The aim of this work is complex estimation of the nitric oxide action on whole blood of healthy people. We tested the reaction of whole human blood (n = 14) to the processing of it with cold NO-containing plasma. We performed direct sparging of blood samples by gaseous flow with NO in a special plant. Cold NO-containing plasma was generated by apparatus “Plazon” (Russia). We tested lactate dehydrogenase activity in direct and reverse reactions, aldehyde dehydrogenase and superoxide dismutase activity, total protein and lactate level, acid-base balance and the partial pressure of gases in blood. For integral assessment of energy metabolism changes a number of derivative parameters (coefficients of energy reaction balance and substrate provision) were used. Our experiments showed that the processing of whole human blood with NO-containing gas flow (NO concentration — 800 ppm) results in significant changes of its physical and chemical parameters. This exposure leads to inhibition of erythrocytes energy metabolism, decreasing plasma antioxidant reserves, developing moderate ionic disorders and acid-base disbalance in blood samples in vitro.  相似文献

The goal of this work was to study the influence of nitric oxide inhalation on parameters of blood proand antioxidant systems in rats under both an intact condition and experimental thermal injury. We studied 40 Wistar rats that were divided into four equal groups. The intact group was subjected to no manipulation exñept a single blood sampling, main group I was subjected to inhalation of a air mixture containing 20 ppm of nitric oxide for 10 days, the control group was subjected to thermal injury and conventional treatment, and main group II was subjected to thermal injury and daily inhalation of nitric oxide (20 ppm) for 10 days. We studied the intensity of lipid peroxidation in the blood plasma, the total antioxidant activity, the peroxide resistance of erythrocytes, the level of malondialdehyde in the blood plasma and erythrocytes, and the activity of superoxide dismutase. It was shown that daily inhalations of a mixture containing a low concentration of nitric oxide (20 ppm) modified blood oxidative metabolism in healthy and burned rats. We hypothesized that the activation of lipid peroxidation in erythrocytes accompanied by a pronounced increase in the catalytic activity of superoxide dismutase is a unified response of healthy and burned rats to exogenous nitric oxide exposure. We also observed a moderate prooxidant effect in the blood plasma of healthy animals comparable to that in the erythrocytes of these rats. In the case of thermal injury, oxidative stress tended to be corrected after the end of the course of inhalation.  相似文献

The aim of this work was to estimate the dynamics of blood physical and chemical parameters when blood specimens were processed by singlet oxygen in vitro. Our experiments were executed with whole blood specimens of healthy people (n = 10). Each specimen was divided into five separate portions of 5 mL. The first portion was a control (without any exposures). The second one was processed by an oxygen-ozone mixture (at ozone concentration of 500 μg/L, the third portion by oxygen, and the fourth and fifth ones were processed by a gas mixture with singlet oxygen (50 and 100% of generator power). In blood samples after processing we studied the activity of lactate dehydrogenase, aldehyde dehydrogenase and superoxide dismutase, erythrocyte and plasma levels of glucose and lactate, acid-base balance and the partial pressure of gases in blood. It was found out, that blood processing by singlet oxygen leads to optimization of energy, detoxication and antioxidant enzymes functioning with changes in plasma and erythrocyte level of glucose and lactate, normalization of blood gases level and acid-base balance. Our results show, that the effect of singlet oxygen on enzyme activity is more pronounced than exposure to an oxygen-ozone gas mixture.  相似文献

The objective of this work was to study how a prolonged course of inhalations of nitric oxide or singlet oxygen modify crystallization of the blood serum in rats. Experiments were performed with 50 adult Wistar rats, which were divided into five equal groups. The control group (n = 10) included intact rats, which were not exposed to any manipulation other than a single blood drawing. Rats of groups 2, 3, and 4 received inhalations of nitric oxide at 20, 50, and 100 ppm, respectively, daily for 30 days. Group 5 rats (n = 10) received a similar course of inhalations of singlet oxygen for 30 days. Blood samples were drawn from the sublingual vein in rats of the test groups immediately after completion of the inhalation course (day 30 of the experiment) and in the recovery period (day 60 of the experiment) and tested for crystallogenic activity. Dried samples were evaluated visually for crystallizability, structure index, facia destruction degree, and marginal zone clarity, using respective scales. A prolonged course of NO inhalations was found to modulate the crystallogenic properties of the rat serum. An optimal response was recorded at the lowest nitric oxide concentration (20 ppm). Higher NO concentrations caused more adverse changes in serum crystallization after the end of the inhalation course and hindered the recovery processes. Singlet oxygen inhalations for 30 days did not exert a considerable adverse effect on the crystallogenic properties of the rat serum.  相似文献

A comparative analysis of the influence of different nitric oxide forms on the character of the dehydration structuring of human serum samples was carried out. The effects of an NO-containing gas flow that was generated by a Plazon device (800 and 80 ppm), an experimental NO generator (20, 50, 75, and 100 ppm), as well as by glutathione-containing dinitrosyl iron complexes (3 mM/L) were investigated using 15 healthy donors. The influence of endogenous sodium on serum crystallization of intact and NO treated blood samples was evaluated. It was found that the effect of NO on the crystallogenic properties of blood serum is directly determined by its concentration and form (free or bound), as well as by the presence of reactive oxygen species in a gas flow. The most pronounced stimulatory effect was observed for the bound NO form, dinitrosyl iron complexes with glutathione ligands. Low NO concentrations modulated the crystallogenic properties of blood serum, while the best stimulatory effect was demonstrated by the gas flow that contained 20 ppm nitric oxide. In contrast, high NO concentrations (800 ppm) inhibited the crystallogenic activity of the biological medium due to an increase in the destruction of structural elements by many times, which lead to the formation of an additional band in the marginal zone of the microscope specimen.  相似文献
