The effect of polyamines on in vitro reconstitution of Escherichia coli 30S and 50S ribosomal subunits has been studied. Spermidine stimulated the reconstitution of 30S particles from 16S rRNA lacking the methyl groups on two neighboring adenines and total proteins of 30S subunits at least 1.6-fold. The reconstitution of 30S particles from normal 16S rRNA and total proteins of 30S subunits exhibited only slight spermidine stimulation. However, the optimal Mg2+ concentration of the reconstitution was decreased from 20 mM to 16 mM in the presence of 3 mM spermidine. In the absence of spermidine the assembly of 30S particles from normal 16S rRNA was more rapid than the assembly from 16S rRNA lacking the methyl groups on two neighboring adenines. The reconstitution of 50S particles from 23S and 5S rRNA and total proteins of 50S subunits was not influenced greatly by spermidine. Gel electrophoresis results, from reconstitution experiments of 30S particles from 16S rRNA lacking the methyl groups on two neighboring adenines and total proteins of 30S subunits, showed that the assembly of S1 and S9 proteins to 23S core particles was stimulated by spermidine during reconstitution. The relationship of polyamine effects on in vitro ribosome assembly from its constituents to in vivo ribosome assembly is discussed. The reconstitution of Bacillus subtilis 30S particles from 16S rRNA and total proteins of 30S subunits was also stimulated approximately 1.3-fold by 3 mM spermidine. 相似文献

We investigated the oligomerization of solid valine and the stabilities of valine and valine peptides under conditions of high temperature (150–200 °C) and high pressure (50–150 MPa). Experiments were performed under non-aqueous condition in order to promote dehydration reaction. After prolonged exposure of monomeric valine to elevated temperatures and pressures, the products were analyzed by liquid chromatography mass spectrometry comparing their retention times and masses. We identified linear peptides that ranged in size from dimer to hexamer, as well as a cyclic dimer. Previous studies that attempted abiotic oligomerization of valine in the absence of a catalyst have never reported valine peptides larger than a dimer. Increased reaction temperature increased the dissociative decomposition of valine and valine peptides to products such as glycine, β-alanine, ammonia, and amines by processes such as deamination, decarboxylation, and cracking. The amount of residual valine and peptide yields was greater at higher pressures at a given temperature, pressure, and reaction time. This suggests that dissociative decomposition of valine and valine peptides is reduced by pressure. Our findings are relevant to the investigation of diagenetic processes in prebiotic marine sediments where similar pressures occur under water-poor conditions. These findings also suggest that amino acids, such as valine, could have been polymerized to peptides in deep prebiotic marine sediments within a few hundred million years. 相似文献

A new species of phocoenid cetacean Haborophocoena , collected from the Early Pliocene Embetsu Formation, in the close vicinity to the site where the holotype of the type species of the genus Haborophocoena was retrieved, improves our knowledge of the anatomy of this cetacean. The new species, represented by a skull, lacking distal half of the rostrum, ear bones, and teeth, is significantly smaller than, but morphologically similar to, the type species Haborophocoena toyoshimai Ichishima and Kimura 2005 . This new record of Haborophocoena confirms the veracity of the genus Haborophocoena as a biological entity, contributes to understanding the interspecific variation, and suggests that the genus diversified in the northwestern Pacific of early Pliocene time. The adult-like degree of ossification in this small skull precludes it from being a juvenile of Haborophocoena toyoshimai . Autapomorphies of Haborophocoena minutus include narrow premaxilla on the dorsal surface of the rostrum, the anterior position of the premaxillary foramen, and a distinct ridge dividing a fossa on the lateral face of the base of the zygomatic process into the dorsal and the ventral halves, the zygomatic process rectangular in lateral profile, and its small size, which is the most compelling feature, making it a distinct species. 相似文献

Creutzfeldt-Jakob disease (CJD) is a transmissible, fatal, neurodegenerative disease in humans. Recently, various drugs have been reported to be useful in the treatment of CJD; however, for such treatments to be useful it is essential to rapidly and accurately diagnose CJD. 124 CJD patients and 87 with other diseases causing rapid progressive dementia were examined. Cerebral spinal fluid (CSF) from CJD patients was analyzed by 2D-PAGE and the protein expression pattern was compared with that from healthy subjects. One of three CJD-specific spots was found to be fatty acid binding protein (FABP), and heart-type FABP (H-FABP) was analyzed as a new biochemical marker for CJD. H-FABP ELISA results were compared between CJD patients and patients with other diseases (n = 211). Visual readout accuracy of the Rapicheck® H-FABP test panel for CSF was analyzed using an independent measure of CSF H-FABP concentration. The distribution of H-FABP in the brains of CJD patients was examined by immunohistochemistry. ELISA sensitivity and specificity were 90.3% and 92.9%, respectively, and Rapicheck® H-FABP sensitivity and specificity were 87.9% and 96.0%, respectively. ELISA and Rapicheck® H-FABP assays provided comparable results for 14-3-3 protein and total tau protein. Elevated H-FABP levels were associated with an accumulation of abnormal prion protein, astrocytic gliosis, and neuronal loss in the cerebral cortices of CJD patients. In conclusion, Rapicheck® H-FABP of CSF specimens enabled quick and frequent diagnosis of CJD. H-FABP represents a new biomarker for CJD distinct from 14-3-3 protein and total tau protein. 相似文献

Human liver glyceraldehyde‐3‐phosphate dehydrogenase (GAPDH) was purified and crystallized using PEG 3350 as a precipitant. However, the crystals were extremely fragile towards osmotic shock. A 1% change in PEG 3350 content causes destruction of the crystals. After many trials for freezing the crystals, X‐ray diffraction data from a native crystal were collected at 2.8 Å resolution using as a cryoprotectant a mixture consisting of paraffin oil and Paratone‐N in a 3:1 ratio and a cryoloop covered with Formver film. Crystals belong to space group P21, with unit‐cell parameters a = 63.23, b = 97.84, c = 84.23 Å, β = 104.1°. Molecular replacement with a starting model consisting of a homology model based on the low‐resolution structure of human skeletal muscle GAPDH, which has 90% identical residues with the liver protein, led to a solution. Most of the current model was assigned properly in the electron‐density map, but the map corresponding to some important regions containing the phosphate‐binding loop was ambiguous. It is planned to crystallize human liver GAPDH in the presence of phosphate ions and/or some kind of inhibitor in order to fix the flexible region. 相似文献

Two fragments of the C‐terminal catalytic domain of human poly(ADP‐ribose) polymerase (catPARP), Met‐catPARP and Gly‐Ser‐catPARP, were purified and crystallized. Both catPARP crystals belong to space group C2, with almost the same unit‐cell parameters. However, the shapes and harvest periods of both crystals were quite different owing to the slight mutation at the N‐terminal position. Gly‐Ser‐catPARP was found to be more suitable for X‐ray crystallography and crystals showed diffraction to at least 3.5 Å resolution. 相似文献
