A series of in vitro experiments were conducted to assess three fibrolytic enzyme preparations as potential feed additives in equine diets. The three fibrolytic enzyme preparations were a concentrated cellulase (E1), an acid cellulase (E2) and a concentrated xylanase (E3). The enzymes were evaluated on their ability to modify the cell wall fraction of high-temperature dried lucerne (HTL) under various experimental conditions including differences in temperature, pH, incubation period, substrate levels and particle size to enable selection of the enzyme preparation most effective in the hydrolysis of lucerne. Results showed enzyme activities (as measured by reducing sugar assays) to be greatest at 50 °C, pH 5 and over an incubation period of greater than 20 h. E1 exhibited the greatest effect on total monosaccharide release from the HTL compared to E2 and E3. Moreover, dry matter (DM) and total non-starch polysaccharide (TNSP) losses were also greater in HTL treated with E1 compared to E2 and E3. Therefore, since the cell wall fraction of HTL contained substantial amounts of cellulose, the enzyme with the highest cellulase activity (Enzyme 1) was most effective in hydrolysing the cell walls of HTL. Consequently, it would appear that the application of exogenous fibrolytic enzyme preparations to forages requires the chemical characterisation of the target forage to enable selection of enzymes that are (a) most suitable to degrade the cell wall components of the candidate forage and (b) effective under field conditions.  相似文献

Vegetative fronds of Spirodela polyrrhiza were induced to form dormant turions by the addition of 1 micromolar abscisic acid or by shading. The cell wall polymers of fronds contained a high proportion of the branched-chain pentose, d-apiose (about 20% of total noncellulosic wall sugar residues), whereas turion cell walls contained only trace amounts (about 0.2%). When the fronds were fed d-[3H]glucuronic acid for 30 minutes, the accumulated UDP-[3H]apiose pool accounted for about 27% of the total phosphorylated [3H]pentose derivatives; in turions, the UDP-[3H]apiose pool accounted for only about 4% of the total phosphorylated [3H]pentose derivatives. We conclude that the developmentally regulated decrease in the biosynthesis of a wall polysaccharide during turion formation involves a reduction in the supply of the relevant sugar nucleotide. One controlling enzyme activity is suggested to be UDP-apiose/UDP-xylose synthase. However, since there was a 100-fold decrease in the rate of polysaccharide synthesis and only a 9-fold decrease in UDP-apiose accumulation, there is probably also control of the activity of the relevant polysaccharide synthase.  相似文献

Previous studies on flagellar adhesion in chlamydomonas (Snell, W. and S. Roseman. 1979. J. Biol. Chem. 254:10820-10829.) have shown that as gametes adhere to flagella isolated from gametes of the opposite mating type, the adhsiveness of the added flagella but not of the gametes is lost. The studies reported here show that the addition of protein synthesis inhibitors (cycloheximide [CH] or anisomycin) to the medium of such cell- flagella mixtures causes the cells to lose their adhesiveness. This loss, however, occurs only after the cells have interacted with 4-8 flagella/cell and does not occur if the cells are kept in CH (7 h) without aggregating. The availability of an impotent (imp) mating type plus (MT(+)) mutant (provided by U.W. Goodenough), which adheres but is unable to undergo the fusion that normally follows adhesion, made it possible to determine whether a similar loss of adhesiveness occurs in mixtures of matting type minus (mt(-)) and imp mt(+) gametes. In the absence of inhibitor, mt(-) and imp mt(+) gametes adhered to each other (without fusing) for several hours; however, in the presence of CH or anisomycin, the gametes began to de-adhere 35 min after mixing, and, by 90 min, 100 percent of the cells were single again. This effect was reversible, and the rapid turnover of cells were single again. This effect was reversible, and the rapid turnover of molecules involved in adhesion occurred only during adhesion inasmuch as gametes pretreated for 4 h with CH were able to aggregate in CH for the same length of time as nonpretreated cells aggregated in CH. By the addition of CH at various times after the mt(-) and imp mt(+) gametes were mixed, measurements were made of the “pool size” of the molecules involved in adhesion. The pool reached a minimum after 25 min of aggregation, rapidly increased for the next 25 min, and then leveled off at the premixing level. These results suggest that flagellar adhesion in chlamydomonas causes modification of surface molecules (receptors, ligands), which brings about their inactivation and stimulates their replacement.  相似文献

