L-lysine α-oxidase (LO) is an L-amino acid oxidase with antitumor, antimicrobial and antiviral properties. Pharmacokinetic (PK) studies were carried out by measuring LO concentration in plasma and tissue samples by enzyme immunoassay. L-lysine concentration in samples was measured spectrophotometrically using LO. After single i.v. injection of 1.0, 1.5, 3.0 mg/kg the circulating T1/2 of enzyme in mice varied from 51 to 74 min and the AUC0–inf values were 6.54 ± 0.46, 8.66 ± 0.59, 9.47 ± 1.45 μg/ml × h, respectively. LO was distributed in tissues and determined within 48 h after administration with maximal accumulation in liver and heart tissues. Mean time to reach the maximum concentration was highest for the liver—9 h, kidney—1 h and 15 min for the tissues of heart, spleen and brain. T1/2 of LO in tissues ranged from 7.75 ± 0.73 to 26.10 ± 2.60 h. In mice, plasma L-lysine decreased by 79% 15 min after LO administration in dose 1.6 mg/kg. The serum L-lysine levels remained very low from 1 to 9 h (< 25 μM, 17%), indicating an acute lack of L-lysine in animals for at least 9 h. Concentration of L-lysine in serum restored only 24 h after LO administration. The results of LO PK study show that it might be considered as a promising enzyme for further investigation as a potential anticancer agent.

This review summarizes data on the properties of L-lysine -oxidase, an enzyme that belongs to the group of oxidases of L-amino acids. This enzyme acts virtually only on L-lysine with a rather low K m yielding -keto--aminocaproic acid. The decrease in the level of the essential amino acid L-lysine and the formation of hydrogen peroxide during the reaction possibly provide the basis for the unique properties of L-lysine -oxidase: cytotoxic, antitumor, antimetastatic, antiinvasive, antibacterial, and antiviral activities, as well as an immunomodulating effect. Native L-lysine -oxidase and its immobilized forms are promising tools for determination of concentration of L-lysine in various biological materials.  相似文献

The paper deals with the distribution of genetic markers (systems ABO, MN, Rh (D), Hp, PTC) and a number of demographic (folding of arms, hand clasping, tongue rolling, right- and left-handedness, of the type of ear lobe, of the types of dermatoglyphic patterns) in the inhabitants of 6 villages in the Mezen District of the Archangelsk Region of the RSFSR (river Peosa basin). The data presented in this work were obtained in the course of examination of over 800 persons. Differences in the interpretation of the results of generally adopted methods of statistical analysis of samples from small populations are discussed. Among the systems analysed in one third of all the cases there was a statistically significant deviation from Hardy-Weinberg's ratios. For the MN blood groups and haptoglobins this was caused by the excess of heterozygotes. The test of Hardy--Weinberg's ratios at the level of two-loci phenotypes revealed no statistically significant deviations either in separate villages or in all the villages taken together. The analysis of heterogeneity with respect to markers inherited according to Mendel's law revealed statistically significant differences between villages in all the systems except haptoglobins. A considerable heterogeneity in the distribution of family names, the frequencies of some of them varying from village to village from 0 to 90%. Statistically significant differences between villages were shown for all the anthropogenetic characters except arm folding, hand clasping and right-left-handedness. Considering the uniformity of the environmental pressure in the region examined, the heterogeneity of the population studied is apparently associated with a random genetic differentiation (genetic drift) and, possibly, with the effect of the progenitor.  相似文献

L-Amino acid oxidases (L-AAO; EC 1.4.3.2) comprise a group of flavoproteins that catalyze oxidative deamination of L-alpha amino acids to corresponding alpha-keto acids, NH3 and H2O2. Most of these enzymes are homodimers with molecular mass of 100–150 kDa that exhibit antiviral, antifungal, antibacterial, and anticancer activity. Among this group of enzymes L-lysine alpha-oxidase (LO) is especially important as its biological effects may differ from the effects of other L-AAO, because this enzyme preferentially oxidizes L-lysine, the essential amino acid for the human body, without any practical effect on other amino acids. Since molecular mechanisms of the cytotoxic action of LO still require better understanding, in this study we have investigated a possible mechanism of action of LO from Trichoderma cf. aureoviride Rifai VKMF-4268D. A rat pheochromocytoma PC12 cell culture was used as a model. Using flow cytometry a dose-dependent cell death induced by LO was shown. The increase in intracellular reactive oxygen species detected by the 2,7-dichlorodihydrofluorescein assay suggests that the oxidative pathway is one of mechanisms underlying the cytotoxic LO action; however, this does not rule out the involvement of other (previously demonstrated) mechanisms of LO effects on cell death.  相似文献

