The interactions between chloroplast membranes and their microenvironment within artificial matrices (albumin-glutaraldehyde matrix, polyurethane foam) where investigated. Particularly, the influence of a high-ionic-strength medium (0.75 M potassium citrate) on the stability of the photosynthetic ferricyanide reduction by immobilized thylakoids has been studied. A method of data analysis based on a nonlinear identification method combined with the numerical integration of the equation of the transient state of the continuous stirred tank reactor (CSTR) is proposed to estimate the actual degradation of the photosynthetic electron transfer. A statistical analysis achieved on the parameter values has allowed a quantitative assessment of the global behavior of immobilized chloroplast membranes. From the mathernatical analysis of the experimental data, we demonstrate that citrate used in the reaction media prevents the photoinactivation of the electron transfer chain whatever the nature of the matrix or the type of the reactor. The use of an albumin-glutaraldehyde matrix or an open reactor during experiments also has allowed a better stabilization of the photosystems under operational conditions.  相似文献

This study was conducted to determine serum lipid levels and the activity of lipoprotein lipase in epididymal white adipose tissue of rats undergoing exercise training. During the 8-week period of treatment, one group of rats was kept sedentary and the remaining animals were exercise trained either continually (1 h of daily treadmill running) or intermittently (alternate weeks of daily running and inactivity). Exercise training, either continual or intermittent, decreased postprandial serum total and high-density lipoprotein cholesterol concentrations, which returned to sedentary levels in the intermittently trained animals following a week of rest. Lipoprotein lipase activity in whole epididymal adipose pad was lower in rats trained continually than in the sedentary group at the end of the treatment. The intermittent training program elicited large fluctuations in both the specific (per milligram of protein) and total (per tissue) activity of lipoprotein lipase in white adipose tissue. During rest periods, enzyme activity rose to levels that were higher than those of sedentary rats, whereas lipase activity was below that of sedentary animals following a week of running. In the last exercise--rest cycle, body weight gain of the intermittently trained rats was nearly abolished during the week of running, but it increased above that of sedentary animals during weeks of rest. The present results suggest that the modulation of lipoprotein lipase activity in white adipose tissue is one of the adaptations that take place to accommodate the fluctuations in the rate of energy deposition that occur in the rat during an intermittent training program.  相似文献

The degree and the origins of quantitative variability of most human plasma proteins are largely unknown. Because the twin study design provides a natural opportunity to estimate the relative contribution of heritability and environment to different traits in human population, we applied here the highly accurate and reproducible SWATH mass spectrometry technique to quantify 1,904 peptides defining 342 unique plasma proteins in 232 plasma samples collected longitudinally from pairs of monozygotic and dizygotic twins at intervals of 2–7 years, and proportioned the observed total quantitative variability to its root causes, genes, and environmental and longitudinal factors. The data indicate that different proteins show vastly different patterns of abundance variability among humans and that genetic control and longitudinal variation affect protein levels and biological processes to different degrees. The data further strongly suggest that the plasma concentrations of clinical biomarkers need to be calibrated against genetic and temporal factors. Moreover, we identified 13 cis‐SNPs significantly influencing the level of specific plasma proteins. These results therefore have immediate implications for the effective design of blood‐based biomarker studies.  相似文献

Malachite green was discovered independently by two researchers in Germany in the 19th century and found immediate employment as a dye and a pigment. Subsequently, other uses, such as staining biological specimens, emerged. A much later application was the control of fungal and protozoan infections in fish, for which the dye remains popular, although illegal in many countries owing to a variety of toxicity problems. In solution, malachite green can exist as five different species depending on the pH. The location of the positive charge of the colored cation on a carbon atom or a nitrogen atom is still debated. Through the canonical LC3 interaction motif (LIR), [W/F/Y]‐X1‐X2‐[I/L/V], protein complexes are recruited to autophagosomes to perform their functions as either autophagy adaptors or receptors. The lipase-catalyzed enantioselective esterification of ibuprofen has been studied in a media, composed only of substrates. When racemic ibuprofen is used, the alcohol-chain length affects the esterification rates of individual enantiomers, but it does not affect the enantioselectivity. Water activity affects the esterification rates of (R)- and (S)-ibuprofen differently, leading to higher enantioselectivity at lower water activities. Experiments were also conducted at various (R)- to (S)-ibuprofen ratios. It appears that the esterification rate of (R)-ibuprofen is always proportional to its concentration, whereas at low water activity the esterification rate of (S)-ibuprofen shows a saturation at higher concentrations. Other 2-phenyl carboxylic acids were studied, and the increase in apparent enantioselectivity at low-water activity was not observed for the molecules tested.  相似文献

A mutant of the cysteine protease papain, displaying nitrile hydratase and amidase activities, was expressed in Pichia pastoris and used for the hydrolysis of peptide nitriles in aqueous-organic media. The rate of hydrolysis of these nitriles is lowered by increasing acetone concentration. This is caused by an increase of the Michaelis constant, and a variation of Vmax proportional to the amount of water in the mixture. The hydrolysis of the amide is less affected by the increase in co-solvent, which results in lower accumulation of this intermediate product. With the peptide nitrile tested, high nitrile concentrations could be used to promote the production of the amide and prevent its hydrolysis to the acid by diminishing the relative rate of amide hydrolysis. A number of non-peptidyl nitriles were also tested as potential substrates but activity was detected for only one compound with structural resemblance to peptide nitriles.  相似文献
