Time-dependent expression of functional proteins in fetal ovaries is important to understand the developmental process of the ovary. This study was carried out to enhance our understanding of the developmental process of porcine fetal ovaries and to better address the differences in fetal ovary development of local and foreign pigs. The objective of the present study is to test the expression of key proteins that regulate the growth and development of fetal ovaries in Meishan and Yorkshire porcine breeds by using proteomics technology. Six Meishan and 6 Yorkshire pregnant gilts were used in this experiment. Fetal ovaries were obtained from Yorkshire and Meishan gilts on days 55 and 90 of the gestation period. Using 2D-DIGE (two dimensional-difference in gel electrophoresis) analysis, the results showed that there are about 1551 and 1400 proteins in gilt fetal ovaries on days 55 and 90, respectively of the gestation. Using MALDI TOF-TOF MS analysis, 27 differentially expressed proteins were identified in the fetal ovaries of the 2 breeds on day 55 of gestation, and a total of 18 proteins were identified on day 90 of gestation. These differentially expressed proteins were involved in the regulation of biological processes (cell death, stress response, cytoskeletal proteins) and molecular functions (enzyme regulator activity). We also found that alpha-1-antitrypsin, actin, vimentin, and PP2A proteins promote the formation of primordial follicles in the ovaries of Yorkshire pigs on day 55 of gestation while low expression heat shock proteins and high expression alpha-fetoproteins (AFP) may promote Meishan fetal ovarian follicular development on day 90 of gestation. These findings provide a deeper understanding of how reduced expression of heat shock proteins and increased expression of AFP can significantly reduce the risk of reproductive disease in obese Meishan sows. Our study also shows how these proteins can increase the ovulation rate and may be responsible for the low reproductive efficiency reported in other obese breeds. The ovarian developmental potential was found to be greater in Meishan pigs than in Yorkshire pigs.  相似文献

People on a diet to lose weight may be at risk of reproductive failure. To investigate the effects of nutrient restriction on reproductive function and the underlying mechanism, changes of reproductive traits, hormone secretions and gene expressions in hypothalamus–pituitary–gonadal axis were examined in postpubertal gilts at anestrus induced by nutrient restriction. Gilts having experienced two estrus cycles were fed a normal (CON, 2.86 kg/d) or nutrient restricted (NR, 1 kg/d) food regimens to expect anestrus. NR gilts experienced another three estrus cycles, but did not express estrus symptoms at the anticipated fourth estrus. Blood samples were collected at 5 days’ interval for consecutive three times for measurement of hormone concentrations at the 23th day of the fourth estrus cycle. Individual progesterone concentrations of NR gilts from three consecutive blood samples were below 1.0 ng/mL versus 2.0 ng/mL in CON gilts, which was considered anestrus. NR gilts had impaired development of reproductive tract characterized by absence of large follicles (diameter ≥ 6 mm), decreased number of corepus lutea and atrophy of uterus and ovary tissues. Circulating concentrations of IGF-I, kisspeptin, estradiol, progesterone and leptin were significantly lower in NR gilts than that in CON gilts. Nutrient restriction down-regulated gene expressions of kiss-1, G-protein coupled protein 54, gonadotropin-releasing hormone, estrogen receptor α, progesterone receptor, leptin receptor, follicle-stimulating hormone and luteinizing hormone and insulin-like growth factor I in hypothalamus–pituitary–gonadal axis of gilts. Collectively, nutrient restriction resulted in impairment of reproductive function and changes of hormone secretions and gene expressions in hypothalamus–pituitary–gonadal axis, which shed light on the underlying mechanism by which nutrient restriction influenced reproductive function.  相似文献

【目的】解析土壤微生物在植物根际的组配机制对于认识和维护农田生态系统的稳定性至关重要。【方法】通过Illumina高通量测序和生物信息学分析方法明确了我国主要种植烟草生态区烤烟根际土壤细菌群落与土壤理化性质的互作关系。【结果】烤烟根际细菌类群主要为放线菌纲(Actinobacteria)、α-变形菌纲(Alphaproteobacteria)、γ-变形菌纲(Gammaproteobacteria)和嗜热油菌纲(Thermoleophilia)。细菌群落组成按生态区聚类,且样本空间距离和细菌群落相似度显著负相关。共现性网络分析表明,烤烟根际细菌群落间协同作用大于拮抗作用,武陵秦巴生态区、黄淮平原生态区、南岭丘陵生态区和沂蒙丘陵生态区细菌群落高度模块化,小单胞菌属(Micromonospora)为南岭丘陵生态区和黄淮生态区细菌共现性网络的网络中心,Bryobacter和气单胞菌属(Arenimonas)为南岭丘陵生态区细菌网络的模块核心,其菌群特性而非相对丰度决定了其在稳定细菌网络中的重要作用。冗余分析结果证实pH、有效铁、交换性镁和有效锰能显著影响烤烟根际细菌群落结构。【结论】烤烟根际细...  相似文献

