A leaf disease of sorghum (Sorghum bicolor) characterised by fine discontinuous chlorotic streaks between the veins, was observed on sorghum grown during the 1987/88 post-rainy season in peninsular India. Early-infected plants were stunted, had shortened internodes, and produced poorly developed panicles. The virus was transmitted by the delphacid planthopper, Peregrinus maidis. Negatively stained leaf dip preparations contained bullet-shaped virus particles (208 ± 4.4 × 66 ± 1.0 nm) resembling those of rhabdoviruses. In ultrathin sections, the particles budded through the inner nuclear membrane and were present in the cytoplasm within membrane-bound vesicles that were apparently contiguous with the distended outer nuclear membrane. A method for purifying the virus was developed utilising polyethylene glycol (PEG) precipitation, Celite filtration and sucrose densitygradient centrifugation. An antiserum was produced in rabbits with a titre of 1/2650 in the precipitin ring interphase test. The virus could be detected in infected sorghum leaf tissues using a direct antigen coating form of enzyme-linked immunosorbent assay (DAC-ELISA). In immuno-double diffusion tests, the virus reacted positively with antisera to maize mosaic virus (MMV) from Reunion (MMV-RN) and Hawaii (MMV-HI), but not with antisera to barley yellow striate mosaic (BYSMV), cereal chlorotic mottle (CCMV), and cynodon chlorotic streak (CCSV) viruses. Thus, the virus isolated from sorghum is designated the MMV-S isolate. In DAC-ELISA tests, MMV-S reacted positively with antisera to MMV-R, MMV-HI, MMV-Florida isolate, CCSV, and CCMV, and weakly with antiserum to BYSMV. SDS-polyacrylamide gel electrophoresis revealed four major proteins of relative mass Mr 70 000, 59 000, 32 000 and 28 000. In electro-blot immunoassay, MMV and CCSV antisera detected the G and N proteins. These data suggest that MMV-S should be placed in the sonchus yellow net virus subgroup of plant rhabdoviruses.  相似文献

Five new species of the genus Myxobolus (Myxosporea) infecting the fresh-water carps are described. M. bhadrensis n. sp. was noticed in the musculature of the fry of Labeo rohita, collected from Bhadra Fish Seed Farm, in September 1979; and M. shettii, M. vedavatiensis, M. venkateshi, and M. hosadurgensis n. spp. were found in the gills of Cirrhina mrigala sampled from Vanivilasa Sagar Fish Farm in April 1980. Also, a new host record for M. curmucae is reported.  相似文献

Through the use of a microthermocouple psychrometer it has becomepossible to measure the water potential over part of the surfaceof a pea seed, starting about 19 hours after sowing in distilledwater, or later in the case of seeds germinating at lower osmoticpotentials, in both instances until well after radicles haveemerged. It has been shown that the potential of air-dry seedsis well below –6,000 joules/kg but increases rapidly duringimbibition, depending upon the water potential of the germinationmedium. Pea seeds subjected to lower external water potentialsgerminate at lower internal water potentials than they exhibitin distilled water. The water potentials of the seeds decreasejust after radicle emergence till the radicle establishes contactwith its germination medium, possibly as a result of demandfor moisture during that period due to incipient cell elongation.No detectable amounts of osmotically active substances are exudedfrom the seed during germination; the pea seed coat restrictsthe entry of polyethylene glycol molecules into the seed untilemergence of the radicle.  相似文献

Drought is the major abiotic constraint affecting groundnut productivity and quality worldwide. Most breeding programmes in groundnut follow an empirical approach to drought resistance breeding, largely based on kernel yield and traits of local adaptation, resulting in slow progress. Recent advances in the use of easily measurable surrogates for complex physiological traits associated with drought tolerance encouraged breeders to integrate these in their selection schemes. However, there has been no direct comparison of the relative efficiency of a physiological trait‐based selection approach (Tr) vis‐à‐vis an empirical approach (E) to ascertain the benefits of the former. The genetic material used in the present study originated from three common crosses and one institute‐specific cross from four collaborating institutes in India (total seven crosses). Each institute contributed six genotypes and each followed both the Tr and E selection approaches in each cross. The field trial of all selections, consisting of 192 genotypes (96 each Tr and E selections), was grown in 2000/2001 in a 4 × 48 alpha design in 12 season × location environments in India. The selection efficiency of Tr relative to E, RETr, was estimated using the genetic concept of response to selection. Based on all the 12 environments, the two selection methods performed more or less similarly (RETr= 1.045). When the 12 environments were grouped into rainy season and post‐rainy season, the relative response to selection in Tr method was higher in the rainy than in the post‐rainy season (RETr= 1.220 vs 0.657) due to a higher genetic variance, lower G × E, and high h2. When the 12 environments were classified into four clusters based on plant extractable soil‐water availability, the selection method Tr was superior to E in three of the four clusters (RETr= 1.495, 0.612, 1.308, and 1.144) due to an increase in genetic variance and h2 under Tr in clustered environments. Although the crosses exhibited significant differences for kernel yield, the two methods of selection did not interact significantly with crosses. Both methods contributed more or less equally to the 10 highest‐yielding selections (six for E and four for Tr). The six E selections had a higher kernel yield, higher transpiration (T), and nearly equal transpiration efficiency (TE) and harvest index (HI) relative to four Tr selections. The yield advantage in E selections came largely from greater T, which would likely not be an advantage in water‐deficient environments. From the results of these multi‐environment studies, it is evident that Tr method did not show a consistent superiority over E method of drought resistance breeding in producing a higher kernel yield in groundnut. Nonetheless, the integration of physiological traits (or their surrogates) in the selection scheme would be advantageous in selecting genotypes which are more efficient water utilisers or partitioners of photosynthates into economic yield. New biotechnological tools are being explored to increase efficiency of physiological trait‐based drought resistance breeding in groundnut.  相似文献

However no evidence was foundof cytoplasmic autophagy, or, during maturation of the egg,of nucleo-cytoplasmic interaction of the kind reported in liverwortsand ferns. The internal lamellar system of the plastids dedifferentiatesalmost entirely, and there is a corresponding increase in thefrequency of osmiophilic droplets. Starch also disappears, andsimultaneously large quantities of mucilage appear in the cytoplasm.The mucilage is secreted into the venter of the archegoniumand envelopes the egg. A modified design of a thermocouple psychrometer is describedwhich utilizes the Peltier effect for producing an exceedinglysmall wet-bulb thermometer. The thermocouple assembly has beenreduced to miniature dimensions and the thermocouple chamberconsists of a silver cylinder (volume 0.06 cm3) whose base isformed by the material under observation. This makes it possibleto measure water potentials over limited surfaces of intactplant tissues, e.g. germinating pea seeds (Pisum sativum L.).The apparatus is capable of measuring water potentials downto a magnitude of about –6,000 joules/kg.3  相似文献
