Substantial differences in the in vivo effect of paraquat (Pq) and DTT on basic peroxidase (GBPx) activity and on H2O2 levels were found in grapevine leaves cv. Sultana. GBPx activity decreased and H2O2 levels increased in illuminated Pq treated leaf-discs. Inactivation of GBPx and accumulation of H2O2 depended on the duration and intensity of the illumination to which discs were exposed. Since GBPx was inactivated directly by H2O2 and not by Pq in leaf extracts, and since GBPx are cytosolic isoenzymes and H2O2 is a stable molecule that can easily permeate chloroplast membranes, we concluded that Pq inactivation of GBPx in vivo is mediated by H2O2. In contrast to the effect induced by Pq, DTT directly inactivated GBPx in leaf extracts. In leaf-discs, however, it reduced GBPx activity in the absence of light, although the levels of H2O2 increased only after exposure of the discs to high irradiance, suggesting that under excess of light, a significant fraction of the photosynthetically produced electrons are dissipated through the water-water cycle and H2O2 accumulates as a consequence of GBPx inactivation.  相似文献

The mannose/glucose-binding Dolichos lablab lectin (designated DLL) has been purified from seeds of Dolichos lablab (hyacinth bean) to electrophoretic homogeneity by affinity chromatography on an ovalbumin- Sepharose 4B column. The purified lectin gave a single symmetric protein peak with an apparent molecular mass of 67 kDa on gel filtration chromatography, and five bands ranging from 10 kDa to 22 kDa upon SDS-PAGE. N-Terminal sequence analysis of these bands revealed subunit heterogeneity due to posttranslational proteolytic truncation at different sites mostly at the carboxyl terminus. The carbohydrate binding properties of the purified lectin were investigated by three different approaches: hemagglutination inhibition assay, quantitative precipitation inhibition assay, and ELISA. On the basis of these studies, it is concluded that the Dolichos lablab lectin has neither an extended carbohydrate combining site, nor a hydrophobic binding site adjacent to it. The carbohydrate combining site of DLL appears to most effectively accommodate a nonreducing terminal alpha-d-mannosyl unit, and to be complementary to the C-3, C-4, and C-6 equatorial hydroxyl groups of alpha-d-mannopyranosyl and alpha-d-glucopyranosyl residues. DLL strongly precipitates murine IgM but not IgG, and the recent finding that this lectin interacts specifically with NIH 3T3 fibroblasts transfected with the Flt3 tyrosine kinase receptor and preserves human cord blood stem cells and progenitors in a quiescent state for prolonged periods in culture, make this lectin a valuable tool in biomedical research.   相似文献

水稻(Oryza sativa L.)幼苗经盐、热激和冷三种不同胁迫预处理均提高了幼苗的抗寒性。与未预处理苗相比,在处理后、低温伤害后和常温下恢复2d的三个时期,不同胁迫预处理苗的可溶性和热不稳定蛋白含量变化趋势甚为相似,但热稳定蛋白含量变化则各有异同。SDS-PAGE图谱分析显示,不同胁迫预处理提高水稻幼苗抗寒性时,其可溶性蛋白、热稳定和热不稳定蛋白组成变化亦各有异同。除诱导出共有的新多肽外,还各自诱导出特有的新多肽。结果表明,植物对不同胁迫的交叉适应存在一定的共同机理,但亦可看出植物对同一种环境胁迫似乎不是以同一的机理去适应。  相似文献

Pig kidney fructose-1,6-bisphosphatase is a homotetrameric enzyme which does not contain tryptophan. In a previous report the guanidine hydrochloride-induced unfolding of the enzyme has been described as a multistate process [Reyes, A. M., Ludwig, H. C., Ya?ez, A. J., Rodriguez, P. H and Slebe, J. C. (2003) Biochemistry 42, 6956-6964]. To monitor spectroscopically the unfolding transitions, four mutants were constructed containing a single tryptophan residue either near the C1-C2 or the C1-C4 intersubunit interface of the tetramer. The mutants were shown to retain essentially all of the structural and kinetic properties of the enzyme isolated from pig kidney. The enzymatic activity, intrinsic fluorescence, size-exclusion chromatographic profiles and 1-anilinonaphthalene-8-sulfonate binding by the mutants were studied under unfolding equilibrium conditions. The unfolding profiles were multisteps, and formation of hydrophobic structures was detected. The enzymatic activity of wild-type and mutant FBPases as a function of guanidine hydrochloride concentration showed an initial enhancement (maximum approximately 30%) followed by a biphasic decay. The activity and fluorescence results indicate that these transitions involve conformational changes in the fructose-1,6-bisphosphate and AMP domains. The representation of intrinsic fluorescence data as a 'phase diagram' reveals the existence of five intermediates, including two catalytically active intermediates that have not been previously described, and provides the first spectroscopic evidence for the formation of dimers. The intrinsic fluorescence unfolding profiles indicate that the dimers are formed by selective disruption of the C1-C2 interface.  相似文献

