Experiments were conducted on cats by conditioned food-procuring method. Behavioural, vegetative reactions and a set of electrophysiological characteristics were recorded. It was found that regulatory influence of an extraneous stimulus and of artificial excitation of the brain was expressed in an increase of reactivity to conditioned and unconditioned sensory stimuli and in disinhibition of effector responses. This influence was realized not only during the development of EEG activation reaction but could be preserved over a long period in conditions of the deactivated brain state. Regulatory influence of interceptive factors is characterized by a decrease of reactivity and by inhibition of effector responses. As in the case of external effects, the realization of this influence begins with a period of brain activation and is preserved for a long time in conditions of deactivated state. 相似文献

Organelle genomics has become an increasingly important research field, with applications in molecular modeling, phylogeny, taxonomy, population genetics and biodiversity. Typically, research projects involve the determination and comparative analysis of complete mitochondrial and plastid genome sequences, either from closely related species or from a taxonomically broad range of organisms. Here, we describe two alternative organelle genome sequencing protocols. The "random genome sequencing" protocol is suited for the large majority of organelle genomes irrespective of their size. It involves DNA fragmentation by shearing (nebulization) and blunt-end cloning of the resulting fragments into pUC or BlueScript-type vectors. This protocol excels in randomness of clone libraries as well as in time and cost-effectiveness. The "long-PCR-based genome sequencing" protocol is specifically adapted for DNAs of low purity and quantity, and is particularly effective for small organelle genomes. Library construction by either protocol can be completed within 1 week. 相似文献
Recent field surveys performed in the newly-discovered Taurida Cave (Crimea, Russia, Late Villafranchian, 1.8–1.5 Ma) enabled us to unearth dentognathic remains of the dirk-toothed cat Megantereon. Here we describe in detail the recovered remains further comparing it anatomically and biometrically with coeval Eurasian and African sites. The performed analysis suggests the inclusion of the Taurida remains in the hypodigm of the Villafranchain European species Megantereon adroveri, a medium-sized saber-toothed cat poorly known in Europe as a consequence of the scarcity and fragmentary nature of the known remains. Our results and the putative transitional characters displayed by the Taurida specimen support the idea put forward by previous authors of a continuous European lineage of dirk-toothed cats.
Sponges (phylum Porifera) are early-branching animals, whose outwardly simple body plan is underlain by a complex genetic repertoire. The transition from a mobile larva to an attached filter-feeding organism occurs by metamorphosis, a process accompanied by a radical change of the body plan and cell transdifferentiation. The continuity between larval cells and adult tissues is still obscure. In a previous study, we have produced polyclonal antibodies against the major protein of the flagellated cells covering the larva of the sponge Halisarca dujardini, used them to trace the fate of these cells and shown that the larval flagellated cells transdifferentiate into the choanocytes. In the present work, we identified the sequence of this novel protein, which we named ilborin. A search in the open databases showed that multiple orthologues of the newly identified protein are present in sponges, cnidarians, flatworms, ctenophores and echinoderms, but none of them has been described yet. Ilborin has two conserved domains: triosephosphate isomerase-barrel, which has enzymatic activity against macroergic compounds, and canonical EF-hand, which binds calcium. mRNA of ilborin is expressed in the larval flagellated cells. We suggest that the new protein is involved in the calcium-mediated regulation of energy metabolism, whose activation precedes metamorphosis. 相似文献
A new acylamidase was isolated from Rhodococcus erythropolis TA37 and characterized. N-Substituted acrylamides (isopropyl acrylamide, N,N-dimethyl-aminopropyl acrylamide, and methylene-bis-acrylamide), acid para-nitroanilides (4′-nitroacetanilide, Gly-pNA, Ala-pNA, Leu-pNA), and N-acetyl derivatives of glycine, alanine, and leucine
are good substrates for this enzyme. Aliphatic amides (acetamide, acrylamide, isobutyramide, n-butyramide, and valeramide) are also used as substrates but with less efficiency. The enzyme subunit mass by SDS-PAGE is
55 kDa. Maximal activity is exhibited at pH 7–8 and 55°C. The enzyme is stable for 15 h at 22°C and for 0.5 h at 45°C. The
Michaelis constant (Km) is 0.25 mM with Gly-pNA and 0.55 mM with Ala-pNA. The acylamidase activity is suppressed by inhibitors of serine proteases
(phenylmethylsulfonyl fluoride and diisopropyl fluorophosphate) but is not suppressed by inhibitors of aliphatic amidases
(acetaldehyde and nitrophenyl disulfides). The N-terminal amino acid sequence of the acylamidase is highly homologous to those
of two putative amidases detected from sequenced R. erythropolis genomes. It is suggested that the acylamidase together with the detected homologs forms a new class within the amidase signature
family. 相似文献
The study of a natural endocranial cast of Paroxyaena has shown that its vision was an important system of afferentation, but did not prevail over others, unlike that of Neohyaenodon. The olfaction was much better developed than in Neohyaenodon. The large gyrus prorealis suggests the presence of a long mobile nose. The well-developed sensomotoric cortex is evidence that the forepaw of Paroxyaena showed a high mobility. 相似文献
