Metabolomic investigation of the freezing-tolerant Arabidopsis mutant esk1 revealed large alterations in polar metabolite content in roots and shoots. Stress metabolic markers were found to be among the most significant metabolic markers associated with the mutation, but also compounds related to growth regulation or nutrition. The metabolic phenotype of esk1 was also compared to that of wild type (WT) under various environmental constraints, namely cold, salinity and dehydration. The mutant was shown to express constitutively a subset of metabolic responses which fits with the core of stress metabolic responses in the WT. But remarkably, the most specific metabolic responses to cold acclimation were not phenocopied by esk1 mutation and remained fully inducible in the mutant at low temperature. Under salt stress, esk1 accumulated lower amounts of Na+ in leaves than the WT, and under dehydration stress its metabolic profile and osmotic potential were only slightly impacted. These phenotypes are consistent with the hypothesis of an altered water status in esk1 , which actually exhibited basic lower water content (WC) and transpiration rate (TR) than the WT. Taken together, the results suggest that ESK1 does not function as a specific cold acclimation gene, but could rather be involved in water homeostasis.  相似文献

The use of long-acting contraceptives as an effective population control mechanism for free-ranging lions was investigated during a three-year study in the Etosha National Park on five lion prides. Lions were immobilized with ketamine hydrochloride and xylazine hydrochloride. Following immobilization, lionesses were weighed, measured and aged, while serum steroid hormone levels were analysed and vaginal smears were obtained. Physical condition and sexual state were assessed. Individuals were used as control animals or given melengestrol acetate (contraceptive) implants, branded and released; the animals were observed for possible changes in behaviour, birth rate and mortality. Ten treated lionesses were recaptured at irregular intervals to reassess weight and steroid hormone levels, while three lionesses had their implants removed to determine if their fertility would return. The contraceptives prevented pregnancy, were reversible when removed and did not alter lion behaviour significantly, except that sexual behaviour was not recorded in treated lionesses.  相似文献

Experiments on the germination of the spores of Funaria have established the following.

    Some growth substances such as vitamins slightly promote the geotropic response of germinating spores in darkness.

    Glucose, even in concentrations as low as 10 mg./L, strongly stimulates the geotropic response.

  • 3. 

    Light (at an intensity of 1000 lux and more) strongly inhibits the geotropic response. This effect is probably phototonic and can be partly counteracted by glucose.

  • 4. 

    The nature of the phototropic and geotropic reactions of chloronema is variable, and depends to a certain extent on environmental influences.

We monitored the radioligand-binding characteristics of thyrotropin-releasing hormone (TRH) receptors, functional activity of Gq/11α proteins, and functional status of the whole signaling cascade in HEK293 expressing high levels of TRH receptors and G11α. Our analyses indicated that disruption of plasma membrane microdomains by cholesterol depletion did not markedly influence the binding parameters of TRH receptors, but it altered efficacy of signal transduction. The functional coupling between TRH receptor and Gq/11α was assessed by agonist-stimulated [35S]GTPγS binding, and results of these measurements pointed out to significantly lower potency of TRH to mediate G protein activation in the plasma membrane fraction isolated from cholesterol-depleted cells; there was a shift in sensitivity by one order of magnitude to the higher concentrations. A markedly lower sensitivity to stimulation with TRH was also observed in our experiments dealing with determination of hormone-induced Ca2+ response. These data suggest that the intact structure of plasma membranes is an important optimum signal transduction initiated by TRH receptors and mediated by Gq/11α proteins.  相似文献

