Taurodeoxycholate 7α-monooxygenase was partially purified from rat liver microsomes. The enzyme was solubilized with cholate, fractionated with polyethylene glycol and chromatographed on a Sepharose 4B column with cholate as ligand. The enzyme activity was eluted from the column into the fraction eluted with 50 mM phosphate buffer containing cholate and KCl, whereas the benzphetamine demethylase activity was eluted in the non-bound fraction. Thus it was established that both enzymes are different entities. The taurodeoxycholate 7α-monooxygenase activity was reconstituted from the partially purified cytochrome P-450, highly purified NADPH-cytochrome P-450 reductase, dilauroylglyceryl-3-phosphorylcholine and NADPH.  相似文献

After the addition of actin to serum, the binding of actin to serum actin-binding proteins was analyzed by the method of immunoblotting using monospecific antibodies against vitamin D-binding protein (DBP) (group-specific component, Gc), human skeletal actin and human plasma gelsolin. When increasing amounts of globular actin were added to serum, actin bound to DBP preferentially. After exhausting DBP, actin began to bind to plasma gelsolin. When equally increasing amounts of filamentous actin were added to serum, actin was bound to both plasma gelsolin and DBP, and then uncomplexed DBP removed one actin molecule from gelsolin-actin 1:2 complex, resulting in a gelsolin-actin 1:1 complex. These results support the theory that the actin-depolymerizing activity of serum is due to the concerted role of plasma gelsolin and DBP.  相似文献

A sensitive method for detecting metallothioneins (MTs) by use of silver staining after sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of carboxymethylated MTs was developed. Carboxymethylation of metallothioneins is indispensable because it prevents their aggregation, thereby allowing each of them to be detected as a single band by SDS-PAGE. However, when the gel was subjected to the silver-staining method of C. R. Merril, D. Goldman, S. A. Sedman, and M. H. Ebert [(1981) Science 211, 1437-1438], the image of carboxymethylated purified MTs was totally negative. Pretreatment of the gel with 1% sodium thiosulfate just prior to the silver-staining procedure successfully reversed the negative image of carboxymethylated MTs. Further, they could be detected with a limit of nanogram levels per lane. This method can be applied to MTs in cell extracts from cultured cell lines treated with cadmium or to those from liver of cadmium-intoxicated mice.  相似文献

The alkaline solution method for detecting uterine implantation sites (metrial glands or spots after delivery), which indicate the position of the embryo, was compared with conventional methods using Turnbull blue (Salewski's technique) and benzyl benzoate (Orsini's technique) in rats. When rat uteri were immersed directly in 2% sodium hydroxide [NaOH], potassium hydroxide [KOH] solution or one of several other alkaline solutions for one hour, the uteri were cleared. Then the metrial glands could be observed without further staining, and the spots after delivery with further staining. After alkaline treatment, unstained metrial glands appeared as yellowish-white degenerated masses and stained spots after delivery appeared as yellowish-brown patches covered with yellowish-white lumps. These yellowish-brown or yellowish-white sites were probably stained by the iron in hemosiderin deposits in the placental scar or in a vestige of the metrial glands left over from pregnancy. Formalin-fixed uteri were not cleared by immersion in a high concentration of NaOH, KOH or other alkaline solutions, but the spots after delivery were stained by this treatment. Turnbull blue did not stain the metrial glands, which contain no iron. Benzyl benzoate cleared fixed or unfixed uteri and allowed observation of the metrial glands and spots after delivery although the entire process required a few days and the specimens became hard and shriveled. The best and most convenient solution for clearing the rat uterus and for staining implantation sites was 2% NaOH or KOH.  相似文献

The high-performance liquid chromatographic separation of bile acid pyrenacyl esters with cyclodextrin-containing mobile phase is presented. Compared with conventional methods, inclusion chromatography gives much more satisfactory separation of derivatized bile acids in a short time. The application of this method to the separation of glycine-conjugated bile acids in human bile is also described.  相似文献

A new specimen of an araucarian cone,Araucaria nihongii, was found attached to the vegetative organs ofYezonia vulgaris, and is described asAraucaria vulgaris comb. nov. Thick branches show characteristic bark structure with lenticular patches. Secondary wood is usually araucarioid. Leaves are arranged helically on shoots, which are imbricate, appressed and fused to surface of the stem. External and anatomical features of leaves closely resembleBrachyphyllum. The seed cone is spherical with winged bracts and thin ovuliferous scales. One seed is borne per cone-scale complex. The seed coat and nucellus wall show typical araucarian structure. An araucarian plant that boreBrachyphyllum-like foliage and aEutacta-like seed cone was predicted by Harris in 1979. This reconstructed plant,Araucaria vulgaris, supports this theory and proves the presence of an extinct characteristic-form of the genus. A new section of the genus was proposed forAraucaria vulgaris. The floristic and structural differentiation of vegetation along the altitudinal gradient in four subalpine forests of different developmental stages on Mt. Fuji has been studied. Near the forest limit a micropattern of vegetation corresponding to the altitudinal zonation has been observed which elucidated the mechanisms of development of the vegetation zonation. As to early stages of vegetation development only two types can be distinguished: the volcanic desert above 1500 m and the pioneer forests below. As to later stages a differentiation of subzones includes from higher to lower altitudes: the Alnus maximowiczii, Betula ermanii, Abies veitchii and Tsuga diversifolia forests. Larix leptolepis and Sorbus americana ssp. japonica, appear as co-dominants in ecotonal communities between the principal subzones and are also important pioneers in early stages. Similarity analyses reveal that the upper subalpine Alnus-Betula forests can be regarded as early successional phases of the climax Abies-Tsuga forests of the lower subalpine zone. The regular arrangement of A. maximowiczii-B. ermanii-A. veitchii is studied along the gradient from the margin to the interior of the forest growing near the forest limit where locally favourable conditions prevail. Growth form, height growth, photosynthetic activity, seed supply, and seedling distribution of the three principal species have been compared, as well as biomass and production relations in contiguous forests of these species. The marginal Alnus type community is productive and disturbance-tolerant, and has a wide ecological and sociological amplitude along the gradient, while the central Abies community is accumulative and disturbance-intolerant, and has a narrower tolerance range, but is superior in competition under stable habitat conditions. A vegetation organization, ‘temporal multi-storeyed structure’, is suggested which means that a zonal pattern of vegetation within a climax region develops by successive replacement of successional species along an environmental gradient.  相似文献

Stimulation of synovial cell prostaglandin production by a factor obtained from casein-induced peritoneal polymorphonuclear (PMN) cells has been investigated. Both the extract and short time cultured medium of rat peritoneal PMN cells stimulate prostaglandin (PG)E2 production as well as collagenase production in the culture of rat synovial cells. PGE2 production by the cells in the presence of the PMN factor is much faster (5 to 24 hr) than collagenase production (24 hr or later, Biomedical Res. 3, 506-516, 1982). This stimulating factor is confirmed to be derived from PMN cells, based on the purification of the cells from peritoneal exudate cells by the Ficoll-Urographin method. Elution profile of the factor on gel filtration has indicated that both PGE2 and collagenase productions by synovial cells are stimulated by the same effluent fractions corresponding to molecular weights of 15,000 - 20,000 daltons and 30,000 - 40,000 daltons. These results suggest that PMN cells are involved in PG production as well as collagenase production in the inflamed tissue by stimulating connective tissue cells such as synovial cells.  相似文献