Seeds of Indian ricegrass (Achnatherum hymenoides), a native bunchgrass common to sandy soils on arid western rangelands, are naturally dispersed by seed‐caching rodent species, particularly Dipodomys spp. (kangaroo rats). These animals cache large quantities of seeds when mature seeds are available on or beneath plants and recover most of their caches for consumption during the remainder of the year. Unrecovered seeds in caches account for the vast majority of Indian ricegrass seedling recruitment. We applied three different densities of white millet (Panicum miliaceum) seeds as “diversionary foods” to plots at three Great Basin study sites in an attempt to reduce rodents' over‐winter cache recovery so that more Indian ricegrass seeds would remain in soil seedbanks and potentially establish new seedlings. One year after diversionary seed application, a moderate level of Indian ricegrass seedling recruitment occurred at two of our study sites in western Nevada, although there was no recruitment at the third site in eastern California. At both Nevada sites, the number of Indian ricegrass seedlings sampled along transects was significantly greater on all plots treated with diversionary seeds than on non‐seeded control plots. However, the density of diversionary seeds applied to plots had a marginally non‐significant effect on seedling recruitment, and it was not correlated with recruitment patterns among plots. Results suggest that application of a diversionary seed type that is preferred by seed‐caching rodents provides a promising passive restoration strategy for target plant species that are dispersed by these rodents.  相似文献

Background

Activation by extracellular ligands of G protein-coupled (GPCRs) and tyrosine kinase receptors (RTKs), results in the generation of second messengers that in turn control specific cell functions. Further, modulation/amplification or inhibition of the initial signalling events, depend on the recruitment onto the plasma membrane of soluble protein effectors. High throughput methodologies to monitor quantitatively second messenger production, have been developed over the last years and are largely used to screen chemical libraries for drug development. On the contrary, no such high throughput methods are yet available for the other aspect of GPCRs regulation, i.e. protein translocation to the plasma membrane, despite the enormous interest of this phenomenon for the modulation of receptor downstream functions. Indeed, to date, the experimental procedures available are either inadequate or complex and expensive.

Results

Here we describe the development of a novel conceptual approach to the study of cytosolic proteins translocation to the inner surface of the plasma membrane. The basis of the technique consists in: i) generating chimeras between the protein of interests and the calcium (Ca2+)-sensitive, luminescent photo-protein, aequorin and ii) taking advantage of the large Ca2+ concentration [Ca2+] difference between bulk cytosolic and the sub-plasma membrane rim.

Conclusion

This approach, that keeps unaffected the translocation properties of the signalling protein, can in principle be applied to any protein that, upon activation, moves from the cytosol to the plasma membrane. Thus, not only the modulation of GPCRs and RTKs can be investigated in this way, but that of all other proteins that can be recruited to the plasma membrane also independently of receptor activation. Moreover, its automated version, which can provide information about the kinetics and concentration-dependence of the process, is also applicable to high throughput screening of drugs affecting the translocation process.  相似文献

Natural and anthropogenic boundaries have been shown to affect population dynamics and population structure for many species with movement patterns at the landscape level. Understanding population boundaries and movement rates in the field for species that are cryptic and occur at low densities is often extremely difficult and logistically prohibitive; however genetic techniques may offer insights that have previously been unattainable. We analysed thirteen microsatellite loci for 739 mountain lions (Puma concolor) using muscle tissue samples from individuals in the Great Basin throughout Nevada and the Sierra Nevada mountain range to test the hypothesis that heterogeneous hunting pressure results in source‐sink dynamics at the landscape scale. We used a combination of non‐spatial and spatial model‐based Bayesian clustering methods to identify genetic populations. We then used a recently developed Bayesian multilocus genotyping method to estimate asymmetrical rates of contemporary movement between those subpopulations and to identify source and sink populations. We identified two populations at the highest level of genetic structuring with a total of five subpopulations in the Great Basin of Nevada and the Sierra Nevada range. Our results suggest that source‐sink dynamics occur at landscape scales for wide‐ranging species, such as mountain lions, and that source populations may be those that are under relatively less hunting pressure and that occupy refugia.  相似文献

