Paepalanthus subgenus Xeractis (Eriocaulaceae) comprises 28 recognized species endemic to the Espinhaço Range, in Minas Gerais state, Brazil. Most species of the subgenus are restricted to small localities and critically endangered, but still in need of systematic study. The monophyly of the subgenus has already been tested, but only with a few species. Our study presents the first phylogenetic hypothesis within the group, based on morphology. A maximum parsimony analysis was conducted on a matrix of 30 characters for 30 terminal taxa, including all species of the subgenus and two outgroups. The biogeographical hypotheses for the subgenus were inferred based on dispersal–vicariance analysis (DIVA). The analysis provided one most‐parsimonious hypothesis that supports most of the latest published subdivisions (sections and series). However, some conflicts remain concerning the position of a few species and the relationships between sections. The distribution and origin(s) of microendemism are also discussed, providing the ground for conservation strategies to be developed in the region. © 2011 The Linnean Society of London, Botanical Journal of the Linnean Society, 2011, 167 , 137–152.  相似文献

A rapid, nondestructive, reproducible and cheap DNA extraction method from body mucus and buccal cells of northern pike and brown trout is described. Buccal cells and body mucus were sampled on FTA Cards; the captured DNA was used directly for microsatellite and polymerase chain reaction–restriction fragment length polymorphism (PCR–RFLP) analyses. A complete concordance with control DNA was found. The genotyping error rate for microsatellite ranged from 1.9% to 3.3% for the northern pike and brown trout, respectively. This methodology, using for the first time these materials as a fish DNA source, combines speed of sampling and processing, with a twofold to a threefold time and costs saving.  相似文献

Small RNA (sRNA)‐guided processes, referred to as RNA silencing, regulate endogenous and exogenous gene expression. In plants and some animals, these processes are noncell autonomous and can operate beyond the site of initiation. Viroids, the smallest self‐replicating plant pathogens known, are inducers, targets and evaders of this regulatory mechanism and, consequently, the presence of viroid‐derived sRNAs (vd‐sRNAs) is usually associated with viroid infection. However, the pathways involved in the biogenesis of vd‐sRNAs are largely unknown. Here, we analyse, by high‐throughput pyrosequencing, the profiling of the Hop stunt viroid (HSVd) vd‐sRNAs recovered from the leaves and phloem of infected cucumber (Cucumis sativus) plants. HSVd vd‐sRNAs are mostly 21 and 22 nucleotides in length and derived equally from plus and minus HSVd RNA strands. The widespread distribution of vd‐sRNAs across the genome reveals that the totality of the HSVd RNA genome contributes to the formation of vd‐sRNAs. Our sequence data suggest that viroid‐derived double‐stranded RNA functions as one of the main precursors of vd‐sRNAs. Remarkably, phloem vd‐sRNAs accumulated preferentially as 22‐nucleotide species with a consensus sequence over‐represented. This bias in size and sequence in the HSVd vd‐sRNA population recovered from phloem exudate suggests the existence of a selective trafficking of vd‐sRNAs to the phloem tissue of infected cucumber plants.  相似文献

Abstract: Direct studies of mammalian carnivores are challenging due to the animals' secretive nature and the high costs associated with their capture and handling. Use of noninvasive hair sampling to survey these reclusive species has great potential as an alternative, with wide applicability in ecology and conservation. Hair-trapping has been extensively used for focal studies of temperate mammals, but its use and applicability as a means to survey mammals in tropical environs has never been addressed. We evaluated the effectiveness of 2 hair-trap types and 2 scents along an elevational gradient within El Cielo Biosphere Reserve (ECBR, Mexico) to detect presence of carnivores. Hair-traps that used roofing nails as a hair-collecting surface collected more hairs and detected a greater number of species than did hair-traps that used velcro strips. Different scent treatments (commercial fragrance and catnip oil) did not differ for these same variables. Of successful nail hair-traps, 60% collected ≥20 hairs (max. = 439), providing enough material for DNA analyses. Hair-trap surveys detected 74% of the potential target mammal species at ECBR with only 19 days of field effort. Developing countries have limited budgets for biodiversity monitoring and hair-traps compare favorably with other methods with a high cost-benefit ratio. Hair-traps are inexpensive, portable, can be made with over-the-counter materials, and can be successfully used to collect data applicable to population and genetic studies of tropical carnivores.  相似文献

The interest shown by ecologists in antioxidants and oxidative stress as potential modulators of life‐history trade‐offs has expanded greatly in recent years. However, we still know very little about natural variation in oxidative damage and antioxidant capacity in free‐living animals. In this study, we describe the natural variation in three components of oxidative balance in nestlings and breeding females in free‐living Great Tits Parus major and Common Starlings Sturnus vulgaris in central Italy, and relate these to breeding conditions and life‐history traits. Our results suggest that there are associations among oxidative physiology, reproductive activity, growth pattern and season in wild birds, but that the nature of these associations might be species‐specific rather than general across species.  相似文献

In the genus Gunnera , the species of subgenus Panke are unique in having large, triangular scales between the leaves on the rhizomes. The morphological significance of these scales has been extensively debated in the past. They have been interpreted as stipules, ligules, or cataphylls, with attempts made to identify homologous structures in representatives of the other subgenera. In the stoloniferous subgenus Misandra, the shoot apex is covered by a hoodlike structure, usually termed ochrea, generally considered homologous to the scales in Panke . Efforts to find similar structures in the remaining subgenera were unsuccessful. Due to the lack of a phylogenetic hypothesis, the homology of the scales has remained unresolved. In this study, the shoot apices of species from all subgenera were examined by light and scanning electron microscopy and the results interpreted in a phylogenetic context. The tip of the stolons in subgenera Pseudogunnera and Milligania carries two opposite cataphylls at the base of the new shoot. These are homologous to the ochrea in Misandra which is in turn homologous to the scales in Panke ; the latter are thus neither stipules nor ligules but cataphylls.
