K-treated cells. These results demonstrate that compound K effectively inhibited adipogenesis and that this process may be mediated in part through changes in the expression of genes involved in angiogenesis and MMP system. Thus, by suppressing adipogenesis, compound K likely has therapeutic potential for the treatment of obesity and related disorders. 相似文献

Inhibition of the actions of peroxisome proliferator-activated receptor alpha on obesity by estrogen
Objective: To determine whether the major ovarian factor estrogen modulates peroxisome proliferator‐activated receptor (PPAR) α actions on obesity and to investigate the mechanism by which estrogen regulates PPARα actions. Research Methods and Procedures: Female ovariectomized mice were randomly divided into four groups (n = 8/group). After they were treated with combinations of high fat, fenofibrate (FF), or 17β‐estradiol (E) for 13 weeks, variables and determinants of obesity and lipid metabolism were measured using in vivo and in vitro approaches. Results: When female ovariectomized mice were given a high‐fat diet with either FF or E, body weight gain and white adipose tissue mass were significantly reduced and serum lipid profiles were improved compared with control mice fed a high‐fat diet alone. When mice were concomitantly treated with FF and E, however, E reversed the effects of FF on body weight gain, serum lipid profiles, and hepatic PPARα target gene expression. Consistent with the in vivo data, E not only decreased basal levels of PPARα reporter gene activation but also significantly decreased Wy14,643‐induced luciferase reporter activity. In addition, inhibition of PPARα functions by E did not seem to occur by interfering with the DNA binding of PPARα. Discussion: Our results demonstrate that in vivo and in vitro treatment of estrogen inhibited the actions of FF‐activated PPARα on obesity and lipid metabolism through changes in the expression of PPARα target genes, providing evidence that FF does not regulate obesity in female mice with functioning ovaries. 相似文献

We report high frequencies of embryo production and plant regeneration through isolated microspore culture of hot pepper (Capsicum annuum L.). Microspores cultured in modified NLN medium (NLNS) divided and developed to embryos. Globular and heart-shaped embryos
were observed from 3 weeks after the beginning of culture, and many embryos reached the cotyledonary stage after 4 weeks of
culture. These cotyledonary embryos developed to plantlets after transfer to solid B5 basal medium. We also optimized conditions
for embryo production by varying the pretreatment media, the carbon sources, and culture densities. Heat shock treatment in
sucrose-starvation medium was more effective than in B5 medium. Direct comparisons of sucrose and maltose as carbon sources
clearly demonstrated the superiority of sucrose compared to maltose, with the highest frequency of embryo production being
obtained in 9% (w/v) sucrose. Microspore plating density was critical for efficient embryonic induction and development, with
an optimal plating density of 8 × 104–10 × 104/ml. Under our optimized culture conditions, we obtained over 54 embryos, and an average of 5.5 cotyledonary embryos when
10 × 104 microspores were grown on an individual plate. 相似文献
