The synthesis of the new radioprotective compounds ATB (I, 2-allylthioureidobenzoic acid), PTB (II, 2-phenylthioureidobenzoic acid), A-2-PTU (III, N-allyl-N"-2-pyridylthiourea), and P-2-PTU (IV, N-phenyl-N"-2-pyridylthiourea) and their influence on biological and genetic effects of gamma rays was studied. In result of investigations it must be noted that PTB displayed radioprotective effect as a result of which more plants in M1 germination and survive in M2 of the induced mutations is increased. The cytological analysis reveals that the studied substance (PTB) decreases chromosome aberration in meristem cells of pea roots almost twice as a result of postirradiation treatment. The effect of A-2-PTU in the experiments with peas greatly depends on the dose of irradiation, i.e., on the degree of damaging of the processes of cell restoration and the possibility of their partial restoration after the treatment with the protector. The results obtained suggest that chemical compounds of N,N"-disubstituted thiourea group (A-2-PTU and P-2-PTU) exert strong radioprotective effect in the experiments with peas. This is of great importance to modern radiobiology and radiation mutagenesis and also to protect hereditary structures against radiation.  相似文献

The review presents our research on the influence of novel thiourea compounds on the biological and genetic effect of gamma-rays using in vivo and in vitro systems of pea. Some novel disubstituted thioureas: o-allylthioureidobenzoic acid (ATB); o-phenylthioureidobenzoic acid (PTB); N-allyl-N'-2-pyridylthiourea (A-2-PTU); N-phenyl-N'-2-pyridylthiourea (P-2-PTU) and 1,4-allylthioureidosalicylic acid (ATUS) were examined. Pea (Pisum sativum L.) seeds from five varieties were used. Experiments in vivo and in vitro were carried under laboratory, greenhouse and field conditions. The data revealed the PTB radioprotective effect demonstrated by: reduction of chromosome aberrations by 2 folds; 50% increase of germinating and surviving plants in M1; twice higher frequency of induced mutations in M2 generation relative to irradiation without PTB treatment; decreasing the level of induced radiation suppression leading to favorable effect on the initial stem and root development of pea. ATB radioprotective effect was demonstrated in vitro by: 25-35% stimulation of organogenesis; by 20-50% increase in bud formation; by 25% stimulation of growth. The effect of A-2-PTU and P-2-PTU depended on the irradiation dose. The protective effect of A-2-PTU is more pronounced at lower irradiation dose, while the effect of P-2-PTU is more pronounced at higher irradiation dose. ATUS, opposite to the other compounds, revealed radiosensibilizing effect by: 16-27% increase in lethality caused by gamma-rays leading to lower number of germinating and surviving plants in M1; 50% decrease in the number of induced mutations in M2 generation; limiting the types of induced mutations at the higher irradiation dose. As a result of the experiments useful mutation forms were obtained, characterized with: earliness, lodging and disease resistance; higher productivity.  相似文献

The complexation between copper(II) and the antihypertensive drug oxprenolol (HOxp) was studied both in methanol and slightly alkaline aqueous media at Cu:HOxp molar ratio from 1:1 to 1:10. Copper(lI) forms two types of complexes-a mononuclear violet one, CuOxp2, with bidentately bound ligands and a green dimeric one, Cu2Oxp2Cl2, in which the two Cu(II) centres are linked by the ligand through oxygen bridges. The crystal structure of the Cu2Oxp2Cl2 complex consists of two crystallographically non-equivalent centrosymmetric copper dimers. Each copper atom is four-coordinated in a distorted square-planar environment. The Cu2O2 structural core is characterized by a Cu1-O1-Cu1' angle of 104.15(13)degrees (Cu2-O2-Cu2' 104.30(13) degrees) and a relatively short Cu1-Cu1' separation of 3.026(1) A (Cu2-Cu2'-3.023(1) A). Magnetic susceptibility and EPR measurements indicate an antiferromagnetic coupling of the copper(II) centers.  相似文献

During the last 2 decades it was proposed that atherogenesis was closely related to the homeostasis of homocysteine (hCys) and/or copper. We hypothesized that the physiological action of hCys may be connected with its ability to form complexes with Cu. Our results showed the presence of two different Cu-hCys complexes. At a molar ratio Cu:hCys 1:1, a blue complex most probably consistent with a tentative dimeric Cu(II)(2)(hCys)(2)(H(2)O)(2) formula was formed, with tetrahedral Cu coordination and anti-ferromagnetic properties. The redox processes between Cu(II) and hCys, in a molar ratio > or =1:3 led to formation of a second yellow Cu(I)hCys complex. Both Cu-hCys complexes affected the metabolism of extracellular thiols more than hCys alone and inhibited glutathione peroxidase-1 activity and mRNA abundance. The biological action of hCys and Cu-hCys complexes involved remodeling and phosphorylation of focal adhesion complexes and paxillin. The adhesive interactions of monocytes with an endothelial monolayer led to the redistribution of both paxillin and F-actin after all treatments, but the diapedesis of monocytes through endothelial cell monolayer was both greater and faster in the presence of the tentative Cu(II)(2)(hCys)(2)(H(2)O)(2) complex. Together, these observations suggest that Cu-hCys complexes actively participate in the biochemical responses of endothelial cells that are involved in the aethiopathogenesis of atherosclerosis.  相似文献