The present work aims to investigate the kinetic characteristics of homodimer enzyme L-lysine α-oxidase from Trichoderma cf. aureoviride Rifai VKM F-4268D, taking into account allosteric effects. The enzyme was first shown to reveal positive cooperativeness, h=2.05±0.15. Using additional opportunities of Hill coefficient the value of the Michaelis–Menten constant has been estimated, Km=1.015?10?5М, indicating high strength of substrate binding to the active site of each subunit. High selectivity and absolute L-stereospecificity of the enzyme were shown. The inhibition of L-lysine conversion by non-cleavable lysine analogs as well as the reaction product was found out to take place. These effects have been evaluated only as the inhibition coefficients (%). A more detailed study of these inhibition effects was complicated because of the cooperativeness of enzyme subunits mentioned above. The kinetic scheme of L-lysine α-oxidase was proposed involving parallel-subsequent action of each of two subunits in the catalytic act.We think that the results obtained will be useful for studying the kinetic properties of other multi-subunit enzymes and improve understanding of the mechanisms of their action.  相似文献

The sequence of 2630 3'-terminal nucleotides has been determined for the genomic RNA of potato virus M (PVM), a type member of the carlavirus group. Analysis of this nucleotide sequence revealed five open reading frames coding for proteins of mol. wt. 25, 12, 7, 34 and 11 kDa (in 5'----3' direction). The PVM genome organization has been shown to be basically analogous to that of potexviruses, except that the latter at their 3' end lack the gene encoding 11K protein. Amino acid sequence comparison between the PVM 34K protein and the coat proteins of potexviruses has shown a high extent of homology in the C-terminal amino acids. Homology has also been found between the 25, 12 and 7K proteins encoded by PVM RNA and corresponding potexvirus proteins. All this gave grounds for suggestion that carlaviruses and potexviruses belong to the same subgroup of phytoviruses.  相似文献

In this article, we report on Some physiological aspects of the synthesis of extracellular L-lysine-??-oxidase (LO) by the fungus Trichoderma cf. aureoviride Rifai VKM F-4268D under salt stress conditions and discuss the possible role of this enzyme for the producer. It has been shown that The synthesis of extracellular LO and proteolytic enzymes is induced in the fungus T. cf. aureoviride Rifai VKM F-4268D during submerged cultivation on wheat bran under salt stress. It has been shown that LO biosynthesis is accompanied by H2O2 accumulation in the growth medium. It seems that the extracellular LO synthesis followed by hydrogen peroxide production under stress conditions provides an adaptive advantage for the producer fungus in its competition with other organisms.  相似文献

Spatial or temporal forest–peatland transition zones were proposed as potential hot spots of methane (CH4) emissions. Consequently, paludified soils are an important component of boreal landscape biogeochemistry. However, their role in the regional carbon cycle remains unclear. This study presents CH4 fluxes from two forest–peatland transition zones, two wet forest sites and two clear-cut sites which were compared to fluxes of open peatlands and dry forest. The median fluxes measured using the closed-chamber technique varied from ? 0.04 to 12.6 mg m?2 h?1 during three climatically different years. The annual mean CH4 emissions of the forest–peatland transition zone were significantly lower than the fluxes of the open peatland sites, 7.9 ± 0.5 and 21.9 ± 1.6 g m?2a?1, respectively. The dry forest site was characterized by a small uptake of CH4 (? 2.3 ± 0.2 g m?2a?1). Although clear-cut forest area drastically increased in European Russia during the last two decades, if water level depths in these forests remains below 10 cm they do not act as CH4 sources. Fluxes of CH4 from the transition zone sites showed a higher response to soil temperature than to water table level. Fluxes of CH4 between the atmosphere and the two investigated peatlands were not significantly different, although a significant difference in water table level could be observed. The meteorological conditions of the investigated summers changed from being hot and dry in 2013 to cold and wet in 2014; the summer of 2015 was characterized as warmer and drier in the first half and colder and wetter in the second half. Significant differences in CH4 fluxes were measured only between 2014 and 2013. Significant differences in CH4 fluxes and in nonlinear regressions showed that the CH4 fluxes of the different site types such as dry forests, transition zones and open peatlands need to be modelled separately on a landscape level. Obviously, underlying processes vary with the ecosystem and (along with regional aspects) have to be understood first before large-scale modelling is possible.  相似文献

L-lysine-α-oxidase (LO) synthesized by the fungus Trichoderma cf. aureoviride Rifai VKM F-4268D under salt stress conditions was isolated and characterized. The newly developed method for the isolation and purification of the enzyme was based on its precipitation from the culture liquid by copper sulfate. The subsequent LO purification by the methods of hydrophobic (Octyl Sepharose) and ion exchange (DEAE ToyoPearl) chromatography yielded a homogeneous enzyme preparation with a high degree of purification (310-fold) and high specific activity (90 U/mg protein). The molecular mass of the enzyme determined by gel filtration and native electrophoresis was 115–116 kDa. According to the data of SDS electrophoresis, LO was a dimer with identical subunits (57–58 kDa). The optical absorption spectrum of LO corresponded to the flavoprotein spectrum with maximums at 278, 390, and 465 (a shoulder at 490) nm. LO is a stereospecific enzyme oxidizing almost exclusively L-lysine (pH optimum 7.8–8.2). Insignificant activity was observed against L-ornithine and L-arginine. LO was shown to be stable at temperatures up to 50°C.  相似文献
