The evoked potential of the tectum opticum during electrical stimulation of the optic nerve was studied in acute experiments on the dogfishSqualus acanthias L. The negative phase of the "classical" negative-positive evoked potential of the contralateral hemisphere of the tectum opticum was shown to be a complex potential, including an early positive component. A similar potential also was recorded from the ipsilateral hemisphere. Enhancement of this positive potential on insertion of the recording electrode deep into the brain, its resistance to functional block on application of potassium chloride to the brain surface, and recording a similar potential from the surface of the floor of the third ventricle after extirpation of the tectum opticum are evidence of the nontectal location of the source of this evoked potential component. On the basis of existence of a focus of maximal activity in the rostral zones of the brain beneath the tectum opticum, and disappearance of the early positive component during functional block and extirpation of this brain region, it is concluded that a leading role in the generation of this component is played by thalamic nuclei.M. The distribution of evoked rhythmic responses on the surface and in the depths of the retina of the carp (Cyprinus carpio) and tench (Tinca tinca) and the dependence of the amplitude — frequency characteristics of the response on stimulus intensity and duration were investigated by recording the local electroretinogram and unit activity of the ganglion cells. Rhythmic on- and off-responses to light differed in various characteristics, including their distribution between the cellular and synaptic layers of the retina. The frequency, amplitude, and number of waves in the response were found to depend on the parameters of the stimulus. On-, off-, and on — off-ganglion cells were represented in the records. Their spike discharge usually corresponded to a phase angle of the negative half-wave of the rhythmic response of close to /2. The results of experiments in which conduction along the optic nerve was blocked by cold showed that the system of generation of the rhythmic response in the retina is under the tonic influence of the centers.M. A 30 kDa beta-galactose-specific lectin named CVL was isolated from the polychaete marine worm Chaetopterus variopedatus (Annelida) and its anti-HIV-1 activity in vitro was determined. Results showed that CVL inhibited cytopathic effect induced by HIV-1 and the production of viral p24 antigen. The EC(50) values were 0.0043 and 0.057 microM, respectively. Time-of-addition analysis of anti-HIV-1 activity indicated its action was at the early stage of virus replication. CVL could blocked the cell-to-cell fusion process of HIV infected and uninfected cells with an EC(50) of 0.073 microM. The inhibition of HIV-1 entry into host cells was demonstrated by using fluorescence-based real-time quantify PCR. At CVL concentration of 0.33 microM and 0.07 microM, 86% and 21% virus attachment were blocked, respectively. The anti-HIV-1 action of CVL might relate to blockade of HIV-1 entry into cells.  相似文献

In all secreted proteins related to the epidermal growth factor (EGF), EGF domains that occur in a mature factor are each encoded by two exons, and those that do not, by one exon. During splicing, additional exon 3a can be inserted between exons 3 and 4, which code for the EGF domain of the mature heparin-binding EGF-like growth factor (HB-EGF). The resulting mRNA codes for the short form of HB-EGF (SF HB-EGF), which retains the signal peptide, the propeptide, and the heparin-binding domain. However, its EGF domain lacks the C-terminal subdomain essential for the interaction with the EGF receptor (EGFR). Structural analysis suggested that SF HB-EGF is a secreted polypeptide that has high affinity for heparin but weakly, if at all, interacts with EGFR. Data obtained in three different systems indicated that SF HB-EGF possesses a mitogenic activity but utilizes a signal transduction pathway other than that of HB-EGF.  相似文献

