Change in the ultraviolet spectrum of solubilized Ca2+-dependent ATPase from sarcoplasmic reticulum due to binding with Ca2+ ions.

Solubilized sarcoplasmic reticulum (SSR) was prepared by solubilizing fragmented sarcoplasmic reticulum (FSR) with a nonionic detergent (C12E8) then displacing the detergent with Tween 80, using a DEAE-cellulose column. The UV absorption of SSR decreased reversibly at about 286 and 292 nm on removal of free Ca2+ ions, while no change in the fluorescence spectrum was detectable. On the other hand, the fluorescence intensity of FSR decreased 3-4% on removal of free Ca2+ ions, as previously reported by Dupont [(1976) Biochem. Biophys. Res. Commun. 71, 544-550]. The UV absorption of FSR increased reversibly at about 270-280 nm on removal of free Ca2+ ions, but the rate of the change was very slow (k = about 0.1 min-1).     

Effects of Exogenous Amino Acids on Iron Uptake in Relation to their Effects on Photoperiodic Flowering in Lemna paucicostata 6746

The flowering of Lemna paucicostata 6746 grown on 14-h photoperiodwas enhanced by the addition of high concentrations of ironto the medium, which also increased the endogenous iron concentration.The addition of asparagine, aspartate, glutamate, -alanine,glycine or serine to the medium also increased the endogenousiron level, resulting in the promotion of flowering. In contrast,the addition of cysteine, cystine, glutamine, arginine, threonineor phenylalanine lowered the endogenous iron level, resultingin the inhibition of flowering. Glycine and asparagine added to the medium during an inductive96-h dark period did not promote iron uptake and had no effecton flowering, but when added during the subsequent 120-h lightperiod, they promoted both iron uptake and flowering response.The increase in the endogenous iron level seems to favor floraldevelopment rather than induction of photoperiodic floweringof Lemna paucicostata 6746. Tree form in a mixed dipterocarp forest in Indonesian Borneo

The form of tropical trees was studied with reference to the production structure of the component individuals of a tropical rain forest stand in Sebulu, East Kalimantan in Indonesian Borneo, since the production structure as a physical or bio-economical basis of tree form still remains obscure in tropical rain forests. The pipe model theory successfully explained the crown shapes of different trees, and its parameter, designated as specific pipe length, suggested an increase in the cost of leaf mass growth with an increase in crown size. A mathematical model consisting of exponential functions of aboveground height was applied for describing stem form, and its properties were examined through changes in its coefficients and by adopting an assumption of the geometrical similarity of individual stem form as a criterion for comparing differences in stem form among individual trees. Furthermore, the cost of buttersses was discussed using the relation between bole- and buttress weight calculated from the mathematical model.     

Preparation ofN-acetylneuraminic acid from delipidated egg yolk

Egg yolk, a large proportion of the egg, was studied for the preparation ofN-acetylneuraminic acid (Neu5Ac). The delipidated hen egg yolk (DEY; 500 kg containing 0.2% w/w, Neu5Ac) was hydrolysed with HCl (pH 1.4) at 80 °C and neutralized with NaOH (pH 6.0). The mixture was filtered and electrodialysed until the conductivity was 240 µS cm^–1. The filtrate was applied on a column of Dowex HCR-W2 (20–50 mesh), followed by a column of Dowex 1-X8 (200–400 mesh). The latter column was washed with water, and then eluted with a linear gradient of HCO₂H (0–2m). The eluates containing Neu5Ac were concentrated using a reverse osmosis membrane and, finally, rotary evaporated at 40 °C. The residue was then lyophilized to yield 500 g Neu5Ac. The purity of Neu5Ac was >98% (TBA method). HPLC, NMR spectroscopy and TLC chromatography of the product obtained from the DEY showed that Neu5Ac was the sole derivative present in egg yolk. Canopy photosynthetic production in a Japanese larch stand. I. Seasonal and vertical changes of leaf characteristics along the light gradient in a canopy

In an 18 year old Japanese larch stand, leaf characteristics such as area, weight, gross photosynthetic rate and respiration rate were studied in order to obtain basic information on estimating canopy photosynthesis and respiration. The leaf growth courses in area and weight from bud opening were approximated by simple logistic curves. The growth coefficient for the area growth curve was 0.155–0.175 day⁻¹, while that for the weight growth was 0.112–0.117 day⁻¹. The larger growth coefficient in area growth caused the seasonal change in specific leaf area (SLA) that increased after bud opening to its peak early in May at almost 300 cm² g⁻¹ and then decreased until it leveled off at about 140 cm²g⁻¹. The change inSLA indicates the possibility that leaf area growth precedes leaf thickness growth. The relationship between the coefficientsa andb of the gross photosynthetic rate (p)-light flux density (1) curve (p=bI/(1+aI)) and the mean relative light flux density (I′/I ₀) at each canopy height were approximated by hyperbolic formulae:a=A/(I′/I ₀)+B andb=C/(I′/I ₀)+D. Leaf respiration rate was also increased with increasingI′/I ₀. Seasonal change of gross photosynthetic rate and leaf respiration rate were related to mean air temperature through linear regression on semilogarithmic co-ordinates.     

Effect of base substitutions in the colicin E1 gene on colicin E1 export and bacteriocin activity

Summary Base substitutions have been introduced into the segment of the colicin E1 gene corresponding to the polypeptide region between the 404th and the 502nd residues which was considered to participate in colicin E1 export and bacteriocin activity. The methods used were in vitro localized mutagenesis with sodium bisulphite and in vivo mutagenesis using either nitrosoguanidine or ethyl methane sulphonate. Cells carrying mutagenized plasmids were screened by their inability to form a clear zone on a lawn of colicin E1 sensitive cells. Mutation sites were determined from the nucleotide sequence analysis and the altered amino acid residues were reduced. The mutant proteins were analysed for their ability to be exported to the periplasmic space and for their bacteriocin activity. Out of eight mutants obtained, three had a single amino acid replacement. Mutant proteins that had Ser and Glu in place of Pro-462 and Gly-502, respectively, showed a decrease in both the export and the bacteriocin activity. A mutant protein having Arg in place of Gly-439 showed a decrease only in the bacteriocin activity. These results suggest that the target region of colicin E1 contributes to the export as well as the bacteriocin activity but the two functions are supported in part by different amino acid residues of the protein.     

Specificity of hemorrhagic proteinase from Crotalus atrox (western diamondback rattlesnake) venom

Hemorrhagic proteinase, HTb, isolated from Crotalus atrox (western diamondback rattlesnake) venom was studied for its specificity. HTb showed fibrinogenase activity, hydrolyzing the A alpha chain of fibrinogen first, followed by the cleavage of the B beta chain. HTb is different from thrombin and did not produce a fibrin clot. The degradation products of fibrinogen were found to be different, indicating that the cleavage sites in the A alpha and B beta chains are different from those of thrombin. N-Benzoyl-Phe-Val-Arg-p-nitroanilide was not hydrolyzed by HTb, although this substrate was hydrolyzed by thrombin and reptilase.