This paper compares the repair of DNA single strand breaks (ssb) induced by γ-radiation in two strains ofChlamydomonas reinhardtii (137C/+/ and UVS-I) and three lines ofPisum sativum (NN 131, 198, 140) differing in the degree of radioresistance. DNA ssb in cells exposed to γ-rays (50, 100, 200, 500 Gy) were measured by electrophoresis and alkaline unwinding method with subsequent chromatography on hydroxyapatite immediately after irradiation and after 30 min of post-irradiation incubation at 25°C. An increase of double-strand DNA (in%) was found in cells after 30 min post-irradiation incubation.C. reinhardtii strains displayed an equal level of DNA degradation and repair efficiency in the DNA single strand breaks. The radioresistant line N 198 ofP. sativum is characterized by a lower level of induced DNA ssb and higher efficiency of repair of these breaks as compared with less radioresistant lines NN 131 and 140.  相似文献

Numerous papers have reported that mesenchymal stem cells (MSCs) can be isolated from various sources such as bone marrow, adipose tissue and others. Nonetheless it is an open question whether MSCs isolated from different sources represent a single cell lineage or if cells residing in different organs are separate members of a family of MSCs. Subendothelial tissue of the umbilical cord vein has been shown to be a promising source of MSCs. The aim of this study was to isolate and characterize cells derived from the subendothelial layer of umbilical cord veins as regards their clonogenicity and differentiation potential. The results from these experiments show that cells isolated from the umbilical cord vein displayed fibroblast-like morphology and grew into colonies. Immunophenotyping by flow cytometry revealed that the isolated cells were negative for the hematopoietic line markers HLA-DR and CD34 but were positive for CD29, CD90 and CD73. The isolated cells were also positive for survivin, Bcl-2, vimentin and endoglin, as confirmed by RT-PCR and immunofluorescence. These cells can be induced to differentiate into osteogenic and adipogenic cells, but a new finding is that these cells can be induced to differentiate into endothelial cells expressing CD31, vWF and KDR-2, and also form vessel-like structures in Matrigel. The differentiated cells stopped expressing survivin, thus showing a diminished proliferative potential. It can be assumed that the subendothelial layer of the umbilical cord vein contains a population of cells with the overall characteristics of MSCs, with the additional capability to transform into endothelial cells.  相似文献

Two cell repair systems--photoreactivation and repair of single-strand DNA breaks have been studied using unicellular green algae as a test-system. Effects of the genotype and the intensity of pico/second UV-laser irradiation on the degree of the photoreactivation have been investigated. It has been shown that the lower intensity (I = 8.10(6) W/cm2) effects less the inactivation of living cells comparing with I = 30.10(6) W/cm2, regardless of the genotype. The clearly expressed higher potentials of strains LARG-1 and 260 to produce and repair alterations of the cyclobutane-pyrimidine dimers type have been established. An analysis of DNA degradation during gamma rays irradiation and after incubation has been carried out for investigation the relationship between strains radioresistance and repair of single-strand break. It has been shown that high efficiency of the repair system is characteristic of the resistant strain obtained from chronically irradiated population.  相似文献

The resistance of differentChlorella vulgaris B. strains obtained as a result of chronic influence with chemical and physical factors have been studied. The aim of our work was to establish the relation between the resistance ofChlorella vulgaris populations with ultrastructural cell organization and the level of endogenic thiols. It was established that destructive processes in the cell are reduced to minimum in the more resistant strains and are of a definite recovering character. The higher resistance is accompanied by higher content of endogenic thiols, enlarging of cell pyrenoid and multilayered cell-covering membrane. From the results obtained it may be supposed that one of the probable mechanisms determining the resistance of theChlorella vulgaris strains obtained by the influence of physical and chemical factors and investigated by us, is connected with the changes which took place in the genetic systems controlling the level of the natural protectors in the cell and is accompanied by structural changes affecting mainly the size and number of cell pyrenoid as well as the thickness of the cell-covering membrane.  相似文献

Regeneration systems from immature embryos of Bulgarian pea genotypes   总被引:1,自引:0,他引:1  
Ten genotypes from Pisum sativum and Pisum arvense were screened for their regeneration abilities. Most of them were created through experimental mutagenesis from Bulgarian varieties and have various valuable agronomic traits. Embryonic axes from immature embryos were plated on modified Murashige and Skoog medium, containing different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-d), α-naphthaleneacetic acid (NAA) and benzyladenine (BA). Two schemes for direct and indirect organogenesis were established. Callus and shoot formation were induced on media containing 0.2 mM 2,4-d or 5 mM BA, respectively. Embryonic axes formed buds directly when plated on medium with 10 mM BA and 1 mM NAA. Organogenesis and adventitious bud formation were maintained on medium supplemented with BA and NAA. Rhizogenesis was induced on Gamborgs' B5 medium. All screened genotypes were able to regenerate plants with a high efficiency (50–100%) although some differences in their organogenetic response were observed.