Synaptic mechanisms of burst activity generation in certain neurons of the tectum opticum and mechanisms of generation of stimulation-induced group discharges by certain secondary neurons of the olfactory bulb were analyzed in carp (Cyprinus carpio L.). Spikes of the spontaneous discharge in neurons of the tectum were accompanied by depolarizing after-potentials, which caused the burst discharges of these cells. Evidence is given in support of the synaptic nature of the after-potential; it is suggested that it is generated by a recurrent collateral mechanism. Synaptic bombardment causing the appearance of a group discharge in olfactory bulb neurons and groups of spikes in their spontaneous activity was found to be intermittent in character. These features of unit activity in the olfactory bulb are shown to be connected with the presence of excitatory synaptic interaction between several neurons, probably dendro-dendritic in nature.M. V. Analysis of postsynaptic unit responses in the visual center ofRana temporaria showed that optic nerve fibers with high and low conduction velocities usually converge on a single neuron of the tectum opticum (TO). In response to stimulation of the optic nerve a complex depolarization potential consisting of 3 (or possibly 4) EPSPs was recorded in one group of neurons; these EPSPs were probably generated through excitation of several groups of afferent fibers. Either an increase or a decrease in the EPSPs can be observed in the TO neurons in response to repetitive and paired stimulation of the optic nerve. Postsynaptic inhibitory responses of some TO neurons, probably of direct and recurrent origin, are discussed.M. V. Lomonosov Moscow State University. The complex formation of lipopolysaccharide (LPS) with chitosan (Ch) was demonstrated using sedimentation velocity analysis in the analytical ultracentrifuge, centrifugation in glycerol gradient and isopicnic centrifugation in cesium chloride. An addition of Ch to the Escherichia coli and Yersinia pseudotuberculosis LPS solutions was found to result in formation of the stable LPS-Ch complexes. The interaction is a complicated process and depends on time and reaction temperature, as well as on the molecular weight of chitosan. A stable LPS-Ch complex could be formed only after preliminary incubation of the initial components at an elevated temperature (37 degrees C). It should be noted that process of LPS complexation with Ch is accompanied by additional dissociating of LPS. The complex formation was shown to be a result not only of ionic binding, but also of other types of interactions. The interaction of Ch with LPS was shown to modulate significantly the biological activity of LPS. The LPS-Ch complex (1:5 w/w) was shown to possess much lower toxicity in a comparison with the parent LPS at injection to mice in the similar concentration. The LPS-Ch complex was shown to maintain an ability to induce of IL-8 and TNF, but induction of IL-8 and TNF biosynthesis by the LPS-Ch complex was lower than that by the parent LPS. The complex LPS-Ch, similarly to the parent LPS, was found stimulated the formation of the IL-8 in the dose-dependent manner in the human embryonal kidney cells (HEK 293 cells) transfected with TLR4 in combination with MD2.  相似文献

Fucoidans and laminarans from Laminaria cichorioides, Laminaria japonica, Fucus evanescens, laminaran from Laminaria gurjanovae, other beta-D-glucans (translam, pustulan and zymosan) and lambda-carrageenan from Chondrus armatus were used to study the effect of water-soluble polysaccharides from seaweeds on the alternative pathway of complement (APC). beta-D-Glucans and fucoidans under study differed appreciably from each other by structural characteristics, and also by degree of purification. beta-D-glucans, on ability to bind complement, ranked in a line according to a degree of their purification. Highly purified beta-D-glucans under study did not reveal an ability to bind complement. The fucoidans were divided conventionally into three groups according to their action on APC. Highly sulfated alpha-L-fucan from L. cichorioides with the greatest activity toward APC and caused 50% inhibition of reaction of activation (RA) of APC in a concentration of 0.5-0.7 mg/ml. Opposite 50% of inhibition of lysis of erythrocytes by sulfated heterogeneous fucoidan from L. japonica was achieved with 20 mg/ml. All other fucoidans and lambda-carrageenan have activity at 6-10 mg/ml concentration. Decreasing the sulfate content from 36% up to 9% in sample fucoidans under study was not reflected practically in the 50% inhibition concentration. Apparently, the degree of sulfating of fucoidans did not influence their action on APC. But the positive influence of fucose in structure of polysaccharide was obvious.  相似文献

Evoked potentials to electrical stimulation of the spinal cord, its dorsal roots and the superficial orbital branch of the facial nerve were recorded in the tectum opticum of the Black Sea piked dogfishSqualus acanthias L. The distribution of evoked potentials over the surface and in the depth of the tectal lamina and also dependence of the parameters of the responses on the strength and frequency of the stimuli and intervals between them in the case of paired stimulation were studied. Evoked potentials were shown to be tectal in origin, to be recorded mainly contralaterally, and to exhibit specific dynamics of changes in response components under the influence of the various procedures and specific dependence of the responses on the location of the recording point. Significant differences were found between the characteristics of evoked potentials generated in superficial and deep layers of the tectum opticum. The nature and physiological role of nonvisual projections to this structure and also its role in the integrative function of the ichthyopsid brain are discussed.M. V.
