Desulfitobacterium chlororespirans Co23 is capable of using 3-chloro-4-hydroxybenzoate as terminal electron acceptor for growth. Membrane preparations from cells grown fermentatively on pyruvate in the presence of 3-chloro-4-hydroxybenzoate dechlorinated this compound at a rate of 3.9 nmol min(sup-1) mg of protein(sup-1). Fivefold-greater dechlorination rates were measured with reduced methyl viologen as the artificial electron donor. Reduced benzyl viologen, NADH, NADPH, reduced flavin adenine dinucleotide, and reduced flavin mononucleotide could not substitute for reduced methyl viologen. The maximal initial rate of catalysis was achieved at pH 6.5 and 60(deg)C. The membrane-bound dechlorinating enzyme system was not oxygen sensitive and was stable at 57(deg)C for at least 2 h. Sulfite inhibited dechlorination in cell-free assays, whereas sulfate did not. Several chlorophenols were dehalogenated exclusively in the ortho position by cell extracts.  相似文献

2-Bromoethanesulfonate (BES) inhibited the reductive dechlorination of chloroethenes in several sediment-free enrichment cultures in the absence of methanogenic archaea. Archaeon-specific PCR primers confirmed the absence of methanogens in the enrichment cultures. BES should not be used to attribute dechlorination activities to methanogens.  相似文献

The decision of the embryonic gonad to differentiate as either a testis or an ovary is a critical step in vertebrate development. The molecular basis of this decision has been the focus of much study, particularly over the past decade. Here we contrast the knowledge of early gonadal development and the switch to testis differentiation with the lack of molecular understanding of ovarian development at early stages. We review current knowledge regarding mechanisms of ovarian morphogenesis and propose a model for the hierarchical control of development of the fetal ovary, incorporating the few genes already known to be important and several signals or factors that are hypothesised to exist in the early ovary.  相似文献

Summary As an approach for a better understanding of the mode of action of rotenone on mammalian cells we have studied the proliferation properties, metabolism and basic cell composition of Ehrlich ascites tumour cells cultured in vitro in the presence of 2,5 µM rotenone and after removal of the inhibitor.Experiments on asynchronous cells showed a rapid cessation of cell division accompanied by increased glycolytic rate, reduced oxygen consumption, moderate increase in DNA content and a fair increase in protein and RNA content of the cultures. DNA histograms obtained by flow-cytometry revealed an accumulation of cells in the G2 and M phase of the cell cycle. Electron micrographs taken after a 24 h treatment of cells illustrated the formation of giant mitochondria and fragmented nuclei.In order to elucidate the dual effect of rotenone — inhibition of mitochondrial energy metabolism and of mitotic processes — the influence on cells of rotenone at different stages of the cell cycle was tested using Ehrlich ascites tumour cells enriched in G1, S and G2 by centrifugal elutriation. DNA histograms and [3H]thymidine labelling index curves of cells from the different fractions cultured in the presence of 2,5 AM rotenone indicated that in addition to the observed accumulation in G2 and mitotic arrest of cells, the cell cycle progression is delayed in G1 phase. This may be explained by an effect of the inhibitor on the respiratory chain. S phase cells seemed to continue the cycle for several hours at a rate comparable to that of controls.Recultivation experiments on rotenone-treated asynchronous cells in inhibitor-free medium confirmed that some cells reinitiate DNA synthesis without preceeding cell division.Thus it must be concluded that cells at all stages of the cycle are affected by rotenone, but the impairment of cellular metabolism becomes manifest and lethal as soon as the acute block at mitosis is abolished and cells reenter the cycle.Abbreviations EAT cells Ehrlich ascites tumour cells - Hanks' solution Hanks' balanced salt solution - Hepes 4-(2-hydroxyethyl)-1-piperazineethane sulfonic acid  相似文献

Increasing ocean temperatures associated with ongoing climate change have resulted in regional reductions in the cover of live coral and increasing concerns that coral reefs will be overgrown by macroalgae. The likelihood of macroalgal overgrowth will, however, depend on the thermal sensitivities of the macroalgae themselves. We exposed recently settled propagules of the common canopy-forming macroalga Sargassum swartzii and adult thalli of three species of Sargassum (S. swatzii, S. cristaefolium, S. polycystum) to three experimental temperatures: ambient, + 2 °C, and + 3.5 °C, reflective of summer minimum, mean, and maximum temperatures for the region. Growth and survival of Sargassum swartzii propagules were assessed over 48 days, and the growth, physical toughness, elemental composition, and susceptibility to herbivory of adult thalli were assessed after short-term exposure (2-weeks) to experimental temperatures. Growth and survival of S. swartzii propagules were reduced by 43% and 84%, respectively, when cultured at the elevated (+ 3.5 °C) temperature compared to ambient temperature. Similarly, elevated temperature resulted in a 17–49% decline in the growth of adult Sargassum thalli relative to controls. Susceptability of S. swartzii and S. cristaefolium to herbivory (i.e. mass removed by herbivores) was 50% less for thalli cultured at elevated (+ 3.5 °C) compared to ambient temperature, but this pattern was not related to changes in the physical or chemical properties of the thalli as a result of elevated temperature. The negative effects of elevated temperatures on the growth and survival of both Sargassum propagules and adult thalli will likely restrict the capacity of Sargassum, and potentially other macroalgae, to establish in new areas, and may also threaten the persistence of existing macroalgal meadows under future ocean temperatures. The thermal sensitivities of tropical Sargassum, together with those of corals, suggest ongoing ocean warming may lead to novel reef ecosystems that are low in both coral cover and macroalgal cover.

Leptospirosis is the most widespread zoonosis in the world and significant efforts have been made to determine and classify pathogenic Leptospira strains. This zoonosis is maintained in nature through chronic renal infections of carrier animals, with rodents and other small mammals serving as the most important reservoirs. Additionally, domestic animals, such as livestock and dogs, are significant sources of human infection. In this study, a multiple-locus variable-number tandem repeat analysis (MLVA) was applied to genotype 22 pathogenic Leptospira strains isolated from urban and periurban rodent populations from different regions of Argentina. Three MLVA profiles were identified in strains belonging to the species Leptospira interrogans (serovars Icterohaemorrhagiae and Canicola); one profile was observed in serovar Icterohaemorrhagiae and two MLVA profiles were observed in isolates of serovars Canicola and Portlandvere. All strains belonging to Leptospira borgpetersenii serovar Castellonis exhibited the same MLVA profile. Four different genotypes were isolated from urban populations of rodents, including both mice and rats and two different genotypes were isolated from periurban populations.  相似文献
