We describe herein the discovery of LASSBio-881 (3c) as a novel in vivo antinociceptive, anti-inflammatory, and in vitro antiproliferative and antioxidant compound, with a cannabinoid ligand profile. We observed that LASSBio-881 (3c) was able to bind to CB1 receptors (71% at 100microM) and also to inhibit T-cell proliferation (66% at 10microM) probably by binding to CB2 receptors, in a non-proapoptotic manner, different from anandamide (1). It was also demonstrated that LASSBio-881 (3c) had an important antioxidant profile toward free radicals (DPPH and hydroxyl), probably due to its particular redox behavior, which reflects the presence of both nitro and 3,5-di-tert-butyl-4-hydroxyphenyl sub-units, as demonstrated by cyclic voltammetry studies. Alzheimer’s Disease (AD) is a neurodegenerative disorder and the most common cause of dementia among the elderly. Efforts have been made to understand the genetic and epigenetic mechanisms involved in the development of this disease. As SORL1 (sortilin-related receptor) and SIRT1 (sirtuin 1) genes have been linked to AD pathogenesis, we aimed to investigate their mRNA expression and promoter DNA methylation in post mortem brain tissues (entorhinal and auditory cortices and hippocampus) from healthy elderly subjects and AD patients. We also evaluated these levels in peripheral blood leukocytes from young, healthy elderly and AD patients, investigating whether there was an effect of age on these profiles. The comparative CT method by Real Time PCR and MALDI-TOF mass spectrometry were used to analyze gene expression and DNA methylation, respectively. SORL1 gene was differently expressed in the peripheral blood leukocytes and might act as a marker of aging in this tissue. Furthermore, we found that SORL1 promoter DNA methylation might act as one of the mechanisms responsible for the differences in expression observed between blood and brain for both healthy elderly and AD patients groups. The impact of these studied genes on AD pathogenesis remains to be better clarified. 相似文献

We report on the clinical observation of a girl patient with few signs of cri-du-chat syndrome. The chromosomal analysis in
lymphocyte culture showed 46,XX,del(5)(p15.3) in 38% of cells. Psychological tests revealed motor, perceptive and visual-spatial
problems, as well as immaturity and emotional dependence. The phoniatric evaluation showed poor vocabulary, difficulty with
repeating words or numbers in sequence, and better receptive than expressive language. The spectrographic measurements showed
disturbance of fundamental frequency (F0) in vocal pronunciation. The anatomic findings of the laryngoscopic evaluation were normal, indicating that the voice and
speech problems were functional disorders. The present case revealed moderate clinical signs and vocal disturbance associated
with a low percentage of 5p-cells and the breakpoint at 5p15.3. The short terminal deletion with a possible loss of the critical
region for cat-like cry and the presence of a normal cell line, explain the cry not so typical at birth (weak but not high-pitched),

Background
Addison’s disease (AD) has been associated with an increased risk of cardiovascular disease. Glucocorticoid receptor polymorphisms that alter glucocorticoid sensitivity may influence metabolic and cardiovascular risk factors in patients with AD. The 9β polymorphism of the glucocorticoid receptor gene is associated with relative glucocorticoid resistance and has been reported to increase the risk of myocardial infarction in the elderly. We explored the impact of this polymorphism in patients with AD.Materials and Methods
147 patients with AD and 147 age, gender and ethnicity matched healthy controls were recruited. Blood was taken in a non-fasted state for plasma lipid determination, measurement of cardiovascular risk factors and DNA extraction.Results
Genotype data for the 9β polymorphism was available for 139 patients and 146 controls. AD patients had a more atherogenic lipid profile characterized by an increase in the prevalence of small dense LDL (p = 0.003), increased triglycerides (p = 0.002), reduced HDLC (p<0.001) an elevated highly sensitive C-reactive protein (p = 0.01), compared with controls. The 9β polymorphism (at least one G allele) was found in 28% of patients and controls respectively. After adjusting for age, gender, ethnicity, BMI and hydrocortisone dose per metre square of body surface area in patients, there were no significant metabolic associations with this polymorphism and hydrocortisone doses were not higher in patients with the polymorphism.Conclusions
This study did not identify any associations between the 9β polymorphism and cardiovascular risk factors or hydrocortisone dose and determination of this polymorphism is therefore unlikely to be of clinical benefit in the management of patients with AD. 相似文献6.

This study aimed to analyze the leaf anatomy of Manihot caerulescens, with emphasis on the ontogeny and secretion composition of the laticifers. Leaf samples were submitted the usual techniques in plant anatomy and histochemical tests to identify the main classes of secondary metabolites. The analyzes were performed using light and scanning electron microscopy. The leaves have a single layered epidermis coated by a uniform cuticle. The epidermal cells are elongated and papillose on the leaf abaxial surface of leaves. The mesophyll is isobilateral and the vascular bundle of the midrib is collateral. The laticifers are originated from ground meristem and procambium, and are of articulated anastomosing type. Besides the laticifers, two types of idioblasts were found: the secretory idioblasts, with a dense content, and the calcium oxalate druses idioblasts. Histochemical tests revealed different classes of chemicals for both secretory idioblasts and laticifers. In idioblasts, a positive reaction was detected for the general phenolic compounds, proteins, total polysaccharides and tannins. The laticifers reacted positively to total lipids and acids, essences, total polysaccharides and rubber. The individuals analyzed here are from caatinga and their leaf anatomical structure showed to be different from that described for the same species growing in the cerrado. 相似文献

