AIMS: To compare galactose-negative strains of Streptococcus thermophilus and Lactobacillus delbrueckii subspecies bulgaricus isolated from fermented milk products and known to produce exopolysaccharides (EPSs). METHODS AND RESULTS: The structures of the EPSs were determined using nuclear magnetic resonance (NMR) and their genetic relationships determined using restriction endonuclease analysis (REA) and random amplification of polymorphic DNA (RAPD). Similar groupings were apparent by REA and RAPD, and each group produced an EPS with a particular subunit structure. CONCLUSION: Although none of the strains assimilated galactose, all inserted a high proportion of galactose into their EPS when grown in skimmed milk, and fell into three distinct groups. Significance and Impact of the Study: This information should help in an understanding of genetic exchanges in lactic acid bacteria.  相似文献

Previous studies demonstrate that gonadotroph responsiveness to GnRH, GnRH binding, and the apparent number of GnRH receptors are all increased by 17 beta-estradiol (E) or inhibin (IN) in ovine pituitary cultures. Progesterone (P) attenuates these effects. To explore differences between the effects of IN and E on GnRH binding, a detailed time-course study was performed. The results indicate that after 48 h, IN had a greater effect on binding of a GnRH agonist (5-fold increase) than E (3-fold increase), but was slower to act initially. A combined treatment of IN and E gave a partially additive effect at 48 h (6.5-fold increase). The mechanism of receptor regulation in this system is not known, but could involve synthesis, recycling, or modification of GnRH receptors. To investigate the contribution of altered receptor biosynthesis to the regulation of receptor levels, a functional Xenopus oocyte-based assay for GnRH receptor mRNA activity was employed. After 48 h of treatment, IN or E each led to a 7- to 8-fold increase in GnRH receptor mRNA activity. Treatment with both hormones led to a 19-fold increase. The increase in mRNA activity induced by either hormone was greatly attenuated by P. Modulation of GnRH receptor mRNA levels suggests that IN, E, and P regulate responsiveness to GnRH in the ovine pituitary at least in part by altering de novo synthesis of GnRH receptors. The differing time courses of action, as assayed by GnRH binding, and the additivity of effects at the mRNA level suggest that IN and E alter mRNA levels via different mechanisms.  相似文献

The marine diatom Cyclotella cryptica was grown over a period of 13 months in a 48-m(2) shallow outdoor flume. The use of foil arrays at intervals of 1.2 m to effect systematic vertical mixing in the flume was found to significantly enhance microalgal production (p = 0.006). Average photosynthetic efficiencies (based on visible irradiance) with and without the foil arrays in place were 9.6 +/- 0.8 and 7.5 +/- 0.5% (+/-95% confidence intervals), respectively. A cost-benefit analysis indicated that the foil arrays were cost-effective if the value of the algae exceeded about $2.28 kg(1) of ash-free dry weight (AFDW). Parallel experiments performed in four 9.2-m(2) flumes showed that production was maximized when the cells were grown on a 2-day batch cycle between harvests rather than on a 1- or 3-day batch cycle. The optimum initial concentration (immediately after harvesting) of the algae was negatively correlated with the time interval between harvests and ranged from approximately 39 g AFDW/m(3) on a 3-day cycle to 213 g AFDW/m(3) on a 1-day cycle. The increase in production resulting from growth on a 2-day rather than a 1-day batch cycle was about 19% and was statistically significant at p = 0.0003. Growth of C. cryptica over a total period of 122 days during the 13-month study in the 48-m(2) flume under near-optimal conditions (2-day batch cycle, initial concentration 155 g AFDW/m(3)) resulted in an average production rate (+/-95% confidence interval) of 29.7 +/- 2.7 g AFDW/m(2) d.  相似文献

Steady-state and time-resolved fluorescence properties of the single tyrosyl residue in oxytocin and two oxytocin derivatives at pH 3 are presented. The decay kinetics of the tyrosyl residue are complex for each compound. By use of a linked-function analysis, the fluorescence kinetics can be explained by a ground-state rotamer model. The linked function assumes that the preexponential weighting factors (amplitudes) of the fluorescence decay constants have the same relative relationship as the 1H NMR determined phenol side-chain rotamer populations. According to this model, the static quenching of the oxytocin fluorescence can be attributed to an interaction between one specific rotamer population of the tyrosine ring and the internal disulfide bridge.  相似文献

A method is presented for determining both the average turnover rate and the standard deviation of the average turnover rate of the adenine nucleotide (AN) pool within a population of microorganisms. The method requires the calculation of the initial slope and curvature of a plot of AN specific activity versus time following the introduction of [3H]adenine. An analysis of noise-corrupted data indicated that the method is capable of detecting a lack of uniformity in the turnover rate when the coefficient of variation of the turnover rate exceeds 39%. An analysis of field data revealed a significant lack of uniformity in the turnover rates of microbial communities in a marine sediment sample and freshwater pond but no significant nonuniformity in the turnover rates of microbial communities in a seawater sample and in a second freshwater pond. Although the method has been applied only to the analysis of AN turnover rates, it is applicable to any intracellular pool for which a suitable radioactive precursor exists.  相似文献

