Prevalence of uterine myoma (MU) was estimated in several Moscow districts. The overall average estimate of the MU prevalence is 2.45% among women of all groups. The prevalence MU estimates increase with the age, its maximum value reaching 8.31% at the age of 50 years. The morbidity risk estimates increased with the age as well, the maximum value being 2.98% at the age of 40-44 years. The value of "cumulative" morbidity risks, i. e. the probability to be affected, is 9.74% for a population living long enough, this value being based on the age-specific estimates of morbidity risks. Taking into consideration the autopsy data, indicating that frequency of MU, including small myomatous nodes, is 20%, the conclusion is made that MU is manifested by clinically expressed disturbances (urging a woman to address to a doctor) in 50% of cases only. Epidemiological data obtained are to be used later for genetic analysis of familial data on MU.  相似文献

Supramembrane structures that connect conjugating agrobacterial cells were visualized for the first time by transmission electron microscopy. The primary contact of cells during conjugation was shown to occur through the formation of long pili containing no VirB1 protein. Pretreatment of agrobacterial cells with acetosyringone resulted in a six-to tenfold increase in the transfer frequency of plasmid pTd33 at 19–25°C and had almost no effect at 30°C. The transfer of plasmid pTd33 fromA. tumefaciens strain GV3101 to plasmid-freeA. tumefaciens strain UBAPF-2 was 16 times decreased after the centrifugation of cells. The transfer efficiency of plasmid pTd33 fromA. tumefaciens strain LBA2525 (virB2::lacZ) to plasmid-freeA. tumefaciens strain UBAPF-2 was one order of magnitude lower than the transfer from the wild-typeA. tumefaciens strain GV3101. Treatment of donor cells with 0.01% SDS before mating decreased the transfer efficiency by a factor of 26. The role of pili in the establishment of contact between conjugating cells of agrobacteria is discussed.  相似文献

Agrobacterial cells produced straight microfibrils not only when in contact with wheat seedling roots, but also when in contact with each other. After 2 h of incubation, agrobacterial cells were found to form aggregates, in which the cells were in contact either directly or through thick straight microfibrils (bridges) of an unknown composition. The majority of the microfibrils were susceptible to attack by cellulase, although some of them showed resistance to this enzyme. Like the wild-type flagellated agrobacteria, their bald mutants produced long straight microfibrils. The cell surface structures of agrobacteria were examined by labeling them immunocytochemically with colloidal gold–conjugated antibodies against O-specific lipopolysaccharides, Vir proteins, and cellulase. Agrobacterial cells treated with acetosyringone and brought into contact were found to contain subpolar and polar cell surface structures. Antibodies against the VirB2 protein were able to interact with a tuft of thin microfibrils located on one pole of the agrobacterial cell whose virgenes were induced by acetosyringone but were unable to interact with the surface structures of the agrobacterial cells aggregated in liquid medium in the absence of wheat seedlings.  相似文献

Agrobacterial Transformation of Uninjured Plants   总被引:1,自引:0,他引:1  
The capacity for agrobacterial transformation (using Agrobacterium tumefaciens strain GV3101 with the activated vir genes) was investigated in uninjured seedlings of tobacco (Nicotiana tabacum L.), rice (Oryza sativa L.), and wheat (Triticum aestivum L.) along with the effect of various factors on the frequency of tobacco seedling transformation. Transformed cells were found at the base of the rice leaf sheath and in the upper part of tobacco leaves, whereas they were absent from wheat leaves. Illumination and temperature, which were below and over the optimum, were the factors limiting agrobacterial transformation of uninjured plants. Preincubation of tobacco seedlings in 100 M ABA, 10 mM EGTA, and 0.1 mM colchicine decreased the frequency of agrobacterial T-DNA penetration into plant cells. The authors conclude that transformation occurs predominantly in young growing cells, and its frequency is related to the state of stomata and plasmodesmata of the target plant cells. The elements of the cytoskeleton probably participate in T-complex transport.  相似文献