The complexity of searches and the volume of genomic data make sequence alignment one of bioinformatics most active research areas. New alignment approaches have incorporated digital signal processing techniques. Among these, correlation methods are highly sensitive. This paper proposes a novel sequence alignment method based on 2-dimensional images, where each nucleic acid base is represented as a fixed gray intensity pixel. Query and known database sequences are coded to their pixel representation and sequence alignment is handled as object recognition in a scene problem. Query and database become object and scene, respectively. An image correlation process is carried out in order to search for the best match between them. Given that this procedure can be implemented in an optical correlator, the correlation could eventually be accomplished at light speed. This paper shows an initial research stage where results were "digitally" obtained by simulating an optical correlation of DNA sequences represented as images. A total of 303 queries (variable lengths from 50 to 4500 base pairs) and 100 scenes represented by 100 x 100 images each (in total, one million base pair database) were considered for the image correlation analysis. The results showed that correlations reached very high sensitivity (99.01%), specificity (98.99%) and outperformed BLAST when mutation numbers increased. However, digital correlation processes were hundred times slower than BLAST. We are currently starting an initiative to evaluate the correlation speed process of a real experimental optical correlator. By doing this, we expect to fully exploit optical correlation light properties. As the optical correlator works jointly with the computer, digital algorithms should also be optimized. The results presented in this paper are encouraging and support the study of image correlation methods on sequence alignment.  相似文献

In order to evaluate the impact of the lesser weever on the ecosystem of the southern North Sea, geographical distribution, density, growth, production and food requirements have been estimated. High densities were found on and around the Brown Ridge, an area with high tidal current velocities, medium grain-size of the sediment and a poor benthic fauna. Growth is restricted to the months of June October. During the winter cessation of growth a considerable loss of weight (about 20%) takes place. Mortality has been estimated by using the average size frequency distribution of all catches made from 1972 to 1984. The resulting convex type of survival curve indicates a high survival rate of the II to IV-group fishes. The production (estimated with Allen's graphical method) of a population of 100 individuals including all age groups (0-VI) amounts to 123.7 g AFDW-year'. In areas with highest densities, consequently, production amounts to 0.018–0.078 g AFDW-m2 -year-1. With an assumed transfer efficiency of 10% through the year, food requirements amounts to 0.18–0.78 g AFDW-m 2 -year -1. Since the lesser weever feeds mainly on fish (85.6%), almost exclusively on gobies (Pomatoschistus sp.), and with an assumed transfer efficiency of approximately 10%, the indirect predatory pressure exerted by it may amount to 1.6 6.7g AFDW-m 2.year -1. A possible feeding by gobies on pelagic organisms (calanoids, mysids) is discussed.  相似文献

Although unadapted germplasms have been used to improve disease and insect resistance in alfalfa, there has been little effort to use these for improving forage yield. We evaluated genetic diversity and combining ability among two unadapted germplasms (Medicago sativa ssp. sativa Peruvian and M. sativa ssp. falcata WISFAL) and three Northern U.S. adapted alfalfa cultivars. Population structure analyses indicated that the WISFAL and Peruvian germplasms were genetically distinct from the cultivars, although Peruvian was relatively closer to the cultivars. Peruvian and WISFAL germplasms were intermated to generate a novel hybrid population. This population was crossed to the three cultivars as testers, and the testcross progenies were evaluated for forage yield along with the hybrid population, the original germplasms (Peruvian, WISFAL and cultivars), testcrosses of Peruvian and WISFAL to the three cultivars and a three-way hybrid of the cultivars. The experiment was carried out in the field in Temuco, Chile and Arlington, Wisconsin, USA, and forage was harvested during two seasons. Results from these evaluations showed that hybrids between the Peruvian × WISFAL population and the cultivar testers yielded as much as the cultivar testers. Heterosis was observed between Peruvian and WISFAL, and between these germplasms and the cultivar testers, suggesting that each germplasm may contain different favorable alleles. If Peruvian and WISFAL populations contain alleles at different loci that complement cultivar testers, then combining and enriching these alleles in a single population could result in improved combining ability with alfalfa cultivars.  相似文献
