OBJECTIVE--To investigate the relation between socioeconomic deprivation and pathological prognostic factors in women with breast cancer as a possible explanation for socioeconomic differences in survival. DESIGN--Retrospective analysis of data from cancer registry and from pathology and biochemistry records. SETTING--Catchment areas of two large teaching hospitals in Glasgow. SUBJECTS--1361 women aged under 75 who had breast cancer diagnosed between 1980 and 1987. MAIN OUTCOME MEASURES--Tumour size, axillary lymph node status, histological grade, and oestrogen receptor concentration in relation to deprivation category of area of residence. RESULTS--There was no significant relation between socioeconomic deprivation and four pathological prognostic factors: 93 (32%) women in the most affluent group presented with tumours less than 20 mm in size compared with 91 (31%) women in the most deprived group; 152 (48%) of the most affluent group presented with negative nodes compared with 129 (46%) of the most deprived group; 23 (22%) of the most affluent group presented with grade I tumours compared with 12 (17%) of the most deprived group; and 142 (51%) of the most affluent group had a low oestrogen receptor concentration at presentation compared with 148 (52%) of the most deprived group. None of these differences was statistically significant. CONCLUSIONS--Differences in survival from breast cancer by socioeconomic deprivation category could not be accounted for by differences in tumour stage or biology. Other possible explanations, such as differences in treatment or in host response, should be investigated. 相似文献

As an approach to evaluating the contribution of classes of endogenous viral sequences to leukemogenesis, a genomic library was prepared from the highly tumorigenic AKR SL12.3 cell line and screened for env-containing proviruses. An extensive battery of virus-derived probes and specific oligonucleotide probes were used to segregate 83 positive clones into related groups. The nonecotropic endogenous retroviruses were identified as members of the polytropic, modified polytropic, or xenotropic groups. At least three unique xenotropic proviruses were detected that differed from the published xenotropic sequence within a variable region of the 5' portion of env. Changes among the xenotropic proviruses included relative insertions and/or deletions that maintain an open reading frame and hence the potential to encode viable envelope gene products. Several recombinant viruses were also detected. Recombination was not random and primarily involved the formation of mink cell focus-inducing class I retroviruses via recombination between polytropic elements and ecotropic virus. One other recombinant was detected which contained ecotropic virus sequences in the 5' region encoding p15 of an otherwise xenotropic provirus. An interesting observation was the finding that certain clones contained more than one provirus within the average 20-kb cloned insert. This would not be expected if integration were totally random. The de novo recombinant proviruses identified here provide a series of potential candidates to be evaluated for their contribution to the tumorigencity of the SL12.3 cell line. 相似文献

The spoOB and spoIVF loci are contiguous on the chromosome of Bacillus subtilis, so that genes in these loci may be parts of a single polycistronic operon. Temperature-sensitive strains having mutations in these loci were isolated, and temperature-shift experiments were carried out to investigate expression of the genes. The temperature-sensitive periods of spoOB mutants extended from the beginning of sporulation until the end of the stage II. The temperature-sensitive periods of spoIVF strains were during stage IV of sporulation. Therefore, although the spoOB and spoIVF loci are contiguous on the chromosome it is unlikely that genes in them are parts of a single polycistronic operon. 相似文献

Trichomes of two oscillatoriacean blue-green algae execute screw-like gliding motion, but the two organisms differ from each other with respect to the screw sense of motion. Electron microscopy of serial longitudinal sections reveals extracellular microfibrils which lie roughly parallel to stream-lines at the surface of each organism. The author proposes that the microfibrils are oriented by shear in a zone just external to the outer unit membrane-like component of the cell wall. 相似文献