The very diverse social systems of sweat bees make them interesting models to study social evolution. Here we focus on the dispersal behaviour and social organization of Halictus scabiosae , a common yet poorly known species of Europe. By combining field observations and genetic data, we show that females have multiple reproductive strategies, which generates a large diversity in the social structure of nests. A detailed microsatellite analysis of 60 nests revealed that 55% of the nests contained the offspring of a single female, whereas the rest had more complex social structures, with three clear cases of multiple females reproducing in the same nest and frequent occurrence of unrelated individuals. Drifting among nests was surprisingly common, as 16% of the 122 nests in the overall sample and 44% of the nests with complex social structure contained females that had genotypes consistent with being full-sisters of females sampled in other nests of the population. Drifters originated from nests with an above-average productivity and were unrelated to their nestmates, suggesting that drifting might be a strategy to avoid competition among related females. The sex-specific comparison of genetic differentiation indicated that dispersal was male-biased, which would reinforce local resource competition among females. The pattern of genetic differentiation among populations was consistent with a dynamic process of patch colonization and extinction, as expected from the unstable, anthropogenic habitat of this species. Overall, our data show that H. scabiosae varies greatly in dispersal behaviour and social organization. The surprisingly high frequency of drifters echoes recent findings in wasps and bees, calling for further investigation of the adaptive basis of drifting in the social insects.  相似文献

Molluscan communities were studied at 114 castles in the CzechRepublic. Altogether 70% of the land fauna species of the CzechRepublic were found at these sites. A range of environmentalfactors were recorded at each, and analysed for their influenceon species diversity. The variability of the molluscan communitieswas assessed. It was shown that castles act as islands (positiveeffect of castle size on species number) and that a high degreeof castle disintegration negatively affected species diversity.Limestone as a rock substrate was found to be a very importantfactor, while the effect of calcium in tree litter was unimportant.Species indicative of some environmental variables were identified.A complete literature review on castle-related molluscan faunawas performed for the Czech Republic. Castles present a characteristicset of habitats markedly differing from the surrounding landscape.Thus, they represent significant refuges for many species, asindicated by the high species diversity and the presence ofrare and geographically isolated species. ABSTRACT. Sand fly and mosquito gregarines have been lumped for a long time in the single genus Ascogregarina and on the basis of their morphological characters and the lack of merogony been placed into the eugregarine family Lecudinidae. Phylogenetic analyses performed in this study clearly demonstrated paraphyly of the current genus Ascogregarina and revealed disparate phylogenetic positions of gregarines parasitizing mosquitoes and gregarines retrieved from sand flies. Therefore, we reclassified the genus Ascogregarina and created a new genus Psychodiella to accommodate gregarines from sand flies. The genus Psychodiella is distinguished from all other related gregarine genera by the characteristic localization of oocysts in accessory glands of female hosts, distinctive nucleotide sequences of the small subunit rDNA, and host specificity to flies belonging to the subfamily Phlebotominae. The genus comprises three described species: the type species for the new genus— Psychodiella chagasi ( Adler and Mayrink 1961 ) n. comb., Psychodiella mackiei ( Shortt and Swaminath 1927 ) n. comb., and Psychodiella saraviae ( Ostrovska, Warburg, and Montoya-Lerma 1990 ) n. comb. Its creation is additionally supported by sequencing data from other gregarine species originating from the sand fly Phlebotomus sergenti . In the evolutionary context, both genera of gregarines from mosquitoes ( Ascogregarina ) and sand flies ( Psychodiella ) have a close relationship to neogregarines; the genera represent clades distinct from the other previously sequenced gregarines.  相似文献

The southern range limit of the planktotrophic, intertidal whelk, Morula marginalba, occurs immediately to the north of the south‐east Australian biogeographical barrier and we predict that its range may be limited by dispersal and biotic interactions. The barrier includes several hundred kilometers without suitable habitat and is the convergence zone of major ocean currents. Within the barrier region, along‐shore larval transport is expected to be erratic, with water moving offshore to form long‐lived eddies. Through comparison of central and range edge populations, we test for evidence of recruitment limitation and interspecific competition with the whelk, Haustrum vinosum, which occurs sympatrically at the range edge. We found similar levels of recruitment and allozyme genetic variation within centre and edge populations where H. vinosum was absent, implying that populations are strongly interconnected. Nevertheless, recruitment and abundance of M. marginalba were lower at its extreme range limit, where it occurs with H. vinosum. Indeed, we detected significant inverse correlations between the abundance of these species, although their habitat use was similar and they consumed similar prey. These lines of evidence suggest a restricted recruitment across the barrier limits the range of M. marginalba but emphasize that multiple factors may act to set species range limits. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 103 , 849–862.  相似文献
