Valinomycin is a potent inhibitor of taurine-stimulated ATP-dependent calcium ion uptake in rat retinal membrane preparations but had no effect on ATP-dependent calcium ion uptake in the absence of taurine and no effect on ATP-independent calcium ion uptake. The presence of potassium ions in the buffer systems were required for valinomycin to be inhibitory. On the contrary, monensin stimulated calcium ion uptake in the ATP-dependent system but had no effect on ATP-independent calcium ion uptake. The crude retinal homogenate was also fractionated into various subcellular components. The fraction which contained photoreceptor cell synaptosomes (P1) had a higher, specific activity for taurinestimulated ATP-dependent calcium ion uptake than the crude homogenate or either the fractions which contained synaptosomes derived from the plexiform layer (P2) or rod outer segments (ROS). No differences in calcium ion uptake were observed in the various subcellular fractions compared to the homogenate when assayed for ATP-dependent calcium ion uptake. Valinomucin inhibited both ATP-dependent and taurine-stimulated ATP-dependent calcium ion uptake in the P1, P2, and ROS fractions while monensin stimulated the ATP-dependent calcium ion uptake in the subcellular fractions.  相似文献

Ischemia was produced by occluding the circumflex branch of the left coronary artery in dogs. The artery remained patent in control animals. Tissue samples were obtained after 4 hours with or without occlusion. Right and left atria contained the highest taurine levels. In controls, an increasing outer to inner gradient in taurine content was observed in the posterolateral wall ventricle. No gradient was found in the right ventricular wall. For the first time, ischemia has been shown to reduce cardiac muscle taurine 47 percent in the left ventricle and 26 percent in right and left atria. Thus heart taurine 1) shows a non-uniform regional distribution in control and ischemic animals, and 2) is decreased after ischemic injury.  相似文献

Summary Taurine is known to increase ATP-dependent calcium ion (Ca2+) uptake in retinal membrane preparations and in isolated rod outer segments (ROS) under low calcium conditions (10M) (Pasantes-Morales and Ordóñez, 1982; Lombardini, 1991). In this report, ATP-dependent Ca2+ uptake in retinal membrane preparations was found to be inhibited by 5M cadmium (Cd2+), suggesting the involvement of cation channel activation. The activation of cGMP-gated cation channels, which are found in the ROS, is a crucial step in the phototransduction process. An inhibitor of cGMP-gated channels, LY83583, was found to inhibit taurine-stimulated ATP-dependent Ca2+ uptake but had no effect on ATP-dependent Ca2+ uptake in the absence of taurine, indicating that taurine may be increasing ATP-dependent Ca2+ uptake through a mechanism of action involving the opening of cGMP-gated channels. The activation of cGMP-gated channels with dibutyryl-cGMP and with phosphodiesterase inhibition using zaprinast caused an increase in ATP-dependent Ca2+ uptake in isolated ROS, but not in taurine-stimulated ATP-dependent Ca2+ uptake. LY83583 had the same effects in isolated ROS as in retinal membrane preparations. Another inhibitor of cGMP-gated channels, Rp-8-Br-PET-cGMPS, produced the same pattern of inhibition in isolated ROS as LY83583. Thus, there appears to be a causal link between taurine and the activation of the cGMP-gated channels in the ROS under conditions of low calcium concentration, a connection that suggests an important role for taurine in the visual signalling function of the retina.  相似文献

Summary Alveolar macrophages isolated by pulmonary lavage from partially taurine-depleted ratsdemonstrated increased (2–2.6 fold) chemiluminescence due to the extracellular reaction between exogenous zymosan and various reactive forms of oxygen compared to macrophages isolated from control animals. Partial taurine depletion was achieved by adding 3% ß-alanine to the drinking water of the rats for 5 weeks prior to harvesting the macrophages. Superoxide dismutase activity was not increased in the lung tissue of the taurine-depleted rats. These data suggest that taurine has antioxidant properties and that taurine depletion is potentially deleterious to alveolar macrophages and pulmonary tissue.  相似文献

Background

The animal efficacy rule addressing development of drugs for selected disease categories has pointed out the need to develop alternative large animal models. Based on this rule, the pathophysiology of the disease in the animal model must be well characterized and must reflect that in humans. So far, manifestations of the acute radiation syndrome (ARS) have been extensively studied only in two large animal models, the non-human primate (NHP) and the canine. We are evaluating the suitability of the minipig as an additional large animal model for development of radiation countermeasures. We have previously shown that the Gottingen minipig manifests hematopoietic ARS phases and symptoms similar to those observed in canines, NHPs, and humans.

