Effect of ring-constrained phenylpropyloxyethylamines on sigma receptors

A series of ring-constrained phenylpropyloxyethylamines, partial opioid structure analogs and derivatives of a previously studied sigma (σ) receptor ligand, was synthesized and evaluated at σ and opioid receptors for receptor selectivity. The results of this study identified several compounds with nanomolar affinity at both σ receptor subtypes. Compounds 6 and 9 had the highest selectivity for both σ receptor subtypes, compared to μ opioid receptors. In addition, compounds 6 and 9 significantly reduced the convulsive effects of cocaine in mice, which would be consistent with antagonism of σ receptors.     

Enzyme Activity of Lectins from the Nitrogen-Fixing Soil Bacterium Bacillus polymyxa

Lectins LI and LII, localized on the surface of the nitrogen-fixing soil bacterium Bacillus polymyxa 1460, were shown to possess proteolytic activity. A relationship was found between the proteolytic and hemagglutinating activities of the lectins. Blocking of hemagglutinating activity with specific carbohydrate haptens led to significant changes in the enzyme activity of both lectins. When lectin activity was blocked with glucuronic acid and fructose-1,6-diphosphate, the proteolytic activity of both LI and LII declined, whereas incubation with d-galactosamine and d-glucosamine promoted increases in the proteolytic activity of LII. This study proposes that the molecules of the B. polymyxa lectins may have two centers on their surfaces: one responsible for lectin activity and the other for proteolytic activity. The effect of the pyridyl nitrogen position in pyridylpiperazine sigma ligands

A series of pyridylpiperazines was synthesized and analyzed for sigma receptor binding affinity to determine the optimal pyridyl nitrogen position and chain length for the σ₁ and σ₂ receptor recognition. The (3-pyridyl)piperazines and (4-pyridyl)piperazines favor σ₁ receptors, while previously studied (2-pyridyl)piperazines favor σ₂ receptors.     

Identification of Streptomyces coelicolor M145 genomic region involved in biosynthesis of teichulosonic acid–cell wall glycopolymer

The cell wall of the model actinomycete Streptomyces coelicolor M145 has recently been shown to contain the novel glycopolymer teichulosonic acid. The major building block of this polymer is 2-keto-3-deoxy-D-glycero-D-galacto-nononic acid (Kdn), suggesting initial clues about the genetic control of biosynthesis of this cell wall component. Here, through genome mining and gene knockouts, we demonstrate that the sco4879–sco4882 genomic region of S. coelicolor M145 is necessary for biosynthesis of teichulosonic acid. Specifically, mutants carrying individual knockouts of sco4879, sco4880 and sco4881 genes do not produce Kdn-containing glycopolymer and instead accumulate the minor cell wall component poly(diglycosyl 1-phosphate). Our studies provide evidence that this region is at least partly responsible for biosynthesis of Kdn, whereas flanking genes might control the other steps of teichulosonic acid formation.     

Role of the Yes and Csk tyrosine kinases in the development of a pathological state in the human retina

Amplification and a cloning of fragments of genes of human retina tyrosine kinases, the nucleotide sequences of which feature a high homology to the gene families of the Yes and Csk tyrosine kinases, and a cloning of the complete coding sequence of the cDNA of the Csk tyrosine kinase gene of the human lymphocytes have been carried out. It has been established that this sequence contains 1,624 bp and encodes a protein that, with a 99% homology, corresponds to the human tyrosine kinase. A comparative analysis of the nucleotide sequences of the full-size cDNA of the Csk tyrosine kinase of the lymphocytes of healthy donors and of patients with an eye choroidal melanoma has shown that a risk of development of an eye choroidal melanoma can be estimated by the frequency of occurrence of a mutant allele in the 10th exon.

Postural stability during static strain before and after a submaximal aerobic bicycle test in athletes

Control of static balance was studied in wrestlers (n = 31) in the normal state and against the background of physical fatigue, i.e., before and after a bicycle PWC ₁₇₀ test, respectively. The static balance was evaluated using a Ritm force platform (Russia) during normal basic stance (BS) and static strain (half-squatting position (HS) with the eyes open). Before physical exercise, i.e., during BS, the differences in postural control of the center of pressure (CP) between athletes and control subjects (n = 40) were nonsignificant. In the HS position, both linear and angular sway velocities of CP (LSV and ASV, respectively) were significantly lower in wrestlers (p < 0.001), and these parameters were negatively correlated with the PWC ₁₇₀ index (r = −0.454 and r = −455, p < 0.001 for LSV and ASV, respectively), which suggests that up to 20% of the sway velocity variance during static strain is dependent on the working capacity of the subjects. After the PWC ₁₇₀ test, the sway velocities of CP increased in both groups during BS (p < 0.01) and HS (p < 0.001), which testifies to the important role of muscular fatigue in the decrease in postural stability in both groups. During BS, an increase in LSV and ASV was about the same in both groups; however, during HS, an increase in the sway velocities of CP after PWC ₁₇₀ was lower in wrestlers (the intergroup differences at p = 0.037 and p = 0.008 for LSV and ASV, respectively); this parameter was negatively correlated with the recovery of the heart rate (HR) after the bicycle test (r = −0.378, p < 0.001 and r = −0.265, p < 0.05 for LSV and ASV, respectively), which suggests that rapid recovery contributes to a certain extent (about 6.5–14.2%) to the postural stability during static strain of wrestlers against the background of their physical fatigue.
