The oriental persimmon (Diospyros kaki Lf) is believed to have originated in China with subsequent introduction into Japan and Korea in ancient times. The species was then brought to Europe, Brazil and the USA from Japan in the 19th century. Recent studies highlighted the poor state of identification of cultivars in these countries due to incorrect labelling and presence of synonyms among local varieties. Thus, molecular marker characterization of germplasm resources is of great value for genetic resource preservation and plant breeding of persimmon. Therefore, to identify accessions for further plant breeding and germplasm management, 37 microsatellite loci were developed from a CT/AG‐enriched persimmon genomic library.  相似文献

Different serotypes of salmonellas were compared for selectivity and efficiency of recovery using 11 plating media. No optimal growth was obtained after 24h incubation in any of the media, but after 48h, brilliant green, brilliant green-phenol red-lactose-sucrose, bismuth sulphite, xylose-lysine-deoxycholate and Hektoen enteric agars showed optimal recovery of all the salmonella serotypes.
Xylose-lysine-deoxycholate and brilliant green-phenol red-lactose-sucrose agars were the most selective media for all salmonella serotypes. Addition of 10 μg/ml of sodium novobiocin to the tryptic soy-xylose-lysine and tryptic soy-brilliant green agars significantly improved their selectivity but reduced or inhibited the growth of some salmonella serotypes, including Salmonella typhi. Xylose-lysine-deoxycholate agar gave the highest recovery percentage of stressed salmonellas with a double-agar layer technique. Good recovery was also obtained on brilliant green-phenol red-lactose-sucrose, tryptic soy-brilliant green, tryptic soy-brilliant green-novobiocin, tryptic, soy-xylose-lysine and tryptic soy-xylose-lysine-novobiocin agars. Salmonella-shigella agar was the least efficient medium for the recovery of salmonellas under stress-induced or non-stressed conditions.  相似文献

Annual sweetclover plants [Melilotus segetalis (Brot) Ser.] were grown for a complete life cycle with and without saline (NaCl treatment of CE=15 dS m−1). Growth and partitioning analyses were performed. Sequential harvests (every 15 d) during the life cycle, and separation of plant material into roots, stems, petioles, leaves and reproductive structures were carried out Salt treatment reduced growth during the early and middle stages of the life of the plant, but did not significantly affect RGR in the reproductive phase. The root–shoot allometric coefficient of salinized plants in the generative phase decreased more than in control plants. We suggest that salinity-induced growth reduction in M. segetalis was primarily a result of a lower unit leaf rate (ULR) despite an increased leaf area ratio (LAR). Earlier flowering, higher biomass allocation to shoot and greater reproductive investment, but similar relative growth rate (RGR), were some of the main characteristics of salt-stressed plants compared to controls during the reproductive phase, these apparently being associated with increased sink strength caused by developing flowers and fruits.  相似文献

By means of hand pollination experiments and fluorescence microscopythe breeding systems and pollen tube growth were studied inDolichandra cynanchoides and Tabebuia nodosa, two bignoniaceousspecies of the Chaco woodland of NE Argentina. Both speciesabscised all selfed pistils within 4–8 d although selfpollen tubes grew successfully to the ovary and penetrated manyovules. Both species therefore show ‘late-acting self-incompatibility’(LSI) or ‘ovarian sterility’ (OS) type control ofselfing. LSI-OS has been reported in 17 species of the Bignoniaceae,and is also clustered in families such as the Bombacaceae-Sterculiaceae,Fabaceae and Myrtaceae. The implications of such family clusteringare discussed, with regard to current hypotheses for the actionof LSI-OS phenomena. The submicroscopic morphology of the vitelline envelope of Bufo arenarum's oocyte change significantly during the maturation and fertilization processes. The morphological changes are related to physiological activity in vivo and can be triggered in vitro by experimental procedures. It is our scope to present the ultrastructure differences of fascicular components of the vitelline envelope in coelomic, "pars recta" conditioned, oviductal, oviposited and fertilized oocytes. Our experimental results indicate that artificial "pars recta" treatment of coelomic oocytes arrange the fascicular components as those of oviposited oocyte, although differences still remain indicating that additional maturation processes take place while the egg pass througth the oviduct. Fertilized or activated oocytes which are refractary to sperm penetration, change the vitelline envelope fascicular components orientation giving a submicroscopical image quite different to those of none fertilized oocytes. These ultrastructural changes define in a clear cut manner the functional states of Bufo arenarum's oocyte.  相似文献