Principal Findings

We establish here the LD50/30 dose (radiation dose at which 50% of the animals succumb within 30 days), and show that at this dose the time of nadir and the duration of cytopenia resemble those observed for NHP and canines, and mimic closely the kinetics of blood cell depletion and recovery in human patients with reversible hematopoietic damage (H3 category, METREPOL approach). No signs of GI damage in terms of diarrhea or shortening of villi were observed at doses up to 1.9 Gy. Platelet counts at days 10 and 14, number of days to reach critical platelet values, duration of thrombocytopenia, neutrophil stress response at 3 hours and count at 14 days, and CRP-to-platelet ratio were correlated with survival. The ratios between neutrophils, lymphocytes and platelets were significantly correlated with exposure to irradiation at different time intervals.

Significance

As a non-rodent animal model, the minipig offers a useful alternative to NHP and canines, with attractive features including ARS resembling human ARS, cost, and regulatory acceptability. Use of the minipig may allow accelerated development of radiation countermeasures.  相似文献

Young olive plants (Olea europaea L.) were grown either in hydroponic or soil culture in a glasshouse over two growing seasons. Plants were exposed to NaCl concentrations between 0 and 200 mM for 34–35 days followed by 30–34 days of relief from stress to determine the effect of salinity on gas exchange of two cultivars ('Frantoio' and 'Leccino') differing in salt-exclusion capacity. Salinity stress brought about a reduction in net CO2 assimilation and stomatal conductance in both cultivars, but the effect was more pronounced in the salt tolerant 'Frantoio' than in the salt-sensitive 'Leccino' cultivar. Therefore, gas exchange parameters may be misleading if used to evaluate the salt tolerance of olive genotypes. Recovery in gas exchange parameters during relief from stress was slower in the salt sensitive cultivar. In general, the decline in assimilation reflected the salt-induced reduction in stomatal conductance, but a marked effect on carboxylation efficiency and CO2 compensation point was measured in plants treated with 200 mM NaCl for four weeks. The cultivar 'Frantoio' showed a 50% reduction in assimilation and stomatal conductance at 146 and 78 mM leaf Na+ concentration (tissue water molar basis) respectively, whereas the corresponding 50% thresholds for the cultivar 'Leccino' were at 275 and 264 mM, respectively.  相似文献

Additional large animal models for the acute radiation syndrome (ARS) would facilitate countermeasure development. We demonstrate here that Gottingen minipigs develop hematopoietic ARS symptoms similar to those observed in canines, non-human primates (NHPs) and humans. Dosimetry for whole-body γ irradiation (0.6 Gy/min) was performed using electronic paramagnetic resonance (EPR) with alanine; National Institute of Standards and Technology (NIST)-calibrated alanine pellets and water-filled Plexiglas phantoms were used. After irradiations of 1.6-2.0 Gy, blood pancytopenia was observed for several weeks, accompanied by the characteristic ARS stages: prodromal symptoms, latent period, illness and recovery or morbidity. Morbidity occurred between days 14 and 27, with a preliminary LD(50/30) estimate between 1.7 and 1.9 Gy. The criterion of whether platelet counts were <200 × 10(3)/μl 7 days postirradiation predicted whether animals would survive in 18 out of 20 cases. The degree of granulocytosis 3 h postirradiation was inversely correlated with survival. Animals euthanized based on preset morbidity criteria displayed signs of multi-organ dysfunction, including widespread internal hemorrhage and alterations in organ function reflected in blood chemistry. Circulating C-reactive protein (CRP), a marker for inflammation, became elevated within hours after irradiation, subsided after several days, and increased again after 14 days. The results support further development of the Gottingen minipig as a model for ARS.  相似文献

Thrombopoietin (TPO) receptor agonists lacking sequence homology to TPO were designed by grafting a known peptide sequence into the hinge and/or kappa constant regions of a human anti-anthrax antibody. Some of these proteins were equipotent to TPO in stimulating cMpl-r activity in vitro and in increasing platelet levels in vivo. ALXN4100TPO (4100TPO), the best agonist in this series with a K(d) of 30 nM for cMpl-r, exhibited potent activity as a radiation countermeasure in CD2F1 mice exposed to lethal total-body radiation from a cobalt-60 γ-ray source. 4100TPO (2 mg/kg, s.c.) administered once either 24 h before or 6 h after TBI showed superior protection to five daily doses given before or after TBI. Prophylactic administration (69 to 94% survival) was superior to therapeutic schedules (60% survival). 4100TPO conferred a significant survival benefit (P < 0.01) when administered 4 days before or even 12 h after exposure and across a dose range of 0.1 to 8 mg/kg. The dose reduction factors (DRFs) with a single dose of 1 mg/kg 4100TPO 24 h before or 12 h after TBI were 1.32 and 1.11, respectively (P < 0.0001). Furthermore, 4100TPO increased bone marrow cellularity and megakaryocytic development and accelerated multi-lineage hematopoietic recovery in irradiated mice, demonstrating the potential of 4100TPO as both a protector and a mitigator in the event of a radiological incident.  相似文献
