Using Toyopearl and cyclohexane: cyclohexanol solvent, fourCl-containing Chls were separated from 36Cl-labeled cells ofthe blue-green, Plectonema boryanum. In normally grown cells,all four Cl-containing chlorophylls amounted to less than 1/2,000of the total Chi and about 1/50 of P700, values much lower thanpreviously reportedcontents of Chi RC I, and varied from algato alga. The level of Cl-containing Chi was markedly enhancedwhen the cells were poisoned with methyl viologen. Homologous pairing is a key step in homologous genetic recombination. In the early stage of trials for the identification of homologous pairing-promoting proteins from a fission yeast, Schizosaccharomyces pombe, we treated DNA products with phenol in the presence of a salt for the removal of tightly bound proteins from DNA before the assay, but we found that this treatment caused very efficient protein-independent double-strand formation from complementary single-stranded DNAs. Using an assay including the phenol treatment, we detected another species of apparent homologous pairing-promoting proteins in the nuclei, in addition to a homologous pairing-promoting protein consisting of three components which we reported previously. However, studies involving the use of an assay without the phenol-treatments revealed that the second one was not really a homologous pairing-protein. Thus, the protein-independent double-strand formation by phenol-treatment in the presence of a salt could cause the erroneous identification of homologous pairing-promoting proteins. 相似文献

Summary We isolated three different repetitive DNA sequences from B. campestris and determined their nucleotide sequences. In order to analyze organization of these repetitive sequences in Brassica, Southern blot hybridization and in situ hybridization with metaphase chromosomes were performed. The sequence cloned in the plasmid pCS1 represented a middle repetitive sequence present only in B. campestris and not detected in closely related B. Oleracea. This sequence was localized at centromeric regions of six specific chromosomes of B. campestris. The second plasmid, pBT4, contained a part of the 25S ribosomal RNA gene, and its copy number was estimated to be 1,590 and 1,300 per haploid genome for B. campestris and B. oleracea, respectively. In situ hybridization with this sequence showed a clear signal at the NOR region found in the second largest chromosome of B. Campestris. The third plasmid, pBT11, contained a 175-bp insert that belongs to a major family of tandem repeats found in all the Brassica species. This sequence was detected at centromeric regions of all the B. campestris chromosomes. Our study indicates that in situ hybridization with various types of repetitive sequences should give important information on the evolution of repetitive DNA in Brassica species. 相似文献

Explants excised from strawberry (Fragaria x ananassa Duch.) plantlets were cultured in vitro for 21 days on half-strength MS (Murashige & Skoog 1962) basal liquid medium with 20 g l-1 sucrose and without sugar in the vessels capped with gas permeable microporous polypropylene film. The experiments were conducted under CO2 nonenriched (350–450 mol mol-1 in the culture room) and CO2 enriched (2,000 mol mol-1 during the photoperiod in the culture room) conditions with a PPF (photosynthetic photon flux) of 200 mol m-2 s-1. The CO2 concentration in the vessels decreased to approximately 200 mol mol-1 during the photoperiod on day 21 under CO2 nonenriched conditions. Immigration of the brown planthopper, Nilaparvata lugens, exhibiting various responses to density in relation to wing morphism 总被引:6,自引:0,他引:6
Considerable inherent variations in the relation between macropterous and brachypterous wing forms, and nymphal density were found in field populations of the brown planthopper, Nilaparvata lugens Stål (Homoptera: Delphacidae), collected from various locations in Japan. When compared under uniform laboratory rearing conditions, most of the female populations exhibited higher ratios of macropters with increasing nymphal density, but some showed extremely high proportion of brachypters and the others were highly macropterous, over broad ranges of density. These results indicate the possibility that the planthoppers in Japan, which are known not to persist in winter, are derived from different migration sources.About ten generations of successive selection for brachyptery from a population showing usual density-dependent wing morphism generated populations similar to highly brachypterous ones mentioned above. Genetic analysis of the inheritance of wing morphism revealed that brachyptery in the females was controlled by a single pair of dominant alleles. However, in the males wing forms did not segregate so clearly in the crossing experiments. This suggests that wing morphism in N. lugens in under sex-limited inheritance.
Einwanderung von Nilaparvata lugens mit unterscheidlicher Reaktion auf Populationsdischte bei der Flügelausbildung
Zusammenfassung In Feldpopulationen von Nilaparvata lugens Stål., welche in verschiedenen Regionen Japans gesammelt wurden, bestand in der Beziehung zwischen makropteren bzw. brachypteren Flügelformen und der Larvendichte eine beträchtliche Variation. Unter einheitlichen Zuchtbedingungen im Laboratorium stieg der Makropterenanteil bei den meisten Weibchenpopulationen mit steigender Temperatur; bei einigen Populationen hingegen war entweder der Brachypterenanteil oder der Makropterenanteil extrem hoch und zwar über weite Dichtebereiche. Dies deutet auf die Möglichkeit hin, dass die Zikade in Japan, wo sie bekanntlich nicht überwintert, jeweils aus verschiedenen Quellen einwandert.Wenn eine Population mit der üblichen dichteabhängigen Flügelausbildung 10 Generationen lang auf Brachypterie selektioniert wurde, entstanden Populationen, die den erwähnten hochbrachypteren Populationen aus dem Feld glichen. Die genetische Analyse der Vererbung der Brachypterie ergab, dass bei Weibchen ein einzelnes dominantes Allel verantwortlich ist. Bei Männchen dagegen trennten sich bei Kreuzungsexperimenten die Flügelformen nicht so klar. Dies deuted auf Unterschiede zwischen den Geschlechtern bei der Vererbung der Flügelformen.相似文献

Summary The translation activity of mRNA coding for argininosuccinate synthetase in total RNA extracted from the liver of three patients with quantitative-type citrullinemia was determined using a cell-free translation system. In two patients, the hepatic content of the enzyme was about 20% of the control value, whereas translatable mRNA level for the enzyme was similar to or slightly lower than those of control livers. In the third patient, the enzyme content was about 50% of the control value, and mRNA activity for the enzyme was low normal. These results indicate that at least in the first two patients, the decrease in the enzyme protein is due either to increased degradation of the enzyme or to decreased translation in the patient's liver. 相似文献

We investigated whether alveolar surface force increased and participated in the lung pressure-volume relationship in bleomycin-induced pulmonary fibrosis in hamsters and, if so, whether lung surfactant was hampered in the lungs. On the air-filled pressure-volume curve, decreases of lung volume from control level were significantly higher at 3-8 cmH2O pressure on day 10 than on day 30. Because the change of lung tissue elasticity evaluated from the saline-filled pressure-volume curve was equal for the 2 days, the higher decrease of air volume on day 10 was due primarily to contribution of alveolar surface force. Pressure differences between deflation limbs of air-filled and saline-filled pressure-volume curves, which represented net alveolar surface force, were significantly higher at any lung volume between 50 and 90% total lung capacity on day 10, but almost no significance was observed on day 30. Phospholipid concentration in bronchoalveolar lavage fluid significantly decreased on day 10 but had improved by day 30. Analysis of phospholipid species in purified lung surfactant showed decreased fractions of disaturated phosphatidylcholine and phosphatidylglycerol on day 10. Surface-active properties of the surfactant, measured by a modified Wilhelmy balance, were remarkably hampered on day 10, but most of them had improved by day 30. We consider that the quantitative and functional abnormalities of lung surfactant have a part in the aggravation of lung mechanics in the acute phase of pulmonary fibrosis. 相似文献