Summary

The development and improvement of artificial incubation techniques for freshwater crayfish eggs and their incorporation into the working schedule of breeding centres is of great interest for commercial production. Factors such as the water circulation system, flow rate, thermal treatment, etc., could strongly influence the success of the process. The present study attempts to test the possible influence of one of these variables, the thermal regime, on both the duration of embryonic development and the efficiency rates obtained in the artificial incubation of white-clawed crayfish (Austropotamobius pallipes) eggs. Four different thermal treatments were tested (three of them included a period at low temperature: 4°-5°C). Survival rates to juvenile stage 2 were similar in the four cases, ranging between 66.7 and 72.7%. We conclude that water cooling (an expensive management procedure) is not necessary in astacid breeding centres provided that egg development takes place at moderately low temperatures (8°-10°C) with a subsequent increase of up to 15°C from the eyed stage. However, the inclusion of periods at low temperature (4°-5°C) allows the staggered production of juvenile batches throughout a 3-week period without adverse effects on efficiency rates. This could be useful to breeding centres in meeting seasonal market requirements. In our study, egg and juvenile losses (mortality rate: 15–20%) were concentrated during the last phases of embryogenesis, particularly from the eyed stage to juvenile stage 2, during which they amounted to more than 90% of the overall mortality which took place during the artificial incubation process.  相似文献

Background aims

Multipotent mesenchymal stromal cell (MSC)-based medicines are extensively investigated for use in regenerative medicine and immunotherapy applications. The International Society for Cell and Gene Therapy (ISCT) proposed a panel of cell surface molecules for MSC identification that includes human leukocyte antigen (HLA)-DR as a negative marker. However, its expression is largely unpredictable despite production under tightly controlled conditions and compliance with current Good Manufacturing Practices. Herein, we report the frequency of HLA-DR expression in 81 batches of clinical grade bone marrow (BM)-derived MSCs and investigated its impact on cell attributes and culture environment.

Methods

The levels of 15 cytokines (interleukin [IL]-1β, IL-4, IL-6, IL-10, IL-17A, IL-17F, IL-21, IL-22, IL-23, IL-25, IL-31, IL-33, interferon-γ, soluble CD40 ligand and tumor necrosis factor-α) were determined in sera supplements and supernatants of BM-MSC cultures. Identity, multipotentiality and immunopotency assays were performed on high (>20% of cells) and low (≤20% of cells) HLA-DR+ cultures.

Results

A correlation was found between HLA-DR expression and levels of IL-17F and IL-33. Expression of HLA-DR did neither affect MSC identity, in vitro tri-lineage differentiation potential (into osteogenic, chondrogenic and adipogenic lineages), nor their ability to inhibit the proliferation of stimulated lymphocytes.

Discussion

Out of 81 batches of BM-MSCs for autologous use analyzed, only three batches would have passed the ISCT criteria (<2%), whereas 60.5% of batches were compliant with low HLA-DR values (≤20%). Although a cause–effect relationship cannot be drawn, we have provided a better understanding of signaling events and cellular responses in expansion culture conditions relating with HLA-DR expression.  相似文献

The post-larval development of the reproductive system in Nassariusvibex was investigated using a histological approach, to establishthe sequence of organogenesis and the contribution of differentorganogenetic components to the adult system. This researchis part of a broader investigation with two primary objectives.The first is to provide a detailed analysis of reproductivesystem development that can be used to re-evaluate existingcharacter sets and generate new characters for reconstructing gastropodphylogenies. The reproductive tract has always been a promisingbut problematic system to work with in phylogenetic research,because of the uncertain homology of many parts of the system.Most of the reproductive system itself develops during post-larvalontogeny, a phase of gastropod development that has been investigatedin very few taxa; thus this line of research has the potentialto provide a wealth of new information. The second objective isto establish comparable organogenetic series for representative gastropodtaxa that can be used to investigate the significance of heterochronicprocesses in the evolution of gastropod organ systems. The reproductive system in Nassarius vibex develops very latein ontogeny from two ontogenetic components in the male andthree in the female. Development and differentiation of theorgan system is not completed until near maturity. In both sexes,ductal components develop first and fuse to form the rudimentaryreproductive tract. Gonads and accessory reproductive structuresdevelop after the ductal components are fused, and glandulartissues do not appear until near maturity. The penis does notdevelop until near maturity. The capsule and albumen glandsin this species are broadly conjoined, unlike the situationin other members of Nassarius. Abstract Argentine haemorrhagic fever (AHF) is caused by Junin (JUN) virus, which is hosted by the drylands vesper mouse (Calomys musculinus). In this work we monitored population abundance of C. musculinus and rodent assemblages for 3 years in and outside the AHF endemic zones (northern Buenos Aires, southern Córdoba and Santa Fe Provinces, Argentina). The study area was divided into endemic and nonendemic zones. In the endemic zone epidemic sites were recognized, characterized by recent emergence and maintenance of AHF cases, and also historical sites, characterized by decreased incidence or disappearance of AHF human cases. In the nonendemic zone AHF has never been recognized. Although differences were statistically significant only during some periods, population abundance of C. musculinus was usually lower in the nonendemic sites. The pattern and magnitude of seasonal fluctuations in C. musculinus populations were also distinct in the nonendemic sites as compared to endemic sites. The relative abundance of C. musculinus in rodent assemblage was lower in nonendemic sites than in the endemic sites. The lower population densities and dampened seasonal dynamics may be at least partly responsible for the absence of AHF cases in the nonendemic zone. It is suggested that the balance between intra and interspecific interactions might be the cause of the pattern of incidence and prevalence of pathogens in the host species.  相似文献
