Summary The use of reticulated polyurethane foam as a support material for the immobilization of methanogenic associations and its application to the anaerobic treatment of fine particulate solid wastes was investigated. The colonization of polyurethane support particles in a continuous upflow reactor fed on a mixture of acetate, propionate and butyrate, was both rapid and dense. The combination of rumen microorganisms and colonized support particles in a two-phase digester resulted in an efficient anaerobic decomposition of papermill sludge.  相似文献

Regional analyses of the interaction between human populations and natural resources must integrate landscape scale environmental problems. An approach that considers human culture, environmental processes, and resource needs offers an appropriate methodology. With this methodology, we analyze problems of food availability in African cattle-keeping societies. The analysis interrelates cattle biomass, forage availability, milk and blood production, crop yields, gathering, food subsidies, population, and variable precipitation. While an excess of cattle leads to overgrazing, cattle also serve as valuable food storage mechanisms during low rainfall periods. Food subsidies support higher population levels but do not alter drought-induced population fluctuations. Variable precipitation patterns require solutions that stabilize year-to-year food production and also address problems of overpopulation.  相似文献

The 100 bp sequence from the beginning of the 16S rRNA gene of archaebacterium Halobacterium halobium and the adjacent 800 bp upstream sequence were determined. Four long (80 bp) direct repeats were found in the region preceeding the structural gene of the 16S rRNA. These repeats are proposed to constitute the promoter region of the rRNA operon of H. halobium.  相似文献

Background

Long-term benefits in animal breeding programs require that increases in genetic merit be balanced with the need to maintain diversity (lost due to inbreeding). This can be achieved by using optimal contribution selection. The availability of high-density DNA marker information enables the incorporation of genomic data into optimal contribution selection but this raises the question about how this information affects the balance between genetic merit and diversity.

Methods

The effect of using genomic information in optimal contribution selection was examined based on simulated and real data on dairy bulls. We compared the genetic merit of selected animals at various levels of co-ancestry restrictions when using estimated breeding values based on parent average, genomic or progeny test information. Furthermore, we estimated the proportion of variation in estimated breeding values that is due to within-family differences.

Results

Optimal selection on genomic estimated breeding values increased genetic gain. Genetic merit was further increased using genomic rather than pedigree-based measures of co-ancestry under an inbreeding restriction policy. Using genomic instead of pedigree relationships to restrict inbreeding had a significant effect only when the population consisted of many large full-sib families; with a half-sib family structure, no difference was observed. In real data from dairy bulls, optimal contribution selection based on genomic estimated breeding values allowed for additional improvements in genetic merit at low to moderate inbreeding levels. Genomic estimated breeding values were more accurate and showed more within-family variation than parent average breeding values; for genomic estimated breeding values, 30 to 40% of the variation was due to within-family differences. Finally, there was no difference between constraining inbreeding via pedigree or genomic relationships in the real data.

Conclusions

The use of genomic estimated breeding values increased genetic gain in optimal contribution selection. Genomic estimated breeding values were more accurate and showed more within-family variation, which led to higher genetic gains for the same restriction on inbreeding. Using genomic relationships to restrict inbreeding provided no additional gain, except in the case of very large full-sib families.  相似文献

Activity patterns of Phyllophaga crinita (Burmeister), Phyllophaga congrua (LeConte), Phyllophaga crassissima (Blanchard), and Cyclocephala lurida (Bland) grubs were monitored with acoustic sensors in small pots of bluegrass, Poa arachnifera Torr, at varying and constant temperatures over multiple-day periods. Experienced listeners readily distinguished three types of sound with distinct differences in frequency and temporal patterns, intensities, and durations. Of approximately 3,000 sounds detected from P. crinita larvae, 7% were identifiable as snaps, with large amplitudes and short durations typically associated with root breakage or clipping activity. Approximately 60% were identifiable as rustles, suggestive of surfaces sliding or rubbing past each other during general movement activity. Another 2% of sounds contained patterns of repeated pulses suggestive of surfaces scraping across a pointed ridge. The remaining 31% had spectral or temporal patterns that fell outside the ranges of easily recognizable sound types. Because the behavioral significance of the different sound types has not yet been fully established, the classified and unclassified sounds were pooled together in analyses of the effects of species, temperature, weight, and time of day. Grubs of all four species produced detectable sounds at rates that increased with temperature [0.45 sounds/((min)(degrees C))] and larval weight [6.3 sounds/((min)(g))]. Mean sound rates were independent of species and time of day. At temperatures <9 degrees C, mean sound rates fell below the typical levels of background noise observed under field conditions. This reduced activity at low temperatures is likely to reduce the effectiveness of acoustic monitoring in the field in cold weather. The consistency of results obtained in these tests over multiple-day periods suggests that acoustic systems have potential as tools for nondestructive monitoring of the efficacy of insect management treatments as well as for biological and ecological studies.  相似文献

Loading of articular cartilage during weight bearing is essential for the maintenance of cartilage function. Although certain cyclic loading protocols stimulate extracellular matrix synthesis, constant or static compression decreases proteoglycan and collagen synthesis in cartilage explants. The goal of this study was to determine whether the compression-induced decrease in proteoglycan synthesis involves an interleukin-1 (IL-1) signaling pathway. Cartilage explants were compressed 50% in the presence of IL-1 receptor antagonist (IL-1ra), and the incorporation of [35S]sulfate into macromolecules was measured. IL-1ra increased sulfate incorporation in compressed cartilage but not in cartilage maintained at the in situ thickness (0% compression). IL-1alpha and IL-1beta mRNAs were detected in cartilage compressed 50% for at least 3h, while nitric oxide synthase II mRNA was only detected in cartilage compressed 50% for 6h. The data support a role for the IL-1 receptor in the pathway linking static compression to reduced proteoglycan synthesis.  相似文献

The ribosomal peptidyl transferase center is responsible for two fundamental reactions, peptide bond formation and nascent peptide release, during the elongation and termination phases of protein synthesis, respectively. We used in vitro genetics to investigate the functional importance of conserved 23S rRNA nucleotides located in the peptidyl transferase active site for transpeptidation and peptidyl-tRNA hydrolysis. While mutations at A2451, U2585, and C2063 (E. coli numbering) did not significantly affect either of the reactions, substitution of A2602 with C or its deletion abolished the ribosome ability to promote peptide release but had little effect on transpeptidation. This indicates that the mechanism of peptide release is distinct from that of peptide bond formation, with A2602 playing a critical role in peptide release during translation termination.  相似文献

Translation of specific short peptides can render the ribosome resistant to macrolide antibiotics such as erythromycin. Peptides act in cis upon the ribosome on which they have been translated. Amino acid sequence and size are critical for peptide activity. Pentapeptides with different consensus sequences confer resistance to structurally different macrolide antibiotics, suggesting direct interaction between the peptide and the drug on the ribosome. Translation of resistance peptides may result in expulsion of the macrolide antibiotics from the ribosome. The consensus sequence of peptides conferring erythromycin resistance is similar to the sequence of the leader peptide involved in translational attenuation of erythromycin resistance genes, indicating that a similar type of interaction between the nascent peptide and antibiotics can occur in both cases.  相似文献
