游离DNA (cell free DNA,cfDNA)作为一种新型分子标志物在疾病预测、肿瘤防治等方面的作用日益突出.但在实现其广泛的临床应用前仍存在诸多障碍,主要是cfDNA相关的标准较少,导致目前的研究报道缺乏可比性,检测结果缺乏稳定的可重复性,从而降低了临床应用的可靠性.本综述集中阐述了近年来cfDNA的相关研究进展,包括cfDNA的样本来源、样本保存、样本提取、定性定量检测和应用,并分析比较了cfDNA研究的各个环节采用的方法及其优缺点,对其中存在的问题进行了剖析和总结,指明了cfDNA相关标准制定的迫切性.  相似文献

利用回交导入系剖析水稻苗期和分蘖期耐盐性的遗传重叠   总被引:4,自引:0,他引:4  
以籼稻品种IR64为受体,来自伊朗的粳稻农家品种Binam为供体培育的99个BC2F8回交导入系为材料,定位了水稻在2叶1心期和分蘖期控制耐盐相关性状的数量性状基因座(QTL).共检测到影响苗期叶片盐害级别、幼苗存活天数和地上部钾、钠离子浓度的QTL 13个,影响分蘖期株高、分蘖数和地上部鲜重的QTL 22个.多数分蘖期QTL对盐胁迫表现出明显的表达差异,根据表达情况将这些QTL分成3组,第1组为在对照条件下表达的11个QTL;第2组为在对照和盐胁迫条件下共同表达的5个QTL,其中有3个(QPh5,QPh8,QTn9)在两种环境下的基因效应大小和方向一致,其表达受盐胁迫影响较小;第3组为受盐胁迫诱导表达的6个QTL.检测到13个影响胁迫与对照差值即性状稳定性的QTL,除QPh4,QTn2和QFw2a外,其余10个基因座增加性状稳定性或提高耐盐性的等位基因均来自Binam.上述受盐胁迫影响较小的3个QTL和影响性状稳定性的13个QTL对耐盐性有贡献,属耐盐QTL.比较苗期和分蘖期耐盐QTL的分布,发现多数(69%)影响苗期和分蘖期的耐盐QTL在遗传上相互独立,仅在第1,2,8和11染色体的4个相同或相邻区域定位到影响两个时期的耐盐QTL,表明苗期和分蘖期的耐盐性存在部分的遗传重叠.通过标记辅助选择将苗期和分蘖期的重要耐盐QTL进行累加,或针对苗期和分蘖期的重叠耐盐QTL进行选择,有可能培育出苗期和分蘖期均耐盐的水稻品种.  相似文献

微卫星(TATG)n基序在香菇菌种中的验证   总被引:10,自引:0,他引:10  
以(TATG)4重复序列为引物对香菇属的3个种13个菌株的微卫星区DNA进行PCR扩增,15%的琼脂糖凝胶电泳,获得了25个条带,并且在供试菌株上表现出多态性,可以实现遗传分类研究。为了验证微卫星分子标记实验准确性,又用RAPD技术对13个供试菌株进行了实验。7个引物在13个菌株上共获得了102条多态性条带。通过聚类分析,RAPD获得的分类结果与微卫星分子标记获得的结果一致。此外,为了证明微卫星分子标记获得的条带不是假阳性,在实验中回收了No.10菌株的PCR扩增产物,进行克隆测序。测序结果显示有(TATG)n基序存在,并且达到了微卫星基序重复数量的最低限度。通过本实验可知,香菇中是存在微卫星(TATG)n基序的, 且基序的多态性可以用于香菇的遗传分类研究。  相似文献

Cytochrome P450 enzymes (P450s) are versatile biocatalysts, which insert a molecular oxygen into inactivated C−H bonds under mild conditions. CYP105D7 from Streptomyces avermitilis has been reported as a bacterial substrate-promiscuous P450 which catalyzes the hydroxylation of 1-deoxypentalenic acid, diclofenac, naringenin, compactin and steroids. In this study, CYP105D7 catalyzes hydroxylation, epoxidation and dehydrogenation of capsaicin, a pharmaceutical agent, revealing its functional diversity. The kinetic parameters of the CYP105D7 oxidation of capsaicin were determined as Km=311.60±87.30 μM and kcat=2.01±0.33 min−1. In addition, we conducted molecular docking, mutagenesis and substrate binding analysis, indicating that Arg81 plays crucial role in the capsaicin binding and catalysis. To our best knowledge, this study presents the first report to illustrate that capsaicin can be catalyzed by prokaryotic P450s.  相似文献