An express method for determination of antibiotic sensitivity in the strains of Cl. perfringens of type A using Soviet dry nutrient media and antibiotics is proposed. The criteria for estimation of the level of the antibiotic sensitivity of the causative agent of gas gangrene in short periods on the basis of comparison of the data of the antibiotic agar diffusion procedure and the antibiotic MIC were worked out. Twelve antibiotics and 45 collection strains of Cl. perfringens of type A were used in the experiment. The antibiotic agar diffusion method with the use of the nutrient media, microbial load and cultivation conditions developed by the authors is recommended for tentative determination of the antibiotic sensitivity in Cl. perfringens of type A for 4 hours. The use of the agar diffusion method and determination of the antibiotic MIC provided complete estimation of tha antibiotic sensitivity of Cl. perfringens of type A within not more than 24 hours.  相似文献

Electron microscopy of noncentrifugated agrobacterial cells on a nitrocellulose membrane labeled with colloid gold-conjugated antibodies to VirB1 showed that the labeled complex bound to acetosyringone (AS)-induced cells but failed to form red-colored stains during incubation with Ti aplasmid cells. Supramembrane structures of AS-treated A. tumefaciens cells were for the first time visualized by transmission electron microscopy. Colloid gold labeling of VirB2-specific antibodies showed that VirB2 proteins produce long thin pilus structures emerging at the poles of AS-induced agrobacterial cells but never on the surface untreated with AS and Ti-plasmid-free agrobacterial cells. As a rule, one (or rarely two) thread-like connections and bridges were observed between the cells at the primary contact stage. The bridges were not destroyed by SDS, did not react with VirB2-specific antibodies, and remained visible at 30 degrees C. Visible close contacts between mating bacteria did not cease after SDS treatment. SDS pretreatment of donor cells or a mating cell suspension significantly modified the efficiency of pTd33 plasmid transfer from donor to recipient agrobacterial cells. In the presence of AS the optimal temperature for transfer was 25 degrees C. The frequency of plasmid pTd33 transfer from A. tumefaciens via vir-dependent pathway decreased 2-4-fold due to increase of temperature from 19.25 to 31 degrees C.  相似文献

The kidney-dependent increase of the haemolytic activity of blood serum after acute decompensated blood loss was demonstrated in the experiments on rats. The preparative ion-exchange chromatography on DEAE-Toyoperal was used to separate the haemolytic active component of the posthaemorrhagic blood serum with the max value of the specific activity of 6.86 A/micrograms protein. The analysis of the separated component by size-exclusion chromatography on TSK-G3000-SW column indicated a molecular mass of 80-100 kDa. In injection in the circulatory system in in vivo experiments the dose-dependent effect of the action of separated component was demonstrated.  相似文献

The dependence of blood loss quantitative indices (duration, number and volume of exfusions) on the initial state of the blood protein system was studied in experiments with dogs. The true correlation was found between the macroprotein contents in plasma and the duration of the arterial pressure compensation phase at the level of 5.3 kPa (40 mm Hg). Hypermacroproteinemia is evident of low organism stability (resistance) and is an unfavourable prognostic characteristic in experimental blood loss.  相似文献

Exocellular structures containing VirB2 proteins were, for the first time, localized on the surface of Agrobacterium by transmission electron microscopy. Using colloidal gold (CG)-labeled VirB2-specific antibodies, it was shown that VirB2 proteins enter into the composition of short surface pili, which emerge at the poles of acetosyringone (AS)-induced Agrobacterium cells. However, cells of the Ti plasmidless A. tumefaciens strain UBAPF-2 and cells not induced with AS were incapable of pilus synthesis. In suspension, mating Agrobacterium cells were connected together by short thick bridges. It was found that these bridges did not include as part of their structure CG-labeled VirB1 and VirB2 proteins. We did not find the tetracycline-resistant transconjugants after mating of A. tumefaciens donor cells harboring binary systems with plasmid-free A. tumefaciens GM-I 9023 in vir-induced and vir-uninduced conditions. However, the same strains can transfer pSUP106 plasmid via a vir-dependent way. We found that activated vir genes slightly stimulate pTd33 plasmid transfer via a tra-dependent pathway to plasmid-free strain UBAPF-2. It seems, that vir-induced T-DNA/plasmid DNA transfer machinery is not essential for the conjugation process between agrobacterial cells but may participate in this process.  相似文献
