ABSTRACT. Bloodstream forms of Trypanosoma musculi, cultured in Schneider's drosophila medium at room temperature, multiply and differentiate through a series of developmental stages into infective metacyclic trypomastigotes in 10 days. Oral inoculation with these culture forms into CBA mice produced a parasitemia similar to that produced by intraperitoneal infection with bloodstream forms except for a three-day longer prepatent period. The effects of temperature on the yield of in vivo modulatedchlorophyll fluorescence were measured in intact leaves of atrazineresistantand -susceptible biotypes of the weed Senecio vulgaris L. At25 ?C, the photochemical quenching (qQ of steady-state chlorophyllfluorescence was reduced by around 30% in the atrazine-resistantmutant as compared to the susceptible wild type, indicatinga higher reduction state of the primary electron acceptor QAof photosystem II in the former biotype. Moderately elevatedtemperatures (above 30 ?C) further increased the steady-stateconcentration of reduced in the mutant. Analysis of the temperature dependence of both the photochemicalquenching qQ and the initial fluorescence level Fo clearly indicateda drastically enhanced heat-sensitivity of the photochemicalapparatus in the atrazine-resistant Senecio leaves. The heat-inducedchanges in F0 and qQ were closely correlated, suggesting thatthe phenomenon responsible for the rise in F0 was also involvedin the inhibition of the photosynthetic electron flux. Low temperaturesalso affected reoxidation but, in contrast to heating, no apparent differences were observed in the behaviourof the two biotypes exposed to chilling stress. ABSTRACT. We describe the in vitro culture of Phytomonas species isolated from Euphorbia characias . The best choice between tested media was SDM-79, in which promastigotes, after 6 days of culture, reached cell densities as high as 4 × 107 cells/ml. Cells growing in LIT or MTL medium showed longer division times and lower cell densities. We succeeded in obtaining Phytomonas sp. amastigote and spheromastigote forms in modified GRACE's medium, yielding transformation rates of up to 70%. Electron microscopy studies were performed in order to characterize the ultrastructural features of these forms obtained in vitro. On the other hand, metabolic studies based on qualitative (nuclear magnetic resonance spectroscopy) and quantitative metabolic methods (enzymatic assays) showed that promastigote forms secreted mainly ethanol, acetate, glycine, glycerol, piruvate and succinate in SDM-79 medium, whereas the major metabolites found after transformation in modified Grace's medium were ethanol, acetate, glycine, piruvate and smaller amounts of glycerol. 相似文献

The effect of orthovanadate on the uptake of phosphate by agedpotato tuber tissue was investigated to study the relationshipwith plasma membrane ATPase activity. Vanadate inhibited therate of phosphate uptake by aged discs with a maximum effectat 500 µM (58% inhibition). When vanadate was added tothe ageing medium for 24 h, the subsequent rate of phosphateuptake was also markedly decreased (68% inhibition). The resultsshow that the inhibition by vanadate was not due to enhancedleakage of phosphate nor to a non-specific toxic effect. Furthermore,complementary experiments with erythrosin B and molybdate wereconsistent with the hypothesis that vanadate acts specificallyon the plasma membrane ATPase and that this enzyme is involvedin maintaining the driving force for active uptake of phosphate(via co-transport with protons) by storage cells of potato tubers. ABSTRACT We studied 31 Phytomonas stocks isolated from various hosts and a broad geographical range by isoenzyme electrophoresis (14 loci) and population genetics analysis. The total variability is considerable since many stocks share no allele. Population genetic analysis strongly suggests that Phytomonas zymodemes behave as natural clones, as already proposed by us for several other protozoan species. These clones should be considered as actual taxa in all applied studies. Latex plants and phloemic plants (coconut and palm trees) harbor distinct sets of clones; hence, latex plants studied in this article are probably not a reservoir for parasites of the coconut and palm tree. 相似文献

A new sand-dwelling ciliate, Discotricha papillifera n.g., n.sp., is described and allocated to the trichostome family Trichopelmidae. The species seems to be one of the most highly differentiated members of the family. The peculiar type of its body ciliature and of its buccal structures suggests affinities with certain other major groups of ciliated protozoa; from an evolutionary point of view the organism appears to show close relationship to the gymnostome family Chlamydodontidae. 相似文献

The mosquito Culex pipiens is subjected to organophosphate treatments in both Corsica and southern Sardinia, but the resistance gene A2-B2, which is currently in a worldwide expansion, has only reached Sardinia. In order to understand this situation, the genetic structure of populations sampled in Sardinia and Corsica was assessed using 15 isozymes. Two loci (HK1 and HK2) were not taken into account because of the possibility of selection. For trie other loci, statistical independence was not rejected for all possible pairs, and no deviation from Hardy-Weinberg expectations was apparent. Low but significant genie differentiation was present between Corsica and Sardinia, as well as between northern and southern Sardinia, despite a large number of effective migrants per generation. These results are discussed in the context of the high probability of extinction/recolonization of breeding sites, the flight migration ability of this mosquito, and the pleiotropic cost of insecticide resistances genes. It is concluded that A2-B2 resistance is unlikely to reach Corsica from southern Sardinia, unless accidental human transportation occurs. 相似文献