We examined the effects of TRPV1 agonists olvanil and piperine on glutamatergic spontaneous excitatory transmission in the substantia gelatinosa (SG) neurons of adult rat spinal cord slices with the whole-cell patch-clamp technique. Bath-applied olvanil did not affect the frequency and amplitude of spontaneous excitatory postsynaptic current (sEPSC), and unchanged holding currents at −70 mV. On the other hand, superfusing piperine reversibly and concentration-dependently increased sEPSC frequency (half-maximal effective concentration: 52.3 μM) with a minimal increase in its amplitude. This sEPSC frequency increase was almost repetitive at an interval of more than 20 min. Piperine at a high concentration produced an inward current in some neurons. The facilitatory effect of piperine was blocked by TRPV1 antagonist capsazepine. It is concluded that piperine but not olvanil activates TRPV1 channels in the central terminals of primary-afferent neurons, resulting in an increase in the spontaneous release of l-glutamate onto SG neurons.  相似文献

In this study, a central venous catheter (CVC)—associated infection model was established in rats to investigate and evaluate the effect of biofilms on the virulence of the pathogens. Twenty-four adult SD rats were randomly divided into biofilm positive (BF+) and biofilm negative (BF?) groups to be challenged with strains of S.epidermidis. Serum levels of inflammatory cytokines were measured and the infection rate and counts of bacteria cells were studied. Compared to rats of BF? group, the serum level of TNF and IL-6 significantly increased in rats of BF+ group (P < 0.01) and the level of IL-10 and IFN-γ significantly decreased (P < 0.01), striking the balance of pro-inflammatory/anti-inflammatory cytokines. The infection rate and bacterial counts in tissues and blood of rats of BF + group were significantly higher than those of rats of BF? group (P < 0.05).Inflammatory cell infiltration in vital organs (heart, lung, liver and kidneys) was more significant in rats of BF+ group than that of rats of BF- group. CVC-associated infection model can be successfully reproduced in rats by injecting 5 × 106 CFU of S.epidermidis. Biofilm formation can significantly enhance the virulence of the bacteria, leading to uncontrolled infection. The serum level of inflammatory cytokines, infection rate and the extent of inflammatory cell infiltration are important markers for evaluating the virulence of biofilm.  相似文献

Cd对蒌蒿生理生化及叶片超微结构的影响   总被引:2,自引:0,他引:2  
采用Cd砂土污染试验,研究了Cd对蒌蒿生理生化特征及叶片超微结构的影响,结果表明在砂土Cd含量>180mg/kg,蒌蒿出现中毒症状,叶绿素含量、a/b值、POD含量均下降,MDA积累,膜质过氧化,叶绿体形状球形化、片层结构解体、双层膜边界模糊、嗜锇颗粒增多,细胞核形状凹陷,核质分布不均匀、染色质凝胶化。低于180mg/kg的砂土Cd浓度,蒌蒿的上述生理生化指标和叶片超微结构基本正常。蒌蒿可以耐受的Cd浓度不超过180mg/kg。  相似文献

采用实验室建立的数字PCR方法 (ddPCR)和荧光定量PCR (qPCR)方法研究HER2基因组DNA标准物质的互通性,评价HER2基因组DNA标准物质与临床样本的互通性,为临床实验室检测提供具有互通性的可溯源标准物质.将研制的5个水平标准物质随机穿插于29例临床样本间,使用ddPCR方法与qPCR方法同时进行检测.参考美国临床实验室标准化协会(CLSI)指南文件EP30和中华人民共和国卫生行业标准基质效应与互通性评估指南WS/T356-2011的推荐,采用Deming回归法评价标准物质的互通性.结果显示,5种标准物质与临床样本之间具有良好的互通性,可以用于临床实验室对HER2基因拷贝数变异检测方法的方法验证和质量控制.  相似文献