Background
The ability to apply standard and interoperable solutions for implementing and managing medical registries as well as aggregate, reproduce, and access data sets from legacy formats and platforms to advanced standard formats and operating systems are crucial for both clinical healthcare and biomedical research settings.Purpose
Our study describes a reproducible, highly scalable, standard framework for a device registry implementation addressing both local data quality components and global linking problems.Methods and Results
We developed a device registry framework involving the following steps: (1) Data standards definition and representation of the research workflow, (2) Development of electronic case report forms using REDCap (Research Electronic Data Capture), (3) Data collection according to the clinical research workflow and, (4) Data augmentation by enriching the registry database with local electronic health records, governmental database and linked open data collections, (5) Data quality control and (6) Data dissemination through the registry Web site. Our registry adopted all applicable standardized data elements proposed by American College Cardiology / American Heart Association Clinical Data Standards, as well as variables derived from cardiac devices randomized trials and Clinical Data Interchange Standards Consortium. Local interoperability was performed between REDCap and data derived from Electronic Health Record system. The original data set was also augmented by incorporating the reimbursed values paid by the Brazilian government during a hospitalization for pacemaker implantation. By linking our registry to the open data collection repository Linked Clinical Trials (LinkedCT) we found 130 clinical trials which are potentially correlated with our pacemaker registry.Conclusion
This study demonstrates how standard and reproducible solutions can be applied in the implementation of medical registries to constitute a re-usable framework. Such approach has the potential to facilitate data integration between healthcare and research settings, also being a useful framework to be used in other biomedical registries. 相似文献8.

BackgroundThe application of the Shuttle Walking Test (SWT) to assess cardiorespiratory fitness and the intensity of this test in healthy participants has rarely been studied. This study aimed to assess and correlate the cardiorespiratory responses of the SWT with the cardiopulmonary exercise testing (CEPT) and to develop a regression equation for the prediction of peak oxygen uptake (VO2 peak) in healthy sedentary adult men.MethodsIn the first stage of this study, 12 participants underwent the SWT and the CEPT on a treadmill. In the second stage, 53 participants underwent the SWT twice. In both phases, the VO2 peak, respiratory exchange ratio (R), and heart rate (HR) were evaluated.ResultsSimilar results in VO2 peak (P>0.05), R peak (P>0.05) and predicted maximum HR (P>0.05) were obtained between the SWT and CEPT. Both tests showed strong and significant correlations of VO2 peak (r = 0.704, P = 0.01) and R peak (r = 0.737, P<0.01), as well as the agreement of these measurements by Bland-Altman analysis. Body mass index and gait speed were the variables that explained 40.6% (R2 = 0.406, P = 0.001) of the variance in VO2 peak. The results obtained by the equation were compared with the values obtained by the gas analyzer and no significant difference between them (P>0.05) was found.ConclusionsThe SWT produced maximal cardiorespiratory responses comparable to the CEPT, and the developed equation showed viability for the prediction of VO2 peak in healthy sedentary men. 相似文献

This work was based on the analysis of digital images of histochemical profile from subcutaneous lesions in sporotrichosis
(ST) and chromoblastomycosis (CM) patients. An additional aim was the detection of carbohydrate expression using lectin histochemical
analysis of the different carbohydrates in the fungal cell wall from four different species (Sporothrix
schenckii, Fonsecaea pedrosoi, Phialophora verrucosa, and Cladophialophora carrionii) associated with diseases mentioned earlier. Slides from tissue biopsies from ST and CM positive patients (n = 10, each) were stained according to routine techniques. Slides were incubated with 25 μg/ml of Con A lectins and WGA conjugated
to peroxidase. Digital image analysis was carried out in a workstation using OPTIMAS™ software system. Routine histochemistry
results indicated that there is significantly higher collagen deposition and elastic fibers in ST characteristic lesions compared
with that found in CM cases. The ST interstitial fibrosis area was larger than in CM lesions. Comparative lectin binding showed
a positive and intense lectin staining pattern in the cell wall of S. schenckii, suggesting a higher expression of glucose/mannose and N-acetyl glucosamine in their cell surface as evidenced by Con A and WGA, respectively. However, these lectins were not effective
to recognize some carbohydrates moieties in the F. pedrosoi, P. verrucosa, and C. carrionii. Such findings contribute to additional information about specific recognition processes between fungal parasites and their
host cell targets may be mediated by the interaction of carbohydrate-binding proteins, such as lectins, on the surface of
one type of cell that combine with complementary sugars on the surface of another cells into fibro-connective tissues associated

The carbon (C) concentration and flux, as dissolved organic carbon (DOC), particulate organic carbon (POC) and macrodetritus (MD), were quantified through 4 tidal cycles in a mangrove tidal creek in Southeastern Brazil. DOC was the major fraction of the total C concentration, accounting for 68 and 61% of the total C concentration during ebb and flood periods respectively. Concentrations of DOC (Ebb = 3.41 +/- 0.57 mgC.L-1 and Flood = 3.55 +/- 0.76 mgC.L-1) and POC (Ebb = 1.73 +/- 0.99 mgC.L-1 and Flood = 1.28 +/- 0.45 mgC.L-1) were relatively similar during the four tidal cycles. Macrodetritus presented a wide variation with concentration peaks probably related to external forces, such as winds, which enrich the ebb flow with leaf litter. DOC and POC fluxes depended primarily on tidal and net water fluxes, whereas MD fluxes were not. The magnitude of the DOC and POC fluxes varied with the area flooded at high tide, but not the MD fluxes. DOC was the major form of carbon export to Sepetiba Bay. During the four tidal cycles, the forest exported a total of 1.2 kg of organic carbon per ha, mostly as DOC (60%), followed by POC (22%) and MD (18%). 相似文献
