Pair-formation behaviour among the Eiders began after the annual moult, in late September. There was a small peak of display in October and November and a large one in the following April and May. As a result, the female population became paired in two phases, up to 50% in the autumn and the remainder in the following spring. The seasonal cycle of the abundance of Leydig cells was bimodal, with a peak in October and a large one in April-May. That the peaks represent increased androgen production was supported by the cycles of Leydig cell enzyme activity and lipid content and of penis weight. The monthly means of Leydig cell abundance and of the rate of display were statistically correlated. Experiments with sexually quiescent males in eclipse plumage demonstrated that testosterone was capable of inducing pair-formation behaviour. Relatively advanced stages of spermatogenesis were found in autumn, and were maintained through the winter, complete development occurring in the spring. Seasonal changes in the production of pituitary gonadotropin followed a bimodal pattern with peaks in autumn and in the spring. Field evidence is presented which suggests that Follicle Stimulating Hormone and Luteinizing Hormone were produced at different phases of a circadian rhythm of photosensitivity.  相似文献

Many diatom-based methods have been proposed in recent years. Besse-Lototskaya et al. (2011) compare some of those developed for assessing inorganic nutrients. However, they fail to address the two questions of greatest interest to those who wish to use such metrics to inform decision-making. These questions are: what is the role of diatom-based trophic metrics in environmental management in Europe in the twenty-first century and do these indices represent causal relationships with underlying pressure gradients? The present economic climate means that Member States of the European Union will look more critically at all methods for assessing freshwater quality and developers need to ensure that their methods are “fit-for-purpose”.  相似文献

Microsomal preparations derived from bovine placenta cotyledons, previously investigated as a convenient source of fatty acyl coenzyme A: estradiol-17 beta-acyl transferase, have been shown to acylate other steroids bearing 3 beta- or 17 beta-hydroxyl groups. In the presence of 0.1 mM oleoyl CoA, the apparent Km values for dehydroepiandrosterone, testosterone, and 5-androstene-3 beta,17 beta-diol (delta 5-DIOL) were 45, 67, and 20 microM, respectively. Acylation of delta 5-DIOL occurred at either the 3 beta- or 17 beta-positions to give monoesters. Testosterone, estradiol-17 beta, and delta 5-DIOL acted as competitive inhibitors for the acylation of the 3 beta-hydroxyl group of dehydroepiandrosterone (Ki values 71, 75, and 41 microM, respectively). Such data indicate that a single enzyme of wide substrate specificity may be involved in these acylation reactions. When estrogen receptor (ER) positive and negative human mammary cancer cell lines were incubated with 10 nM [3H]delta 5-DIOL, intracellular accumulation of delta 5-DIOL long-chain fatty acid esters occurred; rates being higher (p less than 0.001) in ER negative cells (MDA-MB-231 and MDA-MB-330) compared to MCF-7 cells (ER positive), and higher (P less than 0.005) in MDA-MB-231 cells compared to ZR-75-1 cells (ER positive). After exposure to 10 nM [3H]delta 5-DIOL for 16 h, the total labeled steroid fatty acid fraction was composed predominantly of delta 5-DIOL-3 beta- and 17 beta-monoesters (approximately 85%), the remainder containing approximately equal amounts of delta 5-DIOL-diesters and dehydroepiandrosterone-3 beta-esters. Subsequent transfer to medium lacking delta 5-DIOL was accompanied by a breakdown of the labeled esters, which was more rapid in the ER positive cell lines. During this period, intracellular free delta 5-DIOL levels rapidly declined in MDA-MB-330 cells but were maintained in MCF-7 cells, presumably by binding to ER. This behavior parallels that of estradiol-17 beta previously observed in these cell lines and further emphasizes the potential importance of the adrenal-derived estrogen delta 5-DIOL in consideration of a hormone-based etiology of human breast cancer.  相似文献

Evidence regarding the interaction of ultraviolet-B (UV-B, 280-320 nm) radiation and plant competition in terrestrial ecosystems is examined. The competitive interactions of some species pairs were affected even by ambient solar UV-B radiation (as exists without ozone depletion), when compared to control pairs grown without UV-B. Also, the total shoot biomass of these species pairs was depressed under ambient UV-B. Relatively large increases in UV-B radiation (approximating a 40% ozone layer reduction when weighted with the generalized plant action spectrum) altered the competitive interactions of some species pairs grown in pots under field conditions, but did not affect the total shoot biomass production of those pairs. Recent field experiments have examined the competitive interactions of wheat ( Triticum aestivum L. cv. Bannock) and wild oat ( Avena fatua L.) under a simulated increased UV-B regime resulting from a 16% ozone layer reduction when weighted with the generalized plant action spectrum. This increase in UV-B altered the competitive interactions of these two species without affecting the total shoot biomass production of the species pair. The manner in which increased UV-B affected the relative competitive abilities of the two species was highly dependent upon the environmental conditions during the early life stages of the plants. The implications of these results for both agricultural and natural plant communities are discussed.  相似文献

Summary We present a simple model of habitat selection in which individuals differ in their ability to discriminate between resource sites' profitabilities. The model investigates the effects of violating the ideal assumption of the well-known ideal free distribution (IFD). We show that (1) variability in perceptual limits within a population can significantly change the distribution of foraging animals even though the mean perceptual limit is the same, (2) the direction of this change depends on the proportion of the population that choose randomly between resource sites and (3) better perceivers are more likely to be found at individually more profitable sites, which, because of undermatching with respect to the IFD, are also the absolutely more profitable sites. We note that variability in perceptual limits almost always led to an undermatching of organisms to resources, thereby extending previous workers' results implying that the incorporation of any form of perceptual limits leads to undermatching with respect to the IFD.  相似文献

