The phospholipid content and phospholipase D activity in the leaves of two tobacco (Nicotiana tabacum L.) cultivars were investigated. These cultivars are characterized by different response to the infection with tobacco mosaic virus (TMV). In the infected leaves of a susceptible cv. Samsun, phospholipid content and phospholipase D activity did not change within seven days after TMV infection. The development of a hypersensitive response in the leaves of a resistant cv. Xanthy necrotic was not accompanied by a change in the total phospholipid content as compared to the noninfected leaves. However, the appearance of necrotic lesions and their subsequent expansion resulted in a steady decrease in the level of phosphatidylglycerol in infected leaves. At the same time, phosphatidic acid and diphosphatidylglycerol contents increased. Leaf zones remote from the regions of necrosis development were also characterized by an increased level of phosphatidic acid. There was a tendency for an increase in phospholipase D activity in both the sites of necrosis development and in the leaf regions remote from these sites. The changes in phosphatidic acid content were of similar nature, and therefore a relative increase in phosphatidic acid could result from the phospholipase D activity. This fact suggests a possible involvement of phospholipase D in the development of the hypersensitive response, and this suggestion is supported by a higher enzyme activity in the leaves of healthy plants of the resistant cultivar as compared to the susceptible one. Causes for the changes in the content of some phospholipids, as well as the physiological role of phospholipase D in the hypersensitive response are discussed.  相似文献

Abstract: Many articulated brachiopods experience marked life habit variations during ontogeny because they experience their fluid environment at successively higher Reynolds numbers, and they can change the configuration of their inhalant and exhalant flows as body size increases. We show that the extant brachiopod Terebratalia transversa undergoes a substantial ontogenetic change in reorientation governed by rotation around the pedicle. T. transversa′s reorientation angle (maximum ability to rotate on the pedicle) decreases during ontogeny, from 180 degrees in juveniles to 10–20 degrees in individuals exceeding 5 mm, to complete cessation of rotation in individuals larger than 10 mm. Rotation ability is substantially reduced after T. transversa achieves the adult lophophore configuration and preferred orientation with respect to ambient water currents at a length of 2.5–5 mm. We hypothesize that the rotation angle of T. transversa is determined mainly by the position of ventral and dorsal points of attachment of dorsal pedicle muscles relative to the pedicle. T. transversa shows a close correlation between the ontogenetic change in reorientation angle and ontogeny of morphological traits that are related to points of attachment of dorsal pedicle muscles, although other morphological features can also limit rotation in the adult stage. The major morphological change in cardinalia shape and the observed reduction of rotation affect individuals 2.5–10 mm in length. The position of ventral insertions of dorsal pedicle muscles remains constant, but contraction of dorsal pedicle muscles is functionally handicapped because dorsal insertions shift away from the valve midline, rise above the dorsal valve floor, and become limited by a wide cardinal process early in ontogeny (<5 mm). The rate of increase of cardinal process width and of distance between dorsal pedicle muscle scars substantially decreases in the subadult stage (5–10 mm), and most of the cardinalia shell traits grow nearly isometrically in the adult stage (>10 mm). T. transversa attains smaller shell length in crevices than on exposed substrates. The proportion of small‐sized individuals and population density is lower on exposed substrates than in crevices, indicating higher juvenile mortality on substrates prone to grazing and physical disturbance. The loss of reorientation ability can be a consequence of morphological changes that strengthen substrate attachment and maximize protection against biotic or physical disturbance (1) by minimizing torques around the pedicle axis and/or (2) by shifting energy investments into attachment strength at the expense of the cost involved in reorientation.  相似文献

Mitochondria from AA-cells are a unique model with separately finctioning 1st and 3rd points of energy coupling and with completely blocked 2nd coupling point. In vivo the terminal segment of the respiratory chain does not operate, cytochromes c and a+a3 remain oxidized, and dominating terminal oxidase appears to be a component inhibiting by salicylhydroxamate and bound with the respiratory chain via cytochromes b or ubiquinone pool. The blocking of the 2nd coupling point is due to firm and, probably, quantitative binding of AA with cytochromes b, especially with b559, and also to a partial denaturation of the complex III.  相似文献

Samples of Vi-antigen subjected to hydrolysis of different duration have been studied. As revealed in this study, 12-hour hydrolysis causes the depolymerization of the preparation. The fragments thus obtained have been found to possess Vi-specificity. The polymeric form of Vi-antigen contains adhesin differing in specificity from Vi-determinant. This component is absent in the low-molecular fraction of Vi-antigen. Adhesin is the binding element of the molecule of Vi-antigen and ensures its polymeric structure.  相似文献

Enterobacterial Vi-antigen inhibit the Escherichia coli-induced agglutination of red blood cells of guinea pigs and other animals, which indicates that Vi-antigen contains the admixture of adhesion associated with type I pili. The amount of adhesion contained in Vi-antigen can be determined from the degree to which its hemagglutination-inhibiting effect is manifested.  相似文献

The optimum conditions for the isolation and purification of the specific polysaccharide of group B meningococci have been developed. The advantages of the use of synthetic culture media for growing the initial bacterial culture have been demonstrated. The purified polysaccharides have been found to contain about 70% of sialic acid and less than 1% of protein and nucleic acid admixtures. The molecular parameters of group B polysaccharide depended on the growth phase of the bacterial culture. The most valuable culture was obtained at the exponential phase of growth. High serological activity and specificity of the polysaccharide in the passive hemagglutination test recommend it for studies on the development of diagnostic and prophylactic preparations.  相似文献

Spring oilseed rapeBrassica napus L. ssp.oleifera cv. HM-81 was transformed with TL-DNA of the Ri plasmid of the agropine strainAgrobacterium rhizogenes 15834. Selfed progenies (R2 and R3 generations) were studied for changes in values of growth characteristics and fatty acids contents. Transformants are ‘homozygous’ for TL-DNA. Both generations of transformants differed significantly from the nontransformed control plants in reduced length, lower number of pods per plant, lower total mass of seeds and the higher number of branches. The contents of palmitic, linoleic and linolenic acids were significantly higher in transformants when compared with the control. On the contrary, the contents of both stearic and oleic acids were in most of transformants significantly lower. Only traces of erucic acid (less than 0.05 % ) were found, both in transformed and nontransformed plants.  相似文献

The effects of valinomycin, gramicidins A and S, melittin and the protonophore 3,5-di-tert-butyl-4-hydroxybenzylidenmalononitrile on rat liver mitochondria and pea chloroplasts during active electron transport were studied. The canalogenes melittin and gramicidin S as well as gramicidin A and the protonophore increase the proton conductance of the inner mitochondrial membrane and chloroplast tylakoid membrane. The curve for the dependence of the canalogene effects on their concentration is S-shaped for both types of the organelles. Valinomycin reveals no protonophore activity and at high concentrations inhibits electron transport in both types of the coupling membranes. The uncoupling activity of gramicidin A and canalogenes and the inhibiting activity of valinomycin do not depend on the type of organelles when the concentration of these compounds is expressed as concentration in the membrane lipid matrix. At the same time the activity of the protonophore in chloroplasts is 6 times less than that in mitochondria. It is assumed that this difference in the protonophore activity is due to the differences in the mechanism of coupling of electron transport rather than to the peculiarities of lipid composition of mitochondria and chloroplasts. The lack of dependence of activity of peptide perminductors on the membrane lipid composition can probably be due to the fact that their effects is localized in the carbohydrate moiety of the lipid bilayer and does not involve the polar "heads" of the lipids.  相似文献
