Cellular sterol content and sterol metabolism were studied in diploid human kidney cells in early passages in culture. Cholesterol, the main cellular sterol, was present at levels similar to those reported for other cultured mammalian kidneys. Cholesterol biosynthesis was characterized by a slow conversion of sterol precursors with accumulation of lanosterol and 27 carbon-atom sterols. In the absence of exogenous cholesterol, kidney cells grew slowly and intracellular cholesterol decreased; however, sterol formation from labelled acetate was stimulated. These results suggest that the cholesterol concentration in the culture medium influences the rate of sterol formation by the kidney cell. Furthermore, cholesterol appears to be essential to cultured human kidney and de novo synthesis by the cells in culture is not adequate to meet their requirements for growth. 相似文献

Summary Exposure of HTC cells to sodium butyrate caused inhibition of growth. The site of growth inhibition was studied by time-lapse
cinematography and [3H]thymidine incorporation studies. Evidence is presented that sodiunm butyrate affected the cell cycle at a specific point
immediately after mitosis. Inasmuch as it does not modify the interphase duration after its removal, butyrate may be used
for HTC synchronization.

A casein-phosvitin-Sepharose chromatography column allows separation of nuclear protein kinases from their chromosomal phosphoprotein substrates and from at least some protein kinase inhibitors in a single step. The role of c-myc oncogene expression in myogenic differentiation has been established by transfecting rat myoblasts of the L6 cell line with plasmid pMT-myc, in which the c-myc coding sequences were under the control of the metallothionein I promoter. We observed that the constitutive expression of the exogenous c-myc gene inhibits muscular differentiation. A diminution of the endogenous c-myc gene expression occurs within the first 24 h after the transfer of the cells to a differentiating medium. This early decrease of c-myc expression is required for cell differentiation to occur. We have also observed that exogenous myc gene expression has no effect on endogenous myc expression. 相似文献
