A complete human metaphase chromosome has been reconstructed from a series of electron microscopical projections obtained by tilting the specimen stage at 3 degree intervals from –60 to +60 degrees. The reconstructed structure is about 3.0 m long, 1.6 m wide, and 0.8 m thick. The mass distribution was fairly homogeneous within the chromatids and neither a hollow nor a dense core was observed. The distribution and course of fibers observed are most consistent with a looping model of chromosome structure. 相似文献

The electron microscope has been used to determine the weight distribution of isolated subcellular particles from normal rat liver. The following results are reported: (1) There exist at least two well defined weight populations of subcellular particles; their respective median weights are 1.3 x 10-14 and 11 x 10-14 gm. The lighter fraction is considered to consist of lysosomes, the heavier of mitochondria. (2) The mitochondrial fraction shows a log-normal distribution of the particle weight. (3) By the introduction of morphologic criteria, the mitochondrial fraction is divided into two groups, one consisting of a spherical, the other of an oblong type of particle. The data found support the following concepts: (a) Mitochondria increase their weight from a certain size up by linear growth. (b) Mitochondria divide. The division is not necessarily symmetric; in all cases, however, one part of the division product is a spherical particle. It is felt that these results constitute a valuable demonstration of the general capabilities of quantitative electron microscopy and may stimulate many other useful applications of this technique in cytology, bacteriology, and virology. 相似文献

An almost quantitative synthesis of N-methyl-2-anilino-6-naphthalenesulfonyl hydrazine (2,6-mansyl hydrazine) from sulfonyl chloride and hydrazinlum hydroxide is described. The 2,6-mansyl chloride was prepared by different methods from 6-hydroxynaphthalene-2-sulfonic acid (overall yield: 69%). The N- and the O-mansylation of suitable compounds (e.g., amines, amino acids, and phenolic steroids) with 2,6-mansyl chloride and the preparation of oxosteroid-2,6-mansyl hydrazones are deseribed, and the derivatives obtained, their uv spectra, and methods for their thin-layer chromatographic separation are compared with the corresponding data for dansylated compounds. 相似文献

, Pb2+, Ce3+, Cd2+, Fe2+, Fe3+, Al3+, Sn2+, Cs+ and Li+) and the selenium compounds, selenium(IV) oxide and sodium selenite, have been checked with respect to their ability to influence a single or possible several steps of the biosynthesis ofN-acetylneuraminic acid. It could be shown that the following enzymes are sensitive to these metal ions (usually applied at a concentration of 1 mmoll–1):N-acetylglucosamine kinase (inhibited by Zn2+ and vandate), UDP-N-acetylglucosamine-2-epimerase (inhibited by zn2+, Co2+, Cu2+, Hg2+, VO

, Pb2+, Cd2+, Fe3+, Cs+, Li+, selenium(IV) oxide and selenite), andN-acetylmannosamine kinase (inhibited by Zn2+, Cu2+, Cd2+, and Co2+). Dose dependent measurements have shown that Zn2+, Cu2+ and selenite are more efficient inhibitors of UDP-N-acetylglucosamine-2-epimerase than vanadate. As for theN-acetylmannosamine kinase inhibition, a decreasing inhibitory effect exists in the following order Zn2+, Cd2+, Co2+ and Cu2+. In contrast, La3+, Al3+ and Mn2+ (1 mmoll–1) did not interfere with the biosynthesis ofN-acetylneuraminic acid. The three human male specific expressed gene families DAZ, RBM, and TSPY are known to be repetitively clustered in the Y-specific
region of the human Y Chromosome (Chr). RBM and TSPY are Y-specifically conserved in simians, whereas DAZ cannot be detected on the Y chromosomes of New World monkeys. The proximity of SRY to the pseudoautosomal region (PAR) is highly conserved and thus most effectively stabilizes the pseudoautosomal boundary
on the Y (PABY) in simians. In contrast, the non-recombining part of the Y Chrs, including DAZ, RBM, and TSPY, was exposed to species-specific amplifications, diversifications, and rearrangements. Evolutionary fast fixation of any of

