From autoradiographical experiments on water-imbibing pea (Pisum sativum) or Phaseolus mungo cotyledons supplied with glucose-14C or iodoacetic acid-14C, it was shown that the compounds did not penetrate into the innermost part of the cotyledons but remained in the peripheral region during the early germination period. Changes in the rates of incorporation of 14C into alcohol from glucose-14C were examined during the early period of germination, and it was concluded that glucose supplied exo-genously was actively metabolized in the peripheral region. Endogenous alcohol formation was supposed to occur mainly in the peripheral region in the early phases of germination. On the basis of these results, the heterogeneity of the distribution of physiological activities in the cotyledon is discussed.  相似文献

Development of mitochondrial activities (state 3 respiration,respiratory control ratio, ADP/O ratio) in peanut cotyledonsoccurs over the first 5 d from the start of imbibition. Mitochondriain cotyledons with the axis attached develop better than inthose from which the axis has been removed. Initially, mitochondriaare deficient in cytochrome c, but after 2 d from the startof imbibition this deficiency is overcome. Mitochondrial developmentin attached cotyledons, as measured by state 3 respiration,respiratory control ratio, ADP/O ratio, and succinate dehydrogenaseand cytochrome oxidase activities, is severely impaired by cycloheximide.This indicates that de novo synthesis of proteins is necessaryfor mitochondria and their enzymes to develop, a situation whichis in sharp contrast to the situation in pea cotyledons. Electronmicroscope studies also show that there is an increase in thenumbers of mitochondria in peanut cotyledons with time afterthe start of imbibition. Two patterns of mitochondrial developmentexist in legumes: in imbibed peanut cotyledons respiratory activitiesincrease due to biogenesis of mitochondria, whereas in pea cotyledonsthe increases are due to improvement of pre-existing organelles  相似文献

The possibility that the alternative respiration (AR) is involvedin seed germination was followed during and after imbibitionby noting the changes in the sensitivity to the respiratoryinhibitors, KCN and salicylhydroxamic acid (SHAM). This wascarried out by measuring oxygen uptake of mitochondrial preparationsfrom storage cotyledons and of the whole axis in a selectionof legumes (Vigna mungo, V. radiata, V. angularis, V. sinensisPhaseolus vulgaris, Pisum sativum and Dolichos lablab). Theextent of participation of AR in the respiration of cotyledonarymitochondria was variable according to species and time afterimbibition, and varied even within the same genus ( Vigna).In some species, AR was not observed. As to the axis, SHAM treatmentby itself had little effect on the rate of respiration, but,when applied with KCN, reduced oxygen uptake to an extent greaterthan KCN treatment alone. The course of development of mitochondrial activities was studiedin the early stage of germination of Phaseolus mungo seeds.Mitochondrial activities (state 3 oxygen uptake rate, respiratorycontrol, and ADP/O values) increased, while the activities ofglycolytic enzymes (aldolase, glyceraldehyde-3-phosphate dehydrogenase,and pyruvate kinase) hardly changed, during the early periodof imbibition. The activities of mitochondrial enzymes (malateand succinate dehydrogenases, and cytochrome oxidase) increasedduring the period, but the rates were still low compared withthose of glycolytic enzymes. On the basis of these results,a significant difference in activation patterns between glycolyticand mitochondrial activities is discussed.  相似文献

The development of mitochondria in cotyledons during the initialstages following imbibition and the subsequent degenerationwere faster when the embryonic axis was attached. This was moreclearly demonstrated when mitochondrial activity was determinedusing malate or -oxoglutarate as substrate rather than NADHor succinate. Cycloheximide did not inhibit the initial developmentof mitochondria in attached cotyledons, although it inhibitedthe incorporation of 14C-amino acid into protein. Abscisic acidinhibited almost completely the initial increase in mitochondrialactivities of attached cotyledons. The activities of severalenzymes in mitochondrial fractions were almost the same in attachedand detached cotyledons during 4 d after, imbibition. The degenerationof mitochondria was not accompanied by the loss of enzymes.It was inferred that the initial development and the subsequentdegeneration of mitochondria in cotyledons during 4 d afterimbibition was brought about by the structural improvement anddisorganization of mitochondria, respectively. The initial differencesin the development of mitochondrial activities between attachedand detached cotyledons were suggested to be attributable todifferences in the development of the activities of electrontransfer pathway from endogenous NADH to ubiquinone.  相似文献

