The monoclonal antibodies to DNA-dependent RNA-polymerase of bacteriophage T7 have been obtained. Twenty of the obtained 500 clones have inhibited the enzyme activity. Three specificity groups were identified for seven of the clones supporting their affinity for different antigenic determinants.  相似文献

Maternal serum alpha-fetoprotein (MSAFP), human chorionic gonadotropin (hCG) and unconjugated oestriol (uE3) concentrations were measured in maternal serum samples from 21 pregnancies with neural-tube defects, 4 pregnancies with ventral wall defects (VWD) and 1662 unaffected pregnancies in women. These congenital malformations were confirmed by ultrasound scanning. The mean multiplate of the median (MoM) for AFP and uE3 was significantly different from the control values in cases of open NTD (AFP median MoM = 5.95, p < 0.001, uE3 median MoM = 0.2, p < 0.001), while hCG values did not differ from those of matched controls (hCG median MoM = 0.9). The biological basis of altered levels of uE3 in pregnancies with fetal NTDs is unclear.  相似文献

Promorphogenesis, egg cleavage, and gastrulation proceed in Macroposthonia nainitalensis similarly to other phytonematodes of the subclass Tylenchia. Before the onset of gastrulation, the greatest similarity was observed in a descendant of the micromere 2b, which separates macromeres of the third division. Deviations from the general line of development started in this species during embryonal organogenesis with the formation of an external epithelium with a special rough surface layer, which was absent in the embryos of other phytonematodes. In addition, during this period of acceleration, the digestive system developed, while peristaltic changes in body shape appeared at the tadpole stage. Later, somatic muscle development was enhanced in the first instar larvae of M. nainitalensis, while in the second instar larvae of Macroposthonia spp. somatic muscle cells were located at an acute angle to the longitudinal body axis. In addition, denticles--microchaetae appeared in the second to fifth instar larvae of these nematodes. All these features, which have not been observed in other phytonematodes of the subclass Tylenchia, help the peristaltic crawling of Macroposthonia spp. In contrast to other Tylenchia, the musculocutaneous sac did not develop in Macroposthonia spp. However, there are similarities in the development of the digestive and genital systems.  相似文献

Behavioral and neuropathological changes have been widely investigated in murine prion disease but stereological based unbiased estimates of key neuropathological features have not been carried out. After injections of ME7 infected (ME7) or normal brain homogenates (NBH) into dorsal CA1 of albino Swiss mice and C57BL6, we assessed behavioral changes on hippocampal-dependent tasks. We also estimated by optical fractionator at 15 and 18 weeks post-injections (w.p.i.) the total number of neurons, reactive astrocytes, activated microglia and perineuronal nets (PN) in the polymorphic layer of dentate gyrus (PolDG), CA1 and septum in albino Swiss mice. On average, early behavioral changes in albino Swiss mice start four weeks later than in C57BL6. Cluster and discriminant analysis of behavioral data in albino Swiss mice revealed that four of nine subjects start to change their behavior at 12 w.p.i. and reach terminal stage at 22 w.p.i and the remaining subjects start at 22 w.p.i. and reach terminal stage at 26 w.p.i. Biotinylated dextran-amine BDA-tracer experiments in mossy fiber pathway confirmed axonal degeneration and stereological data showed that early astrocytosis, microgliosis and reduction in the perineuronal nets are independent of a change in the number of neuronal cell bodies. Statistical analysis revealed that the septal region had greater levels of neuroinflammation and extracellular matrix damage than CA1. This stereological and multivariate analysis at early stages of disease in an outbred model of prion disease provided new insights connecting behavioral changes and neuroinflammation and seems to be important to understand the mechanisms of prion disease progression.Key words: prion disease, optical fractionator, neuropathology, behavioral changes, albino Swiss mice  相似文献

Four cases of cytogenetic prenatal diagnosis of fetuses with chromosomal aberrations are presented: (1) the Patau syndrome; (2) and (4) the Down syndrome; (3) the Klinefelter syndrome. Cordocentesis has been shown to be expedient for rapid and accurate determination of fetus karyotype. Indicative for cytogenetic examination were ultrasonic data, maternal age, the values of AFP, HGG and nonconjugated estreol in maternal serum. Comparison of ultrasonic examination of fetuses with the data on abortus autotopsia was undertaken. The results demonstrate importance of ultrasonic, cytogenetic, biochemical and morphological research in prenatal malformation diagnosis.  相似文献

4-[N-sodium-N-(5-ethyl-1,3,4-thiadiazol-2-yl)]- sulphanylamido-5-methoxy-1,2-benzoquinone selectively inhibiting lipid peroxidation (LPO) was used to study the hepatotoxic effect of carbon tetrachloride in vivo. It was found that inactivation of the liver microsomal oxidation system during the first few hours after CCl4 injection is due to covalent binding rather than LPO.  相似文献
