Ten, electrophoretically distinct, molecular forms of alcohol dehydrogenase have been isolated from a single human liver by affinity and ion-exchange chromatography. The starch gel electrophoresis patterns after the dissociation-recombination of the forms are consistent with the hypothesis that they arise from the random combination of alpha, beta 1, gamma 1, and gamma 2 subunits into six heterodimeric and four homodimeric isoenzymes. Large differences in kinetic properties are observed for the homodimeric isoenzymes, alpha alpha, beta 1 beta 1, gamma 1 gamma 1, and gamma 2 gamma 2. At pH 7.5, the Km value of beta 1 beta 1 for ethanol is 0.049 mM and that of alpha alpha is 4.2 mM. Forms gamma 1 gamma 1 and gamma 2 gamma 2 do not obey Michaelis-Menten kinetics at pH 7.5 but exhibit negative cooperativity with Hill coefficients of 0.54 and 0.55 and [S]0.5 values of 1.0 and 0.63 mM, respectively. However, all isoenzymes display Michaelis-Menten kinetics for ethanol oxidation at pH 10.0 with Km values ranging from 1.5 to 3.2 mM. The maximum specific activity of beta 1 beta 1 is considerably lower than that of the other three homodimers at both pH 7.5 and 10.0. The Km values of the four homodimers for NAD+ at pH 7.5 range from 7.4 to 13 microM and those for NADH, from 6.4 to 33 microM. Ki values for NADH range from 0.19 to 1.6 microM. At pH 7.5, the kinetic properties of alpha alpha and beta 1 beta 1, prepared in vitro from dissociated and recombined alpha beta 1, are similar to those of the native homodimers. The forms gamma 1 gamma 1 and gamma 2 gamma 2, prepared from dissociated and recombined alpha gamma 1 and beta 1 gamma 2, respectively, exhibit negative cooperativity with Hill coefficients that are similar to those seen with the respective native homodimers.  相似文献

Sections of tendons from the base of the tail of rats were taken at eight time intervals from 18 days in utero until 244 days after birth and were examined in the electron microscope. For each time period, measurements were made of the relative area of fibroblasts, collagen and interstitial material, of the number of fibroblasts per unit area of tendon and of the average area of individual fibroblasts. The spatial arrangement of fibroblasts in the tendon sections was described quantitatively using the "nearest neighbor" method. Initially there was a rapid increase in the area of collagen accompanied by a decrease in the area occupied by fibroblasts but after 104 days of age these values changed very little. The numbers of fibroblasts per unit area decreased steadily from the embryo until 104 days whereas the average size of each cell increased to reach a maximum area at 40 days of age and then declined. At all time intervals cells were arranged in a regular, dispersed pattern across the tendon fascicles. Growth in width of the rat tail appears to involve the secretion of collagen and other intercellular material symmetrically around each fibroblast, so as to gradually separate the cells until a stage is reached at which cells are sufficiently far apart that there is little contact between adjacent cell processes. This may interfere with the integration of metabolic activity in the tissue. As a consequence, there is shrinkage of the cell bodies and a reduction in secretory activity so that, between 55 and 104 days of age, the tendon enters a period of terminal senescence.  相似文献

Soil and aquatic microscopic organisms live and behave in a complex three-dimensional environment. Most studies of microscopic organism behavior, in contrast, have been conducted using microscope-based approaches, which limit the movement and behavior to a narrow, nearly two-dimensional focal field.1 We present a novel analytical approach that provides real-time analysis of freely swimming C. elegans in a cuvette without dependence on microscope-based equipment. This approach consists of tracking the temporal periodicity of diffraction patterns generated by directing laser light through the cuvette. We measure oscillation frequencies for freely swimming nematodes.Analysis of the far-field diffraction patterns reveals clues about the waveforms of the nematodes. Diffraction is the process of light bending around an object. In this case light is diffracted by the organisms. The light waves interfere and can form a diffraction pattern. A far-field, or Fraunhofer, diffraction pattern is formed if the screen-to-object distance is much larger than the diffracting object. In this case, the diffraction pattern can be calculated (modeled) using a Fourier transform.2C. elegans are free-living soil-dwelling nematodes that navigate in three dimensions. They move both on a solid matrix like soil or agar in a sinusoidal locomotory pattern called crawling and in liquid in a different pattern called swimming.3 The roles played by sensory information provided by mechanosensory, chemosensory, and thermosensory cells that govern plastic changes in locomotory patterns and switches in patterns are only beginning to be elucidated.4 We describe an optical approach to measuring nematode locomotion in three dimensions that does not require a microscope and will enable us to begin to explore the complexities of nematode locomotion under different conditions.  相似文献

The submontane belt of the eastern Alps is dominated by beech forests. However, on rocky and steep south-facing slopes, small vegetation mosaics have developed, which, to a certain degree, are similar to the Pannonian forest-steppes. In spite of their unique conservation importance and threatened status, they have received relatively little scientific attention. In this study we analyzed the spatial pattern of such mosaics. More specifically, our objective was to find out how canopy cover value influences the species composition of the herb layer. According to our results, canopy cover of Pinus sylvestris has a rather limited effect on the herb layer composition and species richness. Thus, in the studied canopy cover range (ca. 5–75% canopy cover), most species occurred under all canopy cover grades. This is presumably a result of the canopy characteristics and branching pattern of P. sylvestris: it can be assumed that the physical conditions of the canopy and intercanopy patches are somewhat similar. This is in sharp contrast with the Pannonian forest-steppes dominated by Quercus pubescens. We conclude that, even though the cessation of traditional land-use may not result in a rapid change of the composition in eastern Austrian forest-steppes, every effort must be made to conserve these valuable habitats.  相似文献

