Vast parts of the monsoon tropics of Australia feature semi‐arid habitats, which are generally thought to harbour a depauperate land snail fauna as compared to the mesic continental fringes, in particular the Australian wet tropics. However, our knowledge of land snails inhabiting these often remote environments is still very patchy. In order to improve the understanding of land snail diversity in the monsoon tropics, we revised the camaenid land snail genus Exiligada based on comprehensive collections, undertaken by use of helicopters on remote limestone outcrops in the Northern Territory and in Western Australia. Based on comparative analyses of shell and genital morphology and patterns of molecular differentiation, we recognize 15 species within Exiligada, 13 of which are newly described. In addition, we suggest a revised delimitation of the type species Exiligada negriensis, as compared to the latest available revision, by removing Exiligada qualis from its synonymy and recognizing it as a distinct species. A key for species identification is also provided. Molecular phylogenetic analyses strongly supported the monophyly of Exiligada with respect to other confamilial genera known to occur in the same geographical region. Most Exiligada species are narrowly endemic to restricted limestone outcrops that cover areas with a diameter of about 20 to 150 km. Within the distributional range of Exiligada, the ranges of up to seven species overlap but we never found more than three species to occur in sympatry at a given sampling site. We propose that species originated by allopatric differentiation, followed by secondary range expansion, leading to sympatric distributions. Our study confirms that less complex rock habitats in more xeric environments support no more than three sympatric species, this being likely to be a result of ecological exclusion. © 2012 The Linnean Society of London, Zoological Journal of the Linnean Society, 2012, 166 , 689–722.  相似文献

Identification of genes involved in local adaptation is particularly challenging for species functioning as a network of interconnected populations undergoing frequent extinctions–recolonizations, because populations are submitted to contrasted evolutionary pressures. Using amplified fragment length polymorphism markers, population genetic structure of the mosquito Aedes rusticus was analysed in five geographical areas of the French Rhône‐Alpes region. We included a number of sites that were treated with the bio‐insecticide Bacillus thuringiensis israelensis (Bti) for more than 15 years. Analysis of molecular variance revealed that most of the genetic variability was found within populations (96%), with no significant variation among geographical areas, although variation among populations within areas (4%) was significant. The global genetic differentiation index FST was low (0.0366 ± 0.167). However, pairwise FST values were significant and no isolation‐by‐distance at the regional level was observed, suggesting a metapopulation structure in this species. Bti‐treatment had no effect on genetic structure and on within‐population genetic diversity. Potential signatures of positive selection associated with Bti‐treatment were detected for five loci, even though toxicological bioassays performed on field‐collected larvae showed no significant difference in mortality between Bti‐treated and nontreated sites. The difficulty of detecting moderate resistance in field‐collected larvae together with possible differential persistence of toxins in the environment may explain our inability to detect a toxicological response to Bti in treated sites. The evidence for positive selection occurring at several genomic regions suggests a first step towards Bti resistance in natural mosquito populations treated with this bio‐insecticide. Furthermore, this signal was detectable using genomic tools before any toxicological evidence for resistance could be identified.  相似文献

Frost avoidance and freezing tolerance in Afroalpine 'giant rosette' plants   总被引:5,自引:2,他引:3  
Abstract. The large leaf rosettes of the so-called 'giant rosette' plants which inhabit the alpine region of tropical mountains are composed of a great number of adult leaves surrounding a central cone of developing leaves. Upon onset of the nocturnal frost period the adult leaves nyctinastically bend inwards and form a night-bud around the central leaf bud. The insulating effect of the night-bud was analysed in four species: Senecio keniodendron, Senecio brassica. Lobelia keniensis and Lobelia telekii which grow on Mt Kenya (Kenya). Freezing is avoided by a delay of cooling which is sufficient until rewarming by the next day's sunshine. A consequence of this delay is that the temperature in the nocturnal bud remains higher than that of the outer leaves which are often stiffly frozen after cold nights. Only one freezing point was detected on the leaf temperature recordings. Depending on the water state, the freezing points were in the range — 1°C to —4.2°C. Sucrose, amounting to 38% of the leaf dry weight, may act as a cryoprotectant for the cell membranes. Frost hardiness of the leaves, as determined with a laboratory method, was sufficient to enable the plants to survive during the nocturnal frost temperatures as measured in the field.  相似文献

We have expressed the Paramecium calmodulin gene in Escherichia coli by changing the four TAA codons in this gene to CAAs. This was carried out by three polymerase chain reactions (PCRs) and then cloning the product into the expression vector pKK223-3 immediately downstream of its trp-lac hybrid promoter. JM109 strain of E. coli , transformed with the recombinant plasmid harboring the altered Paramecium calmodulin gene, produces a protein judged to be calmodulin. It is recognized by a monoclonal antibody to Paramecium calmodulin; it migrates with the native protein at nearly the same rate in electrophoreses; and it shows a Ca2+-dependent shift in electrophoretic pattern. The production of calmodulin is about 170 times as efficient with E. coli as with Paramecium in terms of unit volume of packed cells, and is about 400 times as efficient in unit volume of liquid culture. This method appears useful in site-directed mutageneses and in the heterologous productions of other ciliate proteins. A critique of this method is provided. A calmodulin half-molecule, a by-product of this project, is described.  相似文献

Arbuscular mycorrhizal (AM) fungi form mutualistic symbioses with the root systems of most plant species. These mutualisms regulate nutrient exchange in the plant–soil interface and might influence the way in which plants respond to increasing atmospheric CO2. In other experiments, mycorrhizal responses to elevated CO2 have been variable, so in this study we test the hypothesis that different genera of AM fungi differ in their response, and in turn alter the plant's response, to elevated CO2. Four species from three genera of AM fungi were tested. Artemisia tridentata Nutt. seedlings were inoculated with either Glomus intraradices Schenck & Smith, Glomus etunicatum Becker & Gerdemann, Acaulospora sp. or Scutellospora calospora (Nicol. & Gerd.) Walker & Sanders and grown at either ambient CO2 (350 ppm) or elevated CO2 (700 ppm). Several significant inter-specific responses were detected. Elevated CO2 caused percent arbuscular and hyphal colonization to increase for the two Glomus species, but not for Acaulospora sp. or S. calospora . Vesicular colonization was not affected by elevated CO2 for any fungal species. In the extra-radical phase, the two Glomus species produced a significantly higher number of spores in response to elevated CO2, whereas Acaulospora sp. and S. calospora developed significantly higher hyphal lengths. These data show that AM fungal taxa differ in their growth allocation strategies and in their responses to elevated CO2, and that mycorrhizal diversity should not be overlooked in global change research.  相似文献

The length of root hairs of wheat (Triticum aestivum L.) growingin Long Ashton nitrate nutrient solution was affected mainlyby pH and the concentrations of calcium and nitrate. Root hairlength was decreased by high pH (> 7.0) and by low calciumconcentrations but was increased by nitrate. Buffering at pH6.5 was found to affect the relationship between nutrient solutionstrength and the length of root hairs. The interactions betweencalcium, nitrate and buffering were determined. Phosphate concentrationdid not affect the length of root hairs of wheat, although lowconcentrations have been reported to stimulate root hair growthin other plants.