报道了兰科(Orchidaceae)高山铠兰[Corybas himalaicus (King & Pantl.) Schltr.]和山珊瑚(Galeola faberi Rolfe)在中国西藏自治区的分布新记录。高山铠兰形态上因唇瓣先端边缘齿蚀状,花瓣与侧萼片在基部贴生,长7~8 mm,距暗红色而区别于铠兰属其他种;山珊瑚形态上因萼片狭椭圆形,唇瓣上散生褶片状附属物,边缘不规则齿状而区别于山珊瑚属其他种。凭证标本存放于广西药用植物园标本馆(GXMG)。  相似文献

Dietary yeast culture supplementation can contribute to the performance and health of sows and piglets, but few studies have focused on the relationships between the effects of yeast culture and gut microbiota. This study investigated the effect of yeast culture (Saccharomyces cerevisiae) supplementation from late gestation to weaning on the reproductive performance of lactating sows and their faecal microbiota. One hundred and six purebred Landrace sows, of parities two to six were selected and randomly assigned to a control (CON) and yeast culture supplementation (YC) groups based on parity and back fat thickness. The YC sows were individually fed with yeast culture at a dose of 24 g/d from day 90 of gestation to parturition and 40 g/d during lactational period. Blood samples were collected from sows on d 110 of gestation and at weaning at day 21 of lactation for plasma hormone and immunoglobulin analysis. Colostrum and milk on day 20 of lactation were collected for composition analysis. Faecal samples from sows on day 110 of gestation and day 20 of lactation were collected for short-chain fatty acid and faecal microbial analysis. Results showed that the farrowing performance of YC sows did not differ significantly from the CON group (P > 0.05). The average daily feed intake by the YC group during the lactation period was significantly increased by 9.98% (P = 0.004), the weaning-to-oestrus interval was shortened by 0.96 d (P = 0.046) and average daily weight gain of piglets increased by 7.14% (P = 0.036) compared with the CON group. Yeast culture supplementation also significantly improved the average daily milk yield in the first week of lactation (P = 0.035), lactose content in colostrum (P = 0.046), protein (P = 0.033) and DM (P < 0.001) content of milk. In the YC group, concentrations of plasma ghrelin (P = 0.02) and IgG (P = 0.015) were increased compared with the CON group, while that of glucagon-like peptide-1 was decreased (P = 0.006) on d 110 of gestation. The 16S rRNA gene sequencing showed that faecal microbiota changed at taxonomic levels with yeast culture addition (P < 0.05). Dietary yeast culture supplementation from late gestation to lactation improved feed intake, immunity status, milk yield, milk quality and faecal microbiota of sows, resulting in the improved growth performance of piglets.  相似文献

为了研究地下水浅埋区污水灌溉对土壤环境及作物生长的影响,设置地下水埋深(2、3m和4m)和灌水水量(900m3·hm-2和1200m3·hm-2)2个因素,对冬小麦全生育期土壤氮素和冬小麦生长发育指标进行试验,试验在地中渗透仪中进行.结果显示:灌水后,不同处理土壤中硝态氮含量均显著增加,灌水水平越高,土壤中硝态氮含量增加越多;灌水后,灌水水平B1(900 m3·hm-2)不同潜水埋深地下水硝态氮分别增加34.67%、24.94%、20.88%;灌水水平B2(1200 m3·hm-2)不同潜水埋深地下水硝态氮分别增加58.42%、38.98%、27.21%.潜水埋深越浅,地下水中硝态氮的浓度增加越大,这也就表明由于淋溶和硝化作用产生的硝态氮造成浅层地下水污染的风险越大.污水灌溉对冬小麦的生长发育指标和产量有促进作用;相同灌水水平(B1 和B2),地下水埋深影响冬小麦生长发育指标的大小顺序为:2m>3m>4m.  相似文献

Background

Intrauterine growth-restricted (IUGR) neonates impair postnatal skeletal muscle growth. The aim of this study was to investigate whether high nutrient intake (HNI) during the suckling period could improve muscle growth and metabolic status of IUGR pigs.