Pharmacokinetic studies of topical ketoprofen formulations using continuous sampling techniques such as microdialysis (MD) or open-flow microperfusion (OFM) require sensitive assays due to small sample volumes. A simple and easy online-SPE-MS/MS method for ketoprofen analysis was developed for both MD and OFM samples obtained from rat dermal tissue. The quantification range is 25-5000 ng/ml with a limit of detection of 3 ng/ml using only 10 microl sample volume. The method is characterized by a simple setup using a short polymeric SPE column (OASIS HLB) for desalting with 1.5 min run times in combination with a sensitive MS detection in negative ESI MRM mode. An easy sample workup procedure was used which enables high throughput analysis of a large number of samples for pharmacokinetic studies. In addition, a commercial available (fenoprofen) as well as an isotopically labelled (deuterated ketoprofen) standard were investigated as potential internal standards. The method was validated according to FDA guidelines for bioanalytical validation in terms of accuracy, intra-batch and inter-batch precision, linearity, matrix effect, recovery and stability for both internal standards. Accuracies were 98-113% (fenoprofen) and 95-108% (deuterated ketoprofen), intra-batch precision was 2-3% R.S.D. (fenoprofen) and 2-6% R.S.D. (deuterated ketoprofen), and inter-batch precision was 2-6% R.S.D. (fenoprofen) and 3-6% R.S.D. (deuterated ketoprofen) over the entire quantification range. The presented method was applied to dermal interstitial fluid samples obtained in a topical administration study of ketoprofen in rats.  相似文献

New molecular forms of human liver alcohol dehydrogenase (ADH), collectively designated ADHIndianapolis (ADHInd), were recently discovered in 29% of liver specimens from Black Americans [Bosron, W. F., Li, T.-K., and Vallee, B. L. (1981). Proc. Natl. Acad. Sci. USA 77:5784]. Three different ADHInd phenotypes have now been identified by starch gel electrophoresis, and four ADHInd enzyme forms isolated by affinity and ion-exchange chromatography. The most cathodic ADHInd form has a single pH optimum at 7.0 for ethanol oxidation and is a homodimer of a newly discovered subunit, as evidenced by dissociation-recombination studies. The remaining three purified ADHInd forms have dual pH optima for ethanol oxidation at 7.0 and 10.0 and generate two new bands on starch gel electrophoresis after dissociation-recombination. They appear to be heterodimers of this new subunit with the known subunits, , 1, and 1. Based on the occurrence of these four ADHInd isozymes and isozymes containing 1 subunits in the homogenate supernatants of 135 livers, we conclude that ADHInd results from polymorphism at the ADH 2locus, with the variant ADH 2 Ind allele coding for the Ind subunit. The frequency of ADH 2 Ind was 0.16 in Black Americans, and this allele was not observed in any of the 63 livers from White Americans. Aims

Eurasian forest‐steppes are among the most complex non‐tropical terrestrial ecosystems. Despite their considerable scientific, ecological and economic importance, knowledge of forest‐steppes is limited, particularly at the continental scale. Here we provide an overview of Eurasian forest‐steppes across the entire zone: (a) we propose an up‐to‐date definition of forest‐steppes, (b) give a short physiogeographic outline, (c) delineate and briefly characterize the main forest‐steppe regions, (d) explore forest‐steppe biodiversity and conservation status, and (e) outline forest‐steppe prospects under predicted climate change.

Location

Eurasia (29°–56°N, 16°–139°E).

Results and Conclusions

Forest‐steppes are natural or near‐natural vegetation complexes of arboreal and herbaceous components (typically distributed in a mosaic pattern) in the temperate zone, where the co‐existence of forest and grassland is enabled primarily by the semi‐humid to semi‐arid climate, complemented by complex interactions of biotic and abiotic factors operating at multiple scales. This new definition includes lowland forest–grassland macromosaics (e.g. in Eastern Europe), exposure‐related mountain forest‐steppes (e.g. in Inner Asia), fine‐scale forest–grassland mosaics (e.g. in the Carpathian Basin) and open woodlands (e.g. in the Middle East). Using criteria of flora, physiognomy, relief and climate, nine main forest‐steppe regions are identified and characterized. Forest‐steppes are not simple two‐phase systems, as they show a high level of habitat diversity, with forest and grassland patches of varying types and sizes, connected by a network of differently oriented edges. Species diversity and functional diversity may also be exceptionally high in forest‐steppes. Regarding conservation, we conclude that major knowledge gaps exist in determining priorities at the continental, regional, national and local levels, and in identifying clear target states and optimal management strategies. When combined with other threats, climate change may be particularly dangerous to forest‐steppe survival, possibly resulting in compositional changes, rearrangement of the landscape mosaic or even the latitudinal or altitudinal shift of forest‐steppes.  相似文献
