In Saccharomyces cerevisiae the UPF1 protein is required for nonsense-mediated mRNA decay, the accelerated turnover of mRNAs containing a nonsense mutation. Several lines of evidence suggest that translation plays an important role in the mechanism of nonsense mRNA decay, including a previous report that nonsense mRNAs assemble in polyribosomes. In this study we show that UPF1 and ribosomal protein L1 co-localize in the cytoplasm and that UPF1 co-sediments with polyribosomes. To detect UPF1, three copies of the influenza hemagglutinin epitope were placed at the C-terminus. The tagged protein, UPF1-3EP, retains 86% (+/- 5%) of function. Using immunological detection, we found that UPF1-3EP is primarily cytoplasmic and was not detected either in the nucleus or in the mitochondrion. UPF1-3EP and L1 co-distributed with polyribosomes fractionated in a 7-47% sucrose gradient. The sucrose sedimentation profiles for UPF1-3EP and L1 exhibited similar changes using three different sets of conditions that altered the polyribosome profile. When polyribosomes were disaggregated, UPF1-3EP and L1 accumulated in fractions coincident with 80S ribosomal particles. These results suggest that UPF1-3EP associates with polyribosomes. L3 and S3 mRNAs, which code for ribosomal proteins of the 60S and 40S ribosomal subunits, respectively, were on average about 100-fold more abundant than UPF1 mRNA. Assuming that translation rates for L3, S3, and UPF1 mRNA are similar, this result suggests that there are far fewer UPF1 molecules than ribosomes per cell. Constraints imposed by the low UPF1 abundance on the functional relationships between UPF1, polyribosomes, and nonsense mRNA turnover are discussed. 相似文献

Abstract: An adult female rhesus monkey that had received 44.0 Gy of cobalt 60 radiation to 8 cm of the cervical and upper thoracic spinal cord approximately 2.8 years postirradiation developed a sudden onset of self-mutilation and loss of function of the right arm followed progressively by loss of function of the left arm and terminally bilateral paresis of the legs. Histopathologic examination of the cervical spinal cord revealed a glioblastoma multiforme that extended from the cervical medullary junction to the sixth cervical vertebrae. Replication slippage may cause parallel evolution in the secondary structures of mitochondrial transfer RNAs 总被引:9,自引:4,他引:5
Presence of the dihydrouridine (D) stem in the mitochondrial cysteine tRNA
is unusually variable among lepidosaurian reptiles. Phylogenetic and
comparative analyses of cysteine tRNA gene sequences identify eight
parallel losses of the D-stem, resulting in D-arm replacement loops.
Sampling within the monophyletic Acrodonta provides no evidence for
reversal. Slipped-strand mispairing of noncontiguous repeated sequences
during replication or direct replication slippage can explain repeats
observed within cysteine tRNAs that contain a D-arm replacement loop. These
two mechanisms involving replication slippage can account for the loss of
the cysteine tRNA D-stem in several lepidosaurian lineages, and may
represent general mechanisms by which the secondary structures of
mitochondrial tRNAs are altered.

Anthropogenic phosphorus (P) inputs to the Florida Everglades have produced dramatic changes in the wetland vegetation of
this otherwise oligotrophic system. While the proliferation of undesirable plant species in response to enrichment has been
well documented, nutrient-related changes in the physiological and morphological attributes of existing vegetation, prior
to any shifts in species composition or changes in the spatial extent of certain taxa, have yet to be adequately characterized.
In this experiment, three sawgrass-dominated areas were enriched with P for 3 years at rates of 0.4 g P/m2/year (HP), 0.1 g P/m2/year (LP), or 0 g P/m2/year (controls) to assess potential impacts of P-enriched discharges from stormwater treatment areas into the Everglades.
Elevated concentrations of TP in rhizomes and leaves and reduced ratios of leaf N:P were detected in HP plants within ~1 year
at most sites. Live leaf densities, plant heights, and plant densities of the HP groups were generally higher than LP and
control groups after 2 years, a pattern that was evident even after major fire events. Total aboveground biomass was significantly elevated in both HP and LP treatments at two of the three sites after 3 years.
No change in species composition was detected during the study. Planned hydrologic restoration measures will increase P loads
into parts of the Everglades that have not previously experienced anthropogenic P enrichment. Monitoring native vegetation
such as sawgrass can be a sensitive and relatively robust means of detecting unintended P enrichment in these areas prior
to shifts in vegetation community composition or changes in area cover of key species. 相似文献