Comparisons of respiratory metabolism among the phloem, cambiumand xylem of mature carrot root were carried out and the followingresults were obtained. 1) The activity of O2 uptake on a dry weight basis in the cambiumwas higher than that in the xylem. The phloem showed the lowestactivity. 2) RQ was close to unity in all the three tissues.3) The inhibition rate of O2 uptake by malonate was considerablyhigh in the three tissues. The highest inhibition was observedin the cambium. 4) In the phloem and xylem the malonic inhibitionrate of 14CO2 release from G-U-14C was not so high as comparedwith that of O2 uptake. In the cambium, however, those of O2uptake and 14CO2 output were comparable. 5) Malonate treatmentdid not cause any significant change in RQ. 6) The C6/C1 ratiowas highest in the cambium and lowest in the phloem. From these it is concluded that the participation of the TCAcycle in respiration is great in all the three tissues of carrotroot, and among the three the cambium has the highest activityof the cycle. The metabolism of glucose and organic acids was studied in theearly phases of germination of Phaseolus mungo seeds. Duringthe first 5–6 h of imbibition, ethanol fermentation activelyoccurred and in the following stage the TCA cycle seemed tobe markedly activated. CO2 fixation seemed to be low, at leastin the early stage of germination. Aspartic acid was convertedactively into malic acid. This seemed to be a cause of the activesynthesis of malic acid observed.  相似文献

In the early stage of germination of Phaseolus mungo seeds,oxygen uptake was little inhibited by iodoacetate and sodiumfluoride, yet carbon dioxide output was strongly inhibited.The inhibitors also decreased alcohol formation severely inthis stage. It was shown that NaF attacked its target (enolase)and curtailed the glycolytic activity. On the basis of theseresults, a mechanism for the insensitivity of oxygen uptakeis proposed.  相似文献

Changes in respiration rate and in the contents of various constituentsduring the early period of germination of Phaseolus mungo seedswere studied. The course of the respiration developed in threephases. A sharp rise was observed in the first phase (Phasea), followed by the second phase (Phase b) of fairly constantrespiration rate. The respiration rate increases again in thethird phase (Phase c). The O2 uptake in Phases a and b was notinhibited by iodoacetate and fluoroacetate, while that in Phasec was inhibited. The contents of aspartic and glutamic acidsand amides were very high. A decrease of aspartic acid contentwas notable during the first few hours of germination. Citricand malic acids were the major organic acid constituents. Citricacid content in the seeds decreased, while that of malic acidremained unchanged. The leaching of malic acid into the soakingmedium was remarkable during the first 6 h of imbibition  相似文献

Changes in respiratory metabolism accompanied by callus formationin cultured explants of carrot root were followed and the followingresults were obtained. 1) When the explant was cultured on amedium containing kinetin and 2,4-D, active cell division occurredand resulted in callus formation by the 9th–12th days.2) Fresh weight remarkably increased after a lag-time of about5 days. Changes in protein content on fresh weight basis weresimilar to changes in fresh weight. 3) Respiration rate increasedduring the first few days, when growth could not be distinctlymeasured. Accompanying the rise in respiration, the C6/C1 ratioalso increased. As callus developed, the respiratory rate andC6/C1 ratio gradually decreased and RQ, became higher than unity.4) Alcohol dehydrogenase activity increased between the 4thand 9th days after culture. 5) When sub-cultured callus tissuewas fed with G-U-14C, some radioactivity was detected in thealcohol of the tissues. 6) These results suggest that duringthe first 4–6 days after culture the activity of the EMBDEN-MEYERHOF-PARNAS-TCApathway was remarkably increased and, as callus developed, therelative participation of the pentose phosphate pathway graduallyincreased and simultaneously alcohol fermentation occurred.
