A bioassay-guided phytochemical analysis of the triterpene saponins from under ground parts of Gypsophila arrostii var. nebulosa allowed the isolation of two triterpene saponins; nebuloside A, B based on gypsogenin and quillaic acid aglycone. Two new oleanane type triterpenoid saponins (nebuloside A, B) and three known saponins (13) were isolated from the root bark of Gypsophila arrostii var. nebulosa. The structures of the two new compounds were elucidated as 3-O-β-d-galactopyranosyl-(1→2)-[β-d-xylopyranosyl-(1→3)]-β-d-glucuronopyranosyl quillaic acid 28-O-β-d-glucopyranosyl-(1→3)-[β-d-xylopyranosyl-(1→3)-β-d-xylopyranosyl-(1→4)]-α-l-rhamnopyranosyl-(1→2)-β-d-fucopyranosyl ester (nebuloside A) and 3-O-β-d-xylopyranosyl-(1→3)-[β-d-galactopyranosyl(1→3)-β-d-galactopyranosyl-(1→2)]-β-d-glucuronopyranosyl gypsogenin 28-O-β-d-glucopyranosyl-(1→3)-[β-d-xylopyranosyl-(1→3)-β-d-xylopyranosyl-(1→4)]-α-l-rhamnopyranosyl-(1→2)-β-d-fucopyranosyl ester (nebuloside B), on the basis of extensive spectral analysis and chemical evidence. Nebuloside A and B showed toxicity enhancing properties on saporin a type-I RIP without causing toxicity by themselves at 15 μg/mL.  相似文献

Saponinum album (Merck), which is a crude mixture of saponins from Gypsophila paniculata L., was shown to improve the anti cancer therapy when used in vivo in combination with saporin-based targeted toxins. Unfortunately saponinum album cannot be used for further development since Merck has ceased its production in the 1990s. As pure saponins are mandatory for use in medical purposes we developed a convenient method for saponin isolation directly from the roots of Gypsophila paniculata L. The developed method is rapid, cheap and scaling up is also possible. By combining dialysis and HPLC three saponins were isolated in a one-step procedure. Chemical structures of the purified saponins were characterized by extensive one and two-dimensional NMR-spectroscopy and by using ESI-TOF-MS. The biological activities of the purified saponins were also investigated. The method presented herein enabled a rapid and cheap isolation of saponins for tumour therapy.  相似文献

Saponins are secondary plant compounds. They have a triterpenoid or steroidal backbone. Sugars are attached to one or more points of this structure, forming chains that can be branched. This appearance leads to amphiphilic properties giving saponins the ability to interact with both lipophilic and hydrophilic structures. The surfactant behavior lets them lower the surface tension in aqueous solutions and form micelles when reaching the critical micelle concentration (cmc). It also lets them interact with biologic membrane layers that usually consist of phospholipids and cholesterol. This action may perturb the membrane and its function leading to membrane perforation or complete lysis. Thus saponins are also known for their cytotoxicity and membranolytic, respectively hemolytic features. In our studies we wanted to answer the question if there is a correlation between the unspecific detergent behavior when lowering the surface tension and the ability to perforate cell membranes and to act cytotoxic. Do saponins showing a considerable reduction in the surface tension also reveal an evident cytotoxicity or/and a marked cell membrane perforation?We tested a variety of saponins with distinct structures. The reduction in the surface tension and the cmc were analyzed on a tensiometer using the Wilhelmy plate method. The general cytotoxicity was determined in a cell model by DNA quantification. The cell membrane toxicity or membrane perforation was explored in a cell model by quantification of the leakage of the intracellular enzyme lactate dehydrogenase (LDH).The experiments revealed a correlation between the membrane toxicity and the reduction in surface tension.  相似文献

Extracellular administration of a membranepermeable model peptide containing the tripeptide sequence,SLV, at the C-terminus to human endothelial and kidney cellsresulted in an induction of caspase-8 (FLICE), the apicalenzyme of the apoptosis cascade. The unmodified or N-terminally SLV-tagged peptide had no effect, therebyeliminating an unspecific induction of apoptosis as the causeof the caspase activity observed. Drastic alterations ofprimary structure and structure forming properties of thecarrier peptide did not significantly influence the caspase-8inducing activity of the C-terminal SLV-tag, supportingprevious findings that translocation into the cell interior isa more general ability of peptides.  相似文献

A novel soluble non-opioid dynorphin A-binding factor (DABF) was identified and characterized in neuronal cell lines, rat spinal cord, and brain. DABF binds dynorphin A(1-17), dynorphin A(2-17), and the 32 amino acid prodynorphin fragment big dynorphin consisting of dynorphin A and B, but not other opioid and non-opioid peptides, opiates, and benzomorphans. The IC50 for dynorphin A(1-17), dynorphin A(2-17), and big dynorphin is in the 5-10 nM range. Using dynorphin A and big dynorphin fragments a binding epitope was mapped to dynorphin A(6-13). DABF has a molecular mass of about 70 kDa. SH-groups are apparently involved in the binding of dynorphin A since p-hydroxy-mercuribenzoic acid inhibited this process. Upon interaction with DABF dynorphin A was converted into Leu-enkephalin, which remained bound to the protein. These data suggest that DABF functions as an oligopeptidase that forms stable and specific complexes with dynorphin A. The presence of DABF in brain structures and other tissues with low level of prodynorphin expression suggests that DABF as an oligopeptidase may degrade other peptides. Dynorphin A at the sites of its release in the CNS may attenuate this degradation as a competitor when it specifically binds to the enzyme.  相似文献

In view of neurotoxic properties of tetrahydroisoquinolines (TIQ's) there are open questions also in regard to the disturbance of the blood-brain barrier. Because endothelial cells are an important element of this barrier the present study was designed to assess the influence of salsolinol (a TIQ formed by condensation of dopamine and acetaldehyde) on cultivated endothelial cells by physiological, biochemical and morphological investigations. For the investigations we used aortic endothelial cells because of a variety of similarities in physiology and biochemistry to brain capillary endothelial cells. Cytotoxic effects estimated by cell counting after 72 h treatment with salsolinol (IC50=38 mol/l) were possibly caused by mitochondrial damages. Already after 2 h severe ultrastructural alterations of many mitochondria could be observed. The respiration activity of the cells was always inhibited after treatment with salsolinol for some hours. The damage of the mitochondria by salsolinol was not connected with inhibition of the activity of succinate dehydrogenase and cytochrome c+c1. Nevertheless the damages of mitochondrial integrity support the hypothesis that the neurotoxic effect of salsolinol is primarily caused by damaging the endothelial cells associated with a disturbance of blood-brain barrier.  相似文献