Summary Trichomes of Microcoleus vaginatus, a motile blue-green alga of the family Oscillatoriaceae, were studied by light and electron microscopy in an effort to determine the sites of trichome breakage during production of hormogonia.According to the evidence presented herein, transcellular breakage of trichomes is the only mechanism of hormogonium production in M. vaginatus. Tearing of the murein sacculus appears to be necessary and sufficient for transcellular breakage to ensue. As Fuhs and earlier investigators have correctly claimed, this process always involves the death of the cell whose wall is torn.When trichomes of M. vaginatus break across cells to produce hormogonia, the murein sacculus usually tears along a circumferential set of junctional pores. This particular mechanism of trichome breakage is not universal among members of the family Oscillatoriaceae.This report is based on a thesis submitted in partial fulfillment of the requirements for a Ph. The identification of a core region for OVA 323-339, which is critical in determining binding to IAd, has enabled us to generate a series of analog peptides in which this core region was extended at both the N and C termini with different amino acid residues. When assessed for binding capacity, several peptides were shown to have increased affinity for IAd compared with the parent sequence, and in addition, some peptides had acquired binding specificities for class II MHC haplotypes not present for OVA 323-339. These peptides were next examined for their ability to inhibit T cell responses in vitro and in vivo. The correlation between binding and the ability to inhibit T cell activation in vitro was good. However, when assessed in vivo, it was clear that high Ia binding was not sufficient in itself to define the inhibitory capacity of a given peptide. That this discrepancy was due to differences in degradation of the core-extended peptides was suggested by 1) results from an inhibition of Ag presentation assay, in which the pulse period with Ag and inhibitor was extended to 20 h; and 2) direct analysis of peptide stability by using reverse phase HPLC. Finally, by protecting the peptide from degradation with N- and C-terminal substitutions of D-amino acids, the inhibitory capacity of an unstable core-extended peptide in vitro could be greatly enhanced. These data indicate that the core extension approach may be one method by which antagonists for MHC class II molecules may be generated. 相似文献

A method for measuring xylem hydraulic conductance and embolism in entire root and shoot systems 总被引:12,自引:6,他引:6
Current methods for determining the influence of xylem cavitationon hydraulic conductance are limited to unbranched stem or rootsegments with hydraulic conductances above c. 2 mmol s1MPa1. Lower conductances and/or highly branched systemsare encountered in seedlings, arid-land shrubs, herbs, and distalportions of shoot and root systems of trees. In order to quantifythe hydraulic impact of cavitation in such systems, existingtechniques have been modified. Branched shoot or root systemswere prepared for measurement by removal of leaves, or roottips, respectively. The shoot or root system was enclosed ina vacuum chamber with the proximal end protruding and suppliedwith perfusing solution. Flow through the xylem was inducedby chamber vacuum. Hydraulic conductance was determined fromthe slope of the flow rate versus pressure relationship. Xylemembolism was quantified from the increase in hydraulic conductancefollowing high pressure (100 kPa) perfusion of solution throughthe plant. Examples are provided of the application of the methodto cavitation studies in the cold desert shrub Artemisia tridentata. We compared above-ground allocation patterns in mature shrubs of Banksia hookeriana from three 13-year-old populations, growing on nutrient-impoverished sands to determine whether C (dry mass) could be a substitute for mineral nutrients (N, P, K, Ca, Mg and NA). The percentage of reproductive structures to total above-ground growth (reproductive effort; RE) was integrated over nine successive reproductive cycles. Only 0.5% of above-ground dry mass was allocated to seeds compared with 31% to total RE. Allocations of N (24%) and P (48%) to seeds, and N (44%) and P (65%) to RE were much higher. Allocations of K, Ca, Mg and Na to seeds (<1–3%), and RE (21–35%) were closer to that of dry mass. Relative allocation (RA) is defined as the proportion of a nutrient element allocated to a structure relative to its dry mass. RA of P to seeds was 91 and N was 44, but for K, Ca, Mg and Na ranged from only 6 for K to<1 for Na. Thus P, and to a lesser extent N, provide a much more sensitive measure of the relative cost of reproduction than C in this nutrient-limited system. 相似文献

All individuals of all known populations of Banksia goodii were assessed for seed production. Small populations produced no or only a few seeds per unit canopy area. Effects of population size on seed production per unit area and seed production per plant were present over the whole range of population sizes, indicating that even in large populations seed production may still not be at its maximum. Resource differences could not explain this disproportionate decrease in seed production with decline in population size, because there were no differences in soil properties and understorey or overstorey cover between the small and large populations. Although plants in small and large populations were similar in size, seed production per plant was much lower in small populations. This was not because plants in small populations produced fewer cones but because the fraction of these cones that was fertile was much lower. Five of the nine smallest populations (<200 m2) produced no fertile cones over the last 10 years. The number of seeds per fertile cone did not depend on population size.