Experiments in visual cortex have shown that the firing rate of a neuron in response to the simultaneous presentation of a
preferred and non-preferred stimulus within the receptive field is intermediate between that for the two stimuli alone (stimulus
competition). Attention directed to one of the stimuli drives the response towards the response induced by the attended stimulus
alone (selective attention). This study shows that a simple feedforward model with fixed synaptic conductance values can reproduce
these two phenomena using synchronization in the gamma-frequency range to increase the effective synaptic gain for the responses
to the attended stimulus. The performance of the model is robust to changes in the parameter values. The model predicts that

The proof of efficacy of phytopreparations and the determination of their mode of action are permanent challenges for an evidence-based phytotherapy. The technology platform of genomics, proteomics and metabolomics ("-omic-" technologies) are high-throughput technologies. They increase substantially the number of proteins/genes that can be detected simultaneously and have the potential to relate complex mixtures to complex effects in the form of gene/protein expression profiles. Provided that phytopreparation-specific signatures in the form of gene/protein expression profiles can be developed, these technologies will be useful for the chemical and pharmacological standardization and the proof of the toxicological potential of a plant extract. Over a long-term perspective they may economize the proof of efficacy, the determination of the mode of action of phytomedicines and allow to investigate herbal extracts without prominent active principle(s). The application of this genomics revealed already that gene expression profiles induced by single drugs and the ones induced by the combination of the same drugs can be entirely different. These results make the information of the mode of action of isolated "active principles/lead substances" of phytopreparations questionable. The application of the "-omic-" technologies may lead to a change of paradigms towards the application of complex mixtures in medicine and open the new field of phytogenomics, -proteomics and -metabolomics. 相似文献

Clathrin-mediated endocytosis (CME) is the best-studied pathway by which cells selectively internalize molecules from the plasma membrane and surrounding environment. Previous live-cell imaging studies using ectopically overexpressed fluorescent fusions of endocytic proteins indicated that mammalian CME is a highly dynamic but inefficient and heterogeneous process. In contrast, studies of endocytosis in budding yeast using fluorescent protein fusions expressed at physiological levels from native genomic loci have revealed a process that is very regular and efficient. To analyse endocytic dynamics in mammalian cells in which endogenous protein stoichiometry is preserved, we targeted zinc finger nucleases (ZFNs) to the clathrin light chain A and dynamin-2 genomic loci and generated cell lines expressing fluorescent protein fusions from each locus. The genome-edited cells exhibited enhanced endocytic function, dynamics and efficiency when compared with previously studied cells, indicating that CME is highly sensitive to the levels of its protein components. Our study establishes that ZFN-mediated genome editing is a robust tool for expressing protein fusions at endogenous levels to faithfully report subcellular localization and dynamics. 相似文献

In the past decades, many studies have focussed on the relation between the input and output of neurons with the aim to understand
information processing by neurons. A particular aspect of neuronal information, which has not received much attention so far,
concerns the problem of information transfer when a neuron or a population of neurons receives input from two or more (populations
of) neurons, in particular when these (populations of) neurons carry different types of information. The aim of the present
study is to investigate the responses of neurons to multiple inputs modulated in the gamma frequency range. By a combination
of theoretical approaches and computer simulations, we test the hypothesis that enhanced modulation of synchronized excitatory
neuronal activity in the gamma frequency range provides an advantage over a less synchronized input for various types of neurons.
The results of this study show that the spike output of various types of neurons [i.e. the leaky integrate and fire neuron,
the quadratic integrate and fire neuron and the Hodgkin–Huxley (HH) neuron] and that of excitatory–inhibitory coupled pairs
of neurons, like the Pyramidal Interneuronal Network Gamma (PING) model, is highly phase-locked to the larger of two gamma-modulated
input signals. This implies that the neuron selectively responds to the input with the larger gamma modulation if the amplitude
of the gamma modulation exceeds that of the other signals by a certain amount. In that case, the output of the neuron is entrained
by one of multiple inputs and that other inputs are not represented in the output. This mechanism for selective information
transmission is enhanced for short membrane time constants of the neuron. 相似文献
