The removal of polycyclic aromatic hydrocarbons by membrane bioreactor (MBR) under aerobic conditions had been studied using naphthalene (NAP) and phenanthrene (PHE) as model compounds. Three MBRs with submerged ultra-filtration hollow fiber membranes were operated applying different operational conditions during 6.5 months. Complete NAP and PHE removal was obtained applying loads of 7 gNAP kgTSS?1 day?1 and 0.5 gPHE kgTSS?1 day?1, while the organic loading rate was adjusted to 0.26 kgCOD kgTSS?1 day?1, with the biomass concentration being 6000 mgTSS L?1, the hydraulic retention time (HRT) 8 h and the solids retention time (SRT) 30 days. Load increases, as well as HRT and SRT reductions, affected the NAP and PHE removals. Biodegradation was found to be the major NAP and PHE removal mechanism. There was no NAP accumulation in the biomass. Low PHE quantities remain sorbed in the biomass and the contribution of the sorption in the removal of this compound was estimated to be less than 0.01 %. The volatilization does not contribute to the PHE removal in MBRs, but the contribution of NAP volatilization can reach up to 0.6 % when HRT of 8 h is applied. 相似文献

In an attempt to relate karyotype evolution to clinical and hematological data serial chromosomal analyses were performed in 31 patients with chronic myeloid leukemia (CML), both in chronic and acute phases. Our results in Philadelphia chromosome (Ph1)-positive CML are in line with karyotype profiles described in the literature. In addition, we report on chromosomal findings in 4 cases of Ph1-negative disease, one presenting with an iso17q chromosome in the positive CML. The same chromosomal abnormality was observed in a small population of Ph1-negative cells present in one of two patients with mixed Ph1-positive/Ph1-negative CML. The first case of a female patient with the loss of a sex chromosome in Ph1-positive cells is reported. Two patients with unusually long and mild chronic phases despite the presence of trisomy 8 in their karyotypes are described. Our findings suggest that the order of appearance of additional chromosomal changes of CML is of prognostic significance for the progression and the clinical picture of the disease. 相似文献

Taking into account the rising trend of the incidence of cancers of various organs, effective therapies are urgently needed to control human malignancies. However, almost all chemotherapy drugs currently on the market cause serious side effects. Fortunately, several studies have shown that some non‐toxic biological macromolecules, including algal polysaccharides, possess anti‐cancer activities or can increase the efficacy of conventional chemotherapy drugs. Polysaccharides are characteristic secondary metabolites of many algae. The efficacy of polysaccharides on the normal and cancer cells is not well investigated, but our investigations proved a cell specific effect of a newly isolated extracellular polysaccharide from the red microalga Porphyridium sordidum. The investigated substance was composed of xylose:glucose and galactose:manose:rhamnose in a molar ratio of 1:0.52:0.44:0.31. Reversible electroporation has been exploited to increase the transport through the plasma membrane into the tested breast cancer tumor cells MCF‐7 and MDA‐MB231. Application of 75 µg/mL polysaccharide in combination with 200 V/cm electroporation induced 40% decrease in viability of MDA‐MB231 cells and changes in cell morphology while control cells (MCF10A) remained with normal morphology and kept vitality. 相似文献

Balbiani ring (BR) granules are premessenger ribonucleoprotein particles (RNPs) generated in giant chromosomal puffs, the BRs, in the larval salivary glands of the dipteran chironomus tentans. Monoclonal antibodies were raised against nuclear proteins collected on a single-stranded-DNA-agarose affinity column, and two of them were used to identify RNA-binding proteins in BR granules. First, in Western blots (immunoblots), one of the antibodies recognized a 36-kDa protein and the other recognized a 45-KDa protein. Second, both antibodies bound to the BRs in immunocytological experiments. It was shown in cross-linking experiments that the two proteins are associated with heterogeneous nuclear RNP (hnRNP) complexes extracted from C. tentans nuclei. By immunoelectron microscopy of isolated and partly unfolded BR RNPs, it was specifically demonstrated that the BR granules contain the two proteins and, in addition, that both proteins are distributed frequently along the RNP fiber of the particles. Thus, the 36- and 45-KDa proteins are likely to be abundant, RNA-binding proteins in the BR particles. To elucidate to what extent the two proteins are also present in other hnRNPs, we studied the binding of the antibodies to chromosomal puffs in general. It was observed that many puffs in addition to the BRs harbor the two proteins, but there are also puffs containing only one of the components, either the 36- or the 45-kDa protein. We conclude that the two proteins are not randomly bound to all hnRNPs but that each of them seems to be linked to a specific subset of the particles. 相似文献

BACKGROUND: Spectral karyotyping and multiple fluorophore fluorescence in situ hybridisation (M-FISH) facilitate identification of inter-chromosomal rearrangements, but are of low cytogenetic resolution in mapping translocation breakpoints. Reverse chromosome painting yields increased cytogenetic information but isolation of aberrant chromosomes is technically difficult. We have developed the technique of paint-assisted microdissection FISH (PAM-FISH), which enables microdissection of aberrant chromosomes to be carried out easily and rapidly using relatively simple apparatus. METHODS: A selected chromosome paint is hybridised to abnormal metaphases to label a chromosome of interest, which is then microdissected, amplified, labelled by polymerase chain reaction (PCR), and reverse painted onto extended normal metaphases. RESULTS: PAM-FISH was used to reassess structural chromosomal abnormalities identified by molecular cytogenetics in the rhabdomyosarcoma cell line RD. PAM-FISH improved the analysis of virtually all structural abnormalities, identifying six novel translocations and indicating that seven previously described rearrangements were in fact not present in RD. Accuracy of the breakpoint mapping obtained was confirmed by bacterial artificial chromosome-FISH. CONCLUSIONS: PAM-FISH is ideally suited to analysis of tumour metaphases as it is not affected by poor chromosome morphology. Reagents generated by PAM-FISH are also suitable for other investigations, such as mapping using sequence tagged-site PCR or genomic microarrays. PAM-FISH is technically straightforward and could readily be adopted in a routine cytogenetics laboratory for accurate high-throughput analysis of chromosome breakpoints. 相似文献

Studied is the effect of the non-ionic surfactant Tween 80 on the microbial transformation of 4-androstene-3,17-dione into
its 9α-hydroxy-derivative by resting Rhodococcus sp. cells. The surfactant was applied in the cultivation medium as an additional source of carbon, in the transformation
reaction medium as a mediator of the steroid substrate solubility or was used for permeabilization of the glucose grown Rhodococcus sp. cells. Special attention is paid to the fact that Tween 80 accelerates the 9α-steroid hydroxylation reaction carried
out by glucose-grown cells. When the surfactant was applied as a supplementary source of carbon, the rate of the steroid hydroxylation
reaction was significantly lower. In addition, the kinetics of the transformation process changed into a linear one thus indicating
a very slow, if any, product degradation. The fatty acid profile, cell surface hydrophobicity as well as cell ultrastructure
observed by scanning and transmission electron microscopy in the Tween 80- and glucose-grown Rhodococcus sp. cells are compared and related with their 9α-hydroxylating activity. 相似文献
