Summary Remobilization of15N from vegetative tissue of mungbean (Vigna radiata (L.) Wilczek) into pods was measured during the reproductive phase of growth. Plant tissue was labelled with15N during vegetative development. Experiments were conducted in the field at two sites. At one site the soil provided cowpeas
with most of their N but at the other site N fixation provided most of the N. Remobilized N from vegetative tissue to pods
occurred soon after they began to develop. The quantity of the labelled N ultimately remobilized to the pods amounted to 50%
for one cultivar (Tx33) at the high soil N site and 70% at the low N site. For the other cultivar (Tx13) the values were 25%
and 30%, respectively. The two cultivars performed very differently with respect to partitioning of N into pods and the rate
of N fixation. Even though more N was accumulated in the shoots of the high N fixing cultivar (Tx13) less total N was contained

Many cytosolic and nuclear proteins are modified by monomeric O-linked N-acetyl-
-glucosamine (O-GlcNAc). The biological functions of this form of glycosylation are unclear but evidence suggests that it heightens regulation of protein function. To assess the biological function of O-GlcNAc addition, we examined the biological effects of galactosyltransferase (GalT) microinjected into the cytoplasm of Xenopus ovarian oocytes. GalT, which catalyzes β1-4-galactose addition to O-GlcNAc, should inhibit deglycosylation and lectin-like interactions requiring unmodified O-GlcNAc residues. Although GalT injection into diplotene-arrested oocytes has no detectable effects on cell viability, it is toxic to oocytes entering meiosis. Cell-cycle-specific toxicity is recapitulated in vitro as GalT inhibits formation of nuclei and microtubule asters from cell-free extracts of ovulated frog eggs. These observations suggest that regulation of O-GlcNAc is important for cell cycle progression and may be important in diseases in which O-GlcNAc metabolism is abnormal. The methods described here outline a viable experimental scheme for ascribing a biological function to this form of glycosylation. 相似文献

Summary mRNA from estrogen-stimulated rat myometrium, a tissue known to upregulate cell-cell channels in response to this hormone, was microinjected intoXenopus laevis oocytes. The oocytes had been freed from covering layers of follicle cells and vitelline to allow direct cell membrane interactions when paired. About 4 hours after the mRNA injection, paired oocytes become electrically coupled. This coupling was due to the presence of typical cell-cell channels characterized by size-limited intercellular tracer flux, the presence of gap junctions at the oocyte-oocyte interface, and the reversible uncoupling that occurred in the presence of carbon dioxide. The induction of new cell-cell channels in the oocyte membrane was observed against a zero background or a low level of endogenous coupling, depending on the maturation stage of the oocytes. The time course of development of cell-cell coupling after the microinjection of mRNA was determined. The mRNA capable of inducing cell-cell coupling was confined to an intermediate size class when fractionated on a sucrose gradient. 相似文献

Synopsis Distribution of leptocephali ofConger in the Western North Atlantic Ocean was studied using specimens from our collections, specimens from other collections, and various existing collection records. The presence of leptocephali ofConger oceanicus andConger triporiceps < 30 mm long over deep water in the southwestern Sargasso Sea in autumn and winter implies a protracted spawning period there. The subtropical convergence zone, meandering east-west across the Sargasso Sea, is probably the northern limit of spawning of both species. Spawning may also occur close to the Bahamas and Antilles.C. triporiceps may spawn also in the Caribbean Sea judging by the capture of small leptocephali in the western Caribbean and of the more southerly continental distribution of its juveniles. The claim of Johannes Schmidt in 1931 that the EuropeanC. conger spawns across the North Atlantic into the western Sargasso Sea is probably incorrect, because leptocephali ofConger are rare in the eastern Sargasso Sea and becauseC. triporiceps, with myomere numbers overlapping those ofC. conger, was recently described in the western North Atlantic. With increasing size, leptocephali ofC. oceanicus and a portion ofC. triporiceps spread westward and northward in the Florida Current and Gulf Stream, but larger leptocephali especially ofC. triporiceps are found also in the Caribbean and Gulf of Mexico. Spawning ofC. oceanicus in the Sargasso Sea indicates that adults cross the Florida Current-Gulf Stream, and successful leptocephali cross the current in the opposite direction to colonize juvenile habitat on the continental shelf, a migratory pattern similar to that of the American eelAnguilla rostrata (Anguillidae). 相似文献

One-step gene disruption by cotransformation to isolate double auxotrophs in Candida albicans 总被引:11,自引:0,他引:11
Summary The Candida albicans LEU2 gene was disrupted by substituting lambda DNA for a small deletion within the LEU2 gene. Cotransformation with a selectable URA3 ARS vector was used to introduce a linear fragment containing the disruption into the genome of a C. albicans ura3 deletion mutant. Cotransformants containing the lambda DNA were identified by colony hybridization and the URA3 plasmid was subsequently cured. Leu2 disrupted heterozygotes were detected by Southern hybridization and one disruptant was subsequently treated with UV irradiation. Only one leu2 ura3 mutant (SGY-484) was isolated out of 11,000 mutagenized cells. SGY-484 was transformed to Leu+ with either the C. albicans or Saccharomyces cerevisiae LEU2 gene. Southern hybridization analysis revealed that the mutant is not homozygous for the disruption; the leu2 mutation reverts and is most likely a point mutation. Unexpectedly, an ade2 ura3 mutant was isolated from the same mutagenesis. 相似文献

In the European coastal dunes, marram grass (Ammophila arenaria) is planted in order to control sand erosion. In the years 1986 to 1991, workers on the Wadden islands in the Netherlands planting marram grass showed lesions of skin and mucous membranes, suggesting a toxic reaction. Fusarium culmorum dominated the mycoflora of those marram grass culms that were used for planting. This plant material had been cut and stored for more than one week in the open. The Fusarium toxin deoxynivalenol (DON) was detected in the suspect marram grass culms. Isolated F. culmorum strains were able to produce DON in vitro in liquid culture as well as in experimentally inoculated wheat heads. Pathogenicity tests, toxin test as well as RAPD analysis showed that the F. culmorum strains were not specialized for marram grass but may form part of the West-European F. culmorum population infecting cereals and grasses. Storage on old sand-dunes with plant debris may have led to the high occurrence of F. culmorum and contamination with DON. Marram grass culms should be obtained from young plantings on dunes on the seaward slopes and cut culms should not be stored. 相似文献

The ability to plan for future events is one of the defining features of human intelligence. Whether non-human animals can plan for specific future situations remains contentious: despite a sustained research effort over the last two decades, there is still no consensus on this question. Here, we show that New Caledonian crows can use tools to plan for specific future events. Crows learned a temporal sequence where they were (a) shown a baited apparatus, (b) 5 min later given a choice of five objects and (c) 10 min later given access to the apparatus. At test, these crows were presented with one of two tool–apparatus combinations. For each combination, the crows chose the right tool for the right future task, while ignoring previously useful tools and a low-value food item. This study establishes that planning for specific future tool use can evolve via convergent evolution, given that corvids and humans shared a common ancestor over 300 million years ago, and offers a route to mapping the planning capacities of animals. 相似文献