Phenol sulfotransferase (PST) catalyzes the sulfate conjugation of catecholamines and phenol and catechol drugs. The human blood platelet contains a thermolabile (TL) form of PST that catalyzes the sulfate conjugation of dopamine and other monoamines and a thermostable (TS) form that catalyzes the sulfate conjugation of micromolar concentrations of phenol and p-nitrophenol. Experiments were performed to determine whether the brain contains forms of PST analogous to the TL and TS forms found in the human platelet, and to determine whether there are regional variations in human brain PST activity. We found that the human brain contains at least two forms of PST, forms that are similar to the platelet TS and TL forms of the enzyme with respect to substrate specificity, apparent Km constants, thermal stability, and sensitivity to inhibitors. Optimal conditions were determined for the measurement of these two activities in brain homogenates. The stability of PST activities in the human brain after death was determined in five samples of cerebral cortex that were obtained during clinically indicated neurosurgical procedures. An average of 76 +/- 8% and 80 +/- 9% (mean +/- SEM) of the basal TL and TS PST activities, respectively, remained in these five samples of cerebral cortex after 8 h of storage under simulated post-mortem conditions. Six human brains were then obtained less that 8 h after death from patients who had no neurological disease prior to death. The mean activities of the TL and TS forms of PST were measured in 17 different regions of the six brains. If the pituitary was excluded from consideration, TL and TS PST activities both varied approximately fivefold among these regions, and both activities were highest in cerebral cortex. However, the average TS activity in the anterior pituitary, a tissue of non-neural origin embryologically, was 6.5-fold greater than the highest average TS PST activity found in cerebral cortex.  相似文献

Arrays of foils similar in design to airplane wings have been placed in an algal culture flume to create systematic mixing. Vortices are produced in the culture due to the pressure differential created as water flows over and under the foils. In a flume having a flow rate of 30 cm/s, the foil arrays produced vortices with rotation rates of ca. 0.5-1.0 Hz. This rotation rate is satisfactory to take advantage of the flashing light effect if the culture is sufficiently dense. Solar energy conversion efficiencies in an experimental culture of P. tricornutum increased 2.2-2.4 fold with the foil arrays in place versus controls with no foil arrays and solar energy conversion efficiencies averaged 3.7% over a three-month period. Five-day running means of solar energy conversion efficiencies reached as high as 10% during the three-month period. The use of foil arrays appears to be an effective and inexpensive way to utilize the flashing light effect in a dense algal culture system.  相似文献

Two isolates of the marine pennate diatom Phaeodactylum tricornutum Bohlin were grown in semi-continuous, nutrient-sufficient culture at varying irradiances on a 12-h light, 12-h dark illumination cycle. The reponse of the isolates to varying degrees of light limitation differed with respect to all of the compositional parameters measured, including growth rates, elemental composition, chlorophyll content, and the partitioning of cellular carbon into four biochemical classes: proteins, lipids, polysaccharides, and low-molecular weight intermediates. The isolates also differed with respect to the relative contributions of light-period and dark-period uptake to the total uptake of ammonium and phosphate ions, although in all cases uptake took place at a reduced rate in the dark. They did not differ with respect to the diel periodicity of cell division, chlorophyll synthesis, and biochemical synthesis. Slightly more cell division took place during the dark period than during the light period. The specific rate of chlorophyll synthesis in the light period, when expressed as a function of irradiance, saturated rapidly; the rate was nearly constant for all irradiances > 100 βE · m?2 · s?1. Chlorophyll synthesis in the dark was positively correlated with irradiance over the entire range of irradiances, except where photoinhibition was involved. Protein was synthesized in both the light and dark periods, but at a reduced rate in the dark. Polysaccharides were synthesized during the light period and consumed during the dark period. Lipids and low molecular weight intermediates were synthesized during the light period, but showed little net change during the dark period.  相似文献

I. Summary The present paper is based on the examination of lower jaws from some 3000 individuals of Hippopotamus amphibius L. which were collected in the course of cropping operations in the Queen Elizabeth Park, Uganda between 1961 and 1966. It extends an earlier study by W. M. Longhurst, who described 20 relative age groups based on tooth replacement and wear. The age groups, slightly modified, are described and illustrated here and should serve as a useful field guide. Mean chronological ages from 0–43 years have been allocated to the groups. Checks which confirm the validity of the ages allotted are presented and discussed. These include correlation with the mandibular age group of six known-age animals; findings on the age-related incidence of rinderpest-neutralising antibodies; the orderly progress of fusion of the mandibular symphysis and rate of loss of the first premolar. Growth layers present in the teeth are discussed. The growth of the eye lens is also described. After an initial phase of rapid growth the lens continues to grow throughout life and follows a rectilinear pattern from an estimated age of eight years onwards. This is expected from the findings of similar studies on other species for which known-age specimens are available. Variability of lens dry weight at age is relatively small and indicates that the estimated ages are reasonably precise. Growth of the mandible is analysed and is not inconsistent with the ages allotted. A marked sex difference in mandible weight at age allows the sex of found jaws to be determined at ages above eight years. The growth of the teeth is described. Both canines and incisors show well-marked sex differences in growth rate and size. The post-canine teeth do not show sex differences. Cycles of growth, wear and resorption of these teeth are discussed and it is concluded that mechanical senescence of the teeth is a major factor in mortality at ages above 30 years. Growth in body length is briefly discussed and provides further confirmation of the validity and precision of the age criteria. Growth equations are presented. Finally a survivorship curve derived from the ageing of 207 jaws assumed to represent natural deaths is presented. Population models are constructed from the survivorship curves by calculation of estimated natality rates (obtained by applying data on age at first breeding and annual pregnancy rate to the survivorship data) and life tables are constructed. The shape of the survivorship curve and the percentage recruitment agree with expectation and provide further evidence of the consistency of the age criteria.  相似文献