Methods

Twelve pairs of IUGR and normal birth weight (NBW) pigs (7 days old) were randomly assigned to adequate nutrient intake and HNI formula milk groups. Psoas major (PM) muscle sample was obtained after 21 days of rearing.

Results

IUGR decreased cross-sectional areas (CSA) and myofiber numbers, activity of lactate dehydrogenase (LDH), and mRNA expression of insulin-like growth factor 1 (IGF-1), IGF-1 receptor (IGF-1R), mammalian target of rapamycin (mTOR), ribosomal protein s6 (RPS6), eukaryotic translation initiation factor 4E (eIF4E), protein expression of phosphorylated mTOR (P-mTOR), and phosphorylated protein kinase B (P-Akt) in the PM muscle of pigs. Irrespective of birth weight, HNI increased muscle weight and CSA, the concentration of RNA, and ratio of RNA to DNA, as well as ratio of LDH to β-hydroxy-acyl-CoA-dehydrogenase in the PM muscle of pigs. Furthermore, HNI increased percentages of MyHC IIb, mRNA expression of IGF-1, IGF-1R, Akt, mTOR, RPS6, and eIF4E, as well as protein expression of P-mTOR, P-Akt, P-RPS6, and P-eIF4E in the PM muscle of pigs.

Conclusion

The present findings suggest that high nutrient intake during the suckling period could improve skeletal muscle growth and maturity, which is associated with increasing the expression of protein deposition-related genes and accelerating the development of glycolytic-type myofiber in pigs.

马梁遗址地处泥河湾盆地东缘,发现于1983年,并于同年及2006、2008年分别进行小规模发掘,出土了少量石制品和动物化石。2016年,为建立该区域早-中更新世古人类演化的文化序列,河北省文物研究所对低于马梁遗址文化层约3m的马梁第10地点开展试掘,发掘面积18m2,出土石制品197件,动物化石1500余件。石制品原料以燧石、石英砂岩和白云岩为主;类型包括石核、石片、工具、砾石、岩块、断块和碎屑等。技术上,以硬锤锤击为主要打制技术,石核剥坯较为简单,未见修理台面,工具加工缺乏"精制品",表现出较为单一和简单化的石器工业面貌。  相似文献

The study aimed to determine the effects of reduction of dietary crude protein (CP) level with balanced essential amino acids (EAA) on intestinal bacteria and their metabolites of growing pigs. Forty pigs (initial BW 13.50 ± 0.50 kg, 45 ± 2 days of age) were randomly assigned to four dietary treatments containing CP levels at 20.00% (normal crude protein, NP); 17.16% (medium crude protein, MP); 15.30% (low crude protein, LP); and 13.90% (extremely low crude protein, ELP), respectively. Crystalline AAs were added to meet the EAA requirement of pigs. After 4-week feeding, eight pigs per treatment (n = 8) were randomly selected and slaughtered for sampling of ileal, cecal, and colonic digesta and mucosa. Pigs with moderately reduced CP level had increased bacterial diversity, with the Shannon diversity indices for the colon digesta in the LP group and mucosa in the MP and LP groups significantly (P < 0.05) higher than those in the NP and ELP groups. As the CP level reduces, the Bifidobacterium population were linearly decreased (P < 0.05) both in ileum, cecum, and colon, and the ELP group had the lowest Bifidobacterium population in the cecum and colon, with its value significantly lower than NP and MP groups (P < 0.05). However, the ELP group had the highest population of Escherichia coli in the colon, with its value significantly higher than the LP group (P < 0.05). For bacterial metabolites, as CP level decreased, total short-chain fatty acid (T-SCFA), acetate, and butyrate were linearly increased (linear, P < 0.05) in the ileum, while all SCFAs except formate in the cecum and T-SCFA and acetate in the colon, were linearly decreased (P < 0.05). Reducing CP level led to a linear decrease of microbial crude protein (MCP) in the ileum (P < 0.05) and ammonia in all intestine segments (P < 0.05). The spermidine in cecum and total amines, cadaverine, methylamine, and spermidine in colon were shown a quadratic change (P < 0.05) as dietary CP decreases, with the highest concentration in LP group. These findings suggest that moderate reduction of dietary CP level may benefit large intestinal bacterial community and its fermentation, which was negatively affected by extremely low CP diet.
