The six loci, beta-globin (HBBC), parathyroid hormone (PTH), oncogene c-Ha-ras-1 (HRAS1), insulin (INS), calcitonin (CAL) and catalase (CAT) loci, have been mapped to 11p in the order: CAT-CAL-PTH-HBBC-(HRAS1-INS). The purpose of the current study was to examine the linkage relationships, especially the multipoint relationships of these loci in detail. In the 18 families studied, a significant sex difference in recombination was found for the HBBC: HRAS1 linkage with more recombination in the male parent than the female parent (22 vs. 2%). The results of the multipoint analyses provided further evidence for the order CAT-CAL-PTH-HBBC-(HRAS1-INS); however, the order of the last two tightly linked loci is still not clear. 相似文献

A device to measure fluorescein in tissue has been constructed from standard photographic equipment--an electronic strobe and a flashmeter both covered with interference filters. The instrument works well in the light and need not touch the area being measured, an advantage over existing fluorometers. The instrument has been used to measure the amount of dye in flaps in rats, pigs, and three humans. The results revealed that the amount of dye in a freshly made flap was rarely as much as in normal skin, and skin with less than 20 percent of the dye of control areas usually sloughed, although there were exceptions. In the future the instrument will be improved, and its readings will be compared to those obtained from radioactive microspheres, the present "gold standard" of techniques to measure vascularity. The instrument can be used to estimate the blood supply to any tissue and seems to be as reliable as the dermofluorometers already on the market. 相似文献

Summary 1. Studies of the broad-leafed turtle grass,Thalassia testudinum
König, have revealed a diverse range of fungal infestation different in generic composition and dynamics of attack from that found on submerged wood. Certain of the fungi, notably the AscomyceteLindra thalassiae, initiate considerable degradation of leaf tissue and show a developmental cycle in nature related to the physiological state of the host plant.2. Use of fungal-cellulose mats as a trapping substrate has been extremely effective for discernment of ecologically significant shifts in nematode concentrations, especially those of the omnivorous species,Metoncholaimus scissus.3. Patterns of activity ofM. scissus, as well as those of various foliicolous nematodes, suggest that loci of organic material, such as fungal infested leaves and decaying plant tissue, significantly affect biological activity of these animals.4. Laboratory analysis of degraded cotton cellulose filters show a striking incidence of fungal reproduction of the ascomycetous fungusLulworthia, along with development of a considerable associated nematode fauna, especially species ofViscosia (V. macramphida) andLeptolaimus (L. plectoides). Successional patterns in nematode development are noted with continued degradation of the cotton cellulose matrix.

Kurzfassung Untersuchungen am SeegrasThalassia testudinum König haben ergeben, daß sich hier Pilzinfektionen hinsichtlich der Komposition der beteiligten Gattungen und der Dynamik des Befalls von den am untergetauchten Holz festgestellten Infektionen unterscheiden. Bestimmte Pilze, insbesondere der AscomycetLindra thalassiae, leiten eine erhebliche Degradation des Blattgewebes ein und zeigen einen Entwicklungszyklus, welcher in Beziehung steht zum physiologischen Zustand der Wirtspflanze. Die Anwendung von Pilz-Zellulose-Matten als Einfangsubstrat war außerordentlich erfolgreich für das Erkennen ökologisch signifikanter Verschiebungen in den Nematodenkonzentrationen, insbesondere bei der omnivoren ArtMetoncholaimus scissus. Die Aktivitätsmuster vonM. scissus — ebenso wie die verschiedener foliicolöser Nematoden — deuten darauf hin, daß pilzinfizierte und zerfallende Pflanzenteile in entscheidendem Maße die biologische Aktivität dieser Tiere beeinflussen. Laboratoriumsanalysen degradierter Wollzellulosefilter lassen eine überraschend starke Vermehrung des AscomycetenLulworthia erkennen und gleichzeitig die Entwicklung einer beachtlichen Fauna assoziierter Nematodenarten, insbesondere vonViscosia macramphida undLeptolaimus plectoides. Im Verlaufe der weiteren Degradation der Wollzellulosematrix kommt es bei der Nematodenfauna zu entsprechenden Sukzessionen.

One stallion and 54 mares were used in an experiment to evaluate the effect of postbreeding uterine lavage on pregnancy rate in mares. All mares were inseminated with 250 x 10(6) progressively motile sperm every other day during estrus until detection of ovulation. Mares (n = 18) were randomly assigned to one of three treatment groups: 1) no postbreeding uterine lavage (control); 2) uterine lavage at 0.5 h postbreeding; or 3) uterine lavage at 2 h postbreeding. A dilute solution of povidone-iodine (PIS; 0.05%) previously determined to render spermatozoa immotile in vitro was used to lavage the mare uteri. One liter PIS, prewarmed to 40 degrees C, was used for each lavage. Pregnancy status of mares was determined at 21 d and 36 d post ovulation, using transrectal ultrasonography. The pregnancy rate of Group 1 (66.7%) was higher than that of Group 2 (22.2%; P<0.05) or Group 3 (33.3%); P<0.10). The pregnancy rates of Groups 2 and 3 were similar (P>0.70). Evaluation of endometrial biopsies obtained from a separate set of mares (n = 3) on Day 6 post ovulation, both before and after uterine lavage, revealed no difference in the accumulation of inflammatory cells, suggesting adverse effects of lavage on fertility may have been due to excessive removal of spermatozoa from the uterus during the lavage process or damage to oviductal spermatozoa. 相似文献

Human papillomaviruses (HPVs) are implicated in the etiology of anogenital cancers. Expression of the HPV E6 and E7 oncoproteins is believed to contribute to the carcinogenic process. Progressive loss of the ability to differentiate and resistance to the growth-inhibitory effects of endogenous signals also appear important in multistep tumorigenesis. Transforming growth factor beta1 (TGF-beta1) is a potent growth inhibitor for a variety of cultured cells. There have been conflicting reports on the ability of TGF-beta1 to inhibit the growth of HPV-positive keratinocytes in monolayer cultures. We have employed the organotypic (raft) tissue culture system, which more accurately mimics the in vivo cellular environment and architecture. We have investigated the TGF-beta1 response of HPV-positive keratinocytes derived from neoplastic cervical biopsies. Growth of these cell lines as raft tissues showed that many were altered in the ability to stratify and synthesize differentiation-specific proteins. When the organotypic tissues were treated with TGF-beta1, a more complete differentiation of the keratinocytes was induced. Treatment with 12-0-tetradecanoylphorbol-13-acetate gave similar results. TGF-beta1 treatment of HPV-positive raft epithelia led to a dose-dependent increase in E7 RNA expression in contrast to results from previous studies with monolayer cultures. Furthermore, TGF-beta1 interfered with the proliferation of HPV-positive cell lines grown in monolayer cultures. Our results suggest that loss of the ability to express markers of differentiation, a characteristic of malignancy, is a two-step process. Shewanella putrefaciens is a facultatively anaerobic bacterium in the gamma group of the proteobacteria, capable of utilizing a wide variety of anaerobic electron acceptors. An examination of its cytochrome content revealed the presence of a tetraheme, low-redox-potential (E'o = -233 mV), cytochrome c-type cytochrome with a molecular mass of 12,120 Da and a pI of 5.8. The electron spin resonance data indicate a bis-histidine coordination of heme groups. Reduction of ferric citrate was accompanied by oxidation of the cytochrome. The biochemical properties suggested that this protein was in the cytochrome c3 group, which is supported by N-terminal sequence data up to the first heme binding site. 相似文献
