Satellite and compiled in situ observations of sea surface temperatures have greatly increased the ability to detect anomalous and persistent warm water and are being widely used to predict climate change, coral bleaching and mortality. A field-based synoptic view of coral bleaching spanning eight countries and ∼35° of latitude in the western Indian Ocean tested the accuracy of synoptic temperature data derived from satellites and shipboard data to detect and predict bleaching during 2005. The ability to predict the degree of bleaching based on degree heating weeks data was moderate, but increased when past temperature anomalies and coral community susceptibility were included. It is estimated that slightly more than half of the bleaching response is due to anomalous warm water and nearly half due to taxa and community level acclimation or adaptation, where these two factors have opposing effects. Cumulative temperature anomalies do identify general areas with bleaching but both large over and underestimates of bleaching intensity were observed. Consequently, field observations are needed to confirm the synoptic satellite predictions for particular reefs, particularly where acclimation and reorganization of the coral community have occurred due to past bleaching events.  相似文献

The gene encoding thymidine kinase 1 from tomato (toTK1) has in combination with azidothymidine (AZT) recently been proposed as a powerful suicide gene for anticancer gene therapy. The toTK1/AZT combination has been demonstrated to have several advantages for the treatment of glioblastomas because AZT can easily penetrate the blood-brain barrier and toTK1 can efficiently phosphorylate AZT and also AZT-monophosphate. In a pursuit to further understand the properties of toTK1, we examined the oligomerization properties of recombinant toTK1 and its effect on enzyme kinetics. Previously, it has been shown that human TK1 is a dimer in the absence of ATP and a tetramer if preincubated with ATP. However, we show here that ATP preincubation did not result in a structural shift from dimer to tetramer in toTK1. For human TK1 pretreated with ATP, the K(m) value decreased 20-fold, but toTK1's K(m) value did not show a dependence on the presence or absence of ATP. Furthermore, toTK1 was always found in a highly active form.  相似文献

In situ hybridization on cultured promastigotes and sandfly smears were performed with nonradioactively labeled total DNA and recombinant DNA probes containing minicircle kinetoplast DNA (kDNA) or nuclear DNA inserts. Total DNA probes lack specificity whereas recombinant nuclear DNA probes work only if they contain repetitive sequences. Minicircle kDNAs of five Leishmania isolates, representative of five Leishmania taxa found in Kenya, were sequenced. Comparison of the sequences showed a 150-bp region with around 80% homology, whereas the rest of the minicircles had about 50% homology. Nevertheless, application of these probes in in situ hybridization assays as tested on Leishmania promastigotes in the vector gave good specificity and hybridization signal. Two types of labeling were tested: incorporation of biotin-labeled dUTP or directly horseradish peroxidase (HRP)-labeled nucleotides. Both techniques provided good sensitivity and signal-to-noise ratio on cultured promastigotes. Hybridization with HRP-labeled kDNA probes gave a superior signal-to-noise ratio if tested on sandfly preparations. This method provided a reliable and fast identification and facilitated the detection of promastigotes in sandflies. The technique presented here may be helpful in rapid identification of Leishmania promastigotes, and thus make epidemiological studies easier and less time consuming.  相似文献

Module dynamics in the fucoid alga Turbinaria triquetra (J. Agardh) Kützing were studied on a shallow reef flat in the southern Red Sea. Seasonal patterns in thallus density and size were determined, and the initiation, growth, reproduction, and shedding of modules were studied using a tagging approach. The effects of module density and module/thallus size on module initiation, growth, reproduction, and shedding were analyzed, and the occurrence of intraspecific competition among modules was examined. Seasonal variation occurred mainly at the modular level. There was a restricted period of new module formation in the cooler season, followed by fast growth and reproduction, massive shedding of modules from the end of the cooler season onward, and strongly reduced biomass in summer. There was no evidence of suppressed growth in small modules due to intraspecific competition. Module density and thallus/module size had opposite effects on elongation rates. High module densities enhanced maximum elongation rates (fastest‐growing module per thallus), resulting in longer thalli. On the other hand, elongation rates decreased and tissue loss increased with increasing module length. Reproduction had no clear effect on elongation rates, indicating that there was no direct trade‐off between reproduction and growth. The apparent size‐dependence of reproduction was due to delayed fertility in young modules. Module initiation and shedding were independent of module density. Shedding was also independent of module size and reproductive status. We conclude that seasonal changes in the environment affect module initiation, growth, reproduction, and shedding, whereas density and size‐dependent processes mainly affect growth rates.  相似文献

Growth chamber experiments were conducted to determine the influence of the air pollutant ozone (O3) [0.2 and 0.3 ppm (v/v)] on the growth of corn (Zea mays L.) treated with the herbicide atrazine (2.5 and 3.5 kg/ha) and the antioxidant propyl gallate (5 and 8 kg/ha). At both concentrations O3 at 0.3 ppm alone reduced dry weights of corn and chlorophyll. Chlorophyll a was more affected than chlorophyll b by O3 at 0.3 ppm alone and in combination with atrazine. Propyl gallate at both rates protected corn against O3 injury. The interaction between atrazine and O3 at 0.2 ppm was additive whereas at 0.3 ppm it was antagonistic. But combinations of atrazine and propyl gallate did not protect corn from O3 injury.  相似文献

Seasonal variation in density, thallus length and biomass, population size structure, and allometric length‐biomass relationships was investigated in populations of Sargassum ilicifolium (Turner) C. Agardh, Sargassum subrepandum (Forssk.) C. Agardh, and Turbinaria triquetra (J. Agardh) Kütz. (Phaeophyceae) on shallow reef flats in the southern Red Sea. Thallus length and biomass varied strongly with season, with the highest values occurring in the cooler months. Thallus densities showed no significant temporal variation. Log‐total biomass versus log‐density relationships were positive throughout the growth season without any decrease in the slope of the relationship. In two populations, biomass‐density combinations approached the interspecific biomass‐density line, but the massive annual shedding of modules occurred before self‐thinning would set in. Allometric length‐biomass relationships varied with season in all populations and were associated with seasonal module initiation, growth, and shedding. Evidence of a strong asymmetric competition was found in two high‐density populations. These populations showed a predominance of small thalli during peak development, asymmetrical Lorenz curves, increasing Gini coefficients, and increasing thallus length relative to biomass during the main growth phase. In two other less crowded populations, small thalli were absent during peak development, Lorenz curves were symmetrical, and Gini coefficients decreased during the main growth phase. In these populations, size equalization appears to be due to responses at the modular level rather than size‐dependent mortality. We conclude that changes in size structure in this highly seasonal environment are determined by module dynamics, modified by asymmetric competition in some populations, with a minor role of recruitment and no regulatory effect of self‐thinning.  相似文献