Although food abundance is a principal determinant of distribution and abundance of many animals, most previous studies have not quantitatively assessed its importance relative to other factors that may also determine species distributions. We estimated frugivorous phainopepla Phainopepla nitens occupancy and density, food density, and vegetation structure on transects in fragmented mesquite and acacia woodlands over three years in non‐breeding and breeding seasons. Using an AIC framework and controlling for detection probability, we determined relative impacts of food abundance, vegetation structure, and habitat fragmentation on patch occupancy and density, and concomitant extinction and colonization probabilities of phainopeplas. Initial occupancy in winter 2002 was high (0.87 ± 0.047), and primarily positively correlated with food abundance and woodland area (Akaike weights wi= 0.998 and 0.750 respectively). Woodland area more strongly influenced occupancy where food was scarcer. Phainopepla density in both seasons was strongly positively correlated with food abundance, especially in the 2002 drought when density was higher (wi=1.0 for food and year). Density was higher in acacia than mesquite woodlands (wi= 1.0), and moderately negatively correlated with elevation (wi= 0.789). Extinction probability (patches vacated) was low (0.078 ± 0.040), and principally influenced by phainopepla density (wi= 0.968) and tree height (wi= 0.749). Colonization probability was low (0.15 ± 0.034) and determined by vegetation structure (wi= 1.0). Much recorded colonization was reoccupancy of woodlands previously occupied by single males in winter, then vacated in a breeding season. These results suggest that for an animal occupying a highly fragmented landscape, distributions and densities at the habitat patch scale are driven by food abundance, are moderately affected by habitat fragmentation, and are slightly influenced by vegetation structure.  相似文献

Diplochory is seed dispersal by a sequence of two or more steps or phases, each involving a different dispersal agent. Here, we describe five forms of diplochory and derive general characteristics of each phase of seed dispersal. The first and second phases of diplochory offer different benefits to plants. Phase one dispersal often results in escape from density-dependent seed and seedling mortality near the parent plant and can result in the colonization of habitat patches far from the parent. Phase two dispersal often moves seeds to discrete and predictable microsites, where the probability of seedling establishment is disproportionately high (i.e. directed dispersal). Diplochory appears to have evolved because combining two means of seed dispersal can increase the benefits of seed dispersal whilst reducing the likelihood of seed mortality.  相似文献

DNA sequence comparisons of two mitochondrial DNA genes were used to infer phylogenetic relationships among 17 Felidae species, notably 15 in the previously described pantherine lineage. The polymerase chain reaction (PCR) was used to generate sequences of 358 base pairs of the mitochondrial 12S RNA gene and 289 base pairs of the cytochrome b protein coding gene. DNA sequences were compared within and between 17 felid and five nonfelid carnivore species. Evolutionary trees were constructed using phenetic, cladistic, and maximum likelihood algorithms. The combined results suggested several phylogenetic relationships including (1) the recognition of a recently evolved monophyletic genus Panthera consisting of Panthera leo, P. pardus, P. onca, P. uncia, P. tigris, and Neofelis nebulosa; (2) the recent common ancestry of Acinonyx jubatus, the African cheetah, and Puma concolor, the American puma; and (3) two golden cat species, Profelis temmincki and Profelis aurata, are not sister species, and the latter is strongly associated with Caracal caracal. These data add to the growing database of vertebrate mtDNA sequences and, given the relatively recent divergence among the felids represented here (1-10 Myr), allow 12S and cytochrome b sequence evolution to be addressed over a time scale different from those addressed in most work on vertebrate mtDNA.   相似文献