A conformational change accompanying Ca2+ binding to troponin C (TnC) constitutes the initial event in contractile regulation of vertebrate striated muscle. We replaced endogenous TnC in single skinned fibers from rabbit psoas muscle with a modified form of cardiac TnC (cTnC) which, unlike native cTnC, probably contains an intramolecular disulfide bond. We found that such activating TnC (aTnC) enables force generation and shortening in the absence of calcium. With aTnC, both force and shortening velocity were the same at pCa 9.2 and pCa 4.0. aTnc could not be extracted under conditions which resulted in extraction of endogenous TnC. Thus, aTnC provides a stable model for structural studies of a calcium binding protein in the active conformation as well as a useful tool for physiological studies on the primary and secondary effects of Ca2+ on the molecular kinetics of muscle contraction.  相似文献

The early, rapid phase of tension recovery (phase 2) after a step change in sarcomere length is thought to reflect the force-generating transition of myosin bound to actin. We have measured the relation between the rate of tension redevelopment during phase 2 (r), estimated from the half-time of tension recovery during phase 2 (r = t0.5(-1)), and steady-state force at varying [Ca2+] in single fibers from rabbit psoas. Sarcomere length was monitored continuously by laser diffraction of fiber segments (length approximately 1.6 mm), and sarcomere homogeneity was maintained using periodic length release/restretch cycles at 13-15 degrees C. At lower [Ca2+] and forces, r was elevated relative to that at pCa 4.0 for both releases and stretches (between +/- 8 nm). For releases of -3.4 +/- 0.7 nm.hs-1 at pCa 6.6 (where force was 10-20% of maximum force at pCa 4.0), r was 3.3 +/- 1.0 ms-1 (mean +/- SD; N = 5), whereas the corresponding value of r at pCa 4.0 was 1.0 +/- 0.2 ms-1 for releases of -3.5 +/- 0.5 nm.hs-1 (mean +/- SD; N = 5). For stretches of 1.9 +/- 0.7 nm.hs-1, r was 1.0 +/- 0.3 ms-1 (mean +/- SD; N = 9) at pCa 6.6, whereas r was 0.4 +/- 0.1 ms-1 at pCa 4.0 for stretches of 1.9 +/- 0.5 (mean +/- SD; N = 14). The hypothesis that chronic competitive antagonism of nicotinic acetylcholine receptors (nAChR), even in the absence of immobilization or paralysis, induces proliferation of the receptor and tolerance to the competitive antagonist was tested. Chronic antagonism of the nAChR was achieved in rats by an infusion of d-tubocurarine (dTC) via subcutaneously placed osmotic pumps. After 2 wk of dTC or saline, the neuromuscular pharmacodynamics and nAChR number were examined. No differences in weight gain or mobility were observed between groups. Chronic dTC infusion at 2 wk resulted in a baseline concentration of 0.41 +/- 0.07 (SE) micrograms/ml, which, if achieved acutely, would cause a depression of the twitch tension to 60% of control twitch height. Moreover the experimental group was able to develop a baseline twitch tension of 50 g, similar to that of controls. Despite the baseline dTC concentration in the experimental group, the effective doses of dTC for twitch depression were similar to those of controls. The plasma dTC concentration required for steady-state twitch inhibition was significantly (P less than 0.05) higher in the experimental group (0.83 +/- 0.04 vs. 0.50 +/- 0.15 micrograms/ml) as were the extrajunctional nAChR (19.76 +/- 1.77 vs. 13.37 +/- 1.82 fmol/mg protein). The diaphragmatic nAChR were unaltered. This study confirms that chronic doses of dTC cause tolerance to its effects and proliferation of nAChR even in the absence of immobilization. The absence of nAChR changes in the diaphragm may be due to the higher margin of safety of the diaphragm for muscle relaxants than for peripheral muscles. Intensive Care Unit patients receiving chronic infusions of dTC to facilitate mechanical ventilation will require increased doses with time.  相似文献

There is an increasing awareness of the need to assess the impact of nutrient enrichment on river ecosystems separately from the impacts of organic effluents. A range of methods have been proposed and some have moved from the development stage to practical use by water management organizations. The methods can be applied to broad surveys or provide baseline information to assess possible future change. In the latter case it is recommended that several different methods are used, especially where it is important to get reliable information on the long-term impact of improvements in effluent quality. Estimates of biomass measured as chlorophyll a have often been used for phytoplankton and sometimes also for benthic communities. However, a lot of care is needed in applying this method, because of the range of factors besides nutrient concentration which can influence values. Approaches based on the whole community have been developed by a number of research groups, usually involving semiquantitative estimates of abundance. There has also been a rapid increase in the use of indices based on the relative proportions of epilithic diatom species. The methodologies used by a number of research groups in Europe are broadly similar, making it possible to compare results between different regions. The development of indices based on macrophyte floristic composition in relation to river nutrient status is also under development, especially in France and UK. However, interpretation of the results is complicated where long-term changes are taking place in nutrient concentrations in the water, because of the varying contributions of sediment and water to different species of rooted plant. Bioassays can be especially helpful where it is desired to establish whether either N or P is limiting for a population of community.  相似文献
