Microsatellite loci were isolated from Carnaby's black cockatoo (Calyptorhynchus latirostris: Aves), a highly valued, endangered, and endemic species of bird from Western Australia. This study describes three dinucleotide and one tetranucleotide microsatellite loci for which the primers produced clear and polymorphic amplification patterns with between two and nine alleles and moderate levels of variability. Two additional dinucleotide markers which were monomorphic in the Carnaby's cockatoo were able to amplify and were polymorphic in two other species of black cockatoo, greatly increasing the utility of these markers.  相似文献

We assessed the extent to which plants can acquire amino acids when supplied as single N-sources or when plants have access to a mixture of amino- and inorganic N sources. Because the uptake of different N-sources is temperature-dependent, the effects of temperature on amino-N uptake were also tested. Lolium perenne (perennial rye-grass) was grown hydroponically at 11 °C or 21 °C. Uptake of N was determined using 15N tracers at the growth temperature from solutions containing either nitrate, ammonium or glycine as single N sources and from a mixture containing all three N-forms. Estimates of the relative importance of amino acids such as glycine to the total N budget of plants will have been underestimated in studies where uptake was determined in single source solutions compared with those from solutions containing a mixture of N-forms. The proportion of total N acquired from the mixed N source as ammonium increased as temperature was reduced. Regarding the uptake and initial metabolism of glycine, uptake was probably the rate limiting step at 11 °C whilst it was the metabolism of glycine to serine at 21 °C. Although 15N incorporation into the plant amino-N pool was generally in proportion to the abundance of individual amino acids, its incorporation into the glycine pool was sometimes significantly less than predicted.  相似文献

Net copper uptake by cellulose discs, isolated root cell walls,and by live and dead roots of whole ryegrass seedlings, werestudied using 64Cu as a tracer. Uptake by cellulose discs stoppedafter around 10 h while uptake by isolated root cell walls continuedfor up to 50 h. An initial fast phase of uptake consisting predominantlyof cell wall adsorption was similar in live and dead tissuefor up to 19 h. A slower phase of uptake continued for up to50 h, greater in live than in dead tissue, the slower phaseof uptake in live tissue consisting of both a living and a deadcomponent. Based on these results, an alternative to the desorptionmethod for estimating the apoplastic contribution to total copperuptake is presented. Time-course studies with seedlings givena variety of growing solution/uptake solution regimes, and therelationship between copper uptake and external copper concentration,for short (4.8 h) and long (42.4 h) term uptakes, suggest thatdiffering contributions of cell wall adsorption and symplasmicabsorption may be responsible for differing effects of externalcopper concentration on uptake being expressed by the same tissue.Water flux had little effect on total uptake of copper althougha possible effect on absorption could not be ruled out. Plants of Lolium perenne L. cv. S23 were grown in sand culturesupplied with either ammonium (NH4+) or nitrate (NO3)in an otherwise complete nutrient solution at 12°C or 20°C.Three weeks after germination, plants were clipped weekly tosimulate grazing. After 10 weeks growth all nitrogen (N) wassupplied enriched with 15N to quantify the effects of form ofN supply and temperature on the relative ability of currentroot uptake and remobilization to supply N for laminae regrowth. The form of N supply had no effect on the dry matter partitioning,while at 20°C more dry weight was allocated to laminae regrowthand less to the remaining plant material. The current root uptakeof N, which subsequently appeared in the laminae regrowth, wassimilar for plants supplied with NH4+ or NO3, and bothwere equally reduced at the lower temperature of growth. Remobilizationof N to laminae regrowth was greater for plants receiving NH4+than NO3; remobilization with either form of N supplywas reduced at the lower temperature of growth. Remobilizationwas reduced to a lesser extent at 12°C than current rootuptake. It was concluded that remobilization became relativelymore important in supplying N for regrowth of laminae at lowertemperatures. Seeds of cauliflower cv. Hipop and Brussels sprouts cv. Asmer Aries were aged at 20% moisture content for 24 h; all seeds retained a germination of over 70% after ageing although the mean germination time increased. Prolonged aerated hydration for up to 32 h at 20°C followed drying resulted in improved performance of both unaged and aged cauliflower seeds and aged Brussels sprouts. Thus, all seed showed reductions in the mean germination time to the extent that after 32 h hydration the aged cauliflower seeds performed as well as high quality unaged seed. The improvement of aged seeds was also revealed an increase in germination after the controlled deterioration test following up to 24 h (cauliflower) or 32 h (Brussels sprouts) aerated hydration. This increase was indicative of a decrease in the extent of deterioration present after aerated hydration. Deleterious effects of prolonged hydration were observed in Brussels sprouts after 32 h although these may be explained desiccation injury after treatment since radicle emergence had occurred during hydration. The improvements in seed performance may be explained the activation of metabolic repair occurring during the early part of the hydration period therereducing the extent of deterioration that has been sustained during ageing, with further improvements due to the advancement of the germination process.  相似文献

The ground parrot (Pezoporus wallicus) has a coastal distribution in Victoria and has declined in range since European colonization. Its habitat consists of two major vegetation communities: coastal heathland and sedgeland. These are further divided into various sub-communities. These habitats contained high densities of cyperaceous and restionaceous plants, the seeds of which form the bulk of the bird's diet. Seeds and small fruits of some dicotyledonous plants and grasses are also eaten. Seed production by sedges remains fairly constant over time in sedgelands but varies in heathland, depending on time since burning. This variation is reflected by parallel changes in ground parrot population density. Both long unburnt and very frequently burnt heathlands are unsuitable for ground parrots. The birds also require dense vegetation cover. Ground parrots appear to live in territorial breeding pairs with enforced natal dispersal. This is an adaptation to an environment where habitat is patchily distributed in space and variable in quality over time.  相似文献
