Homologous amino acid sequences of phospholipases A2 (PLA2) of snakes belonging to the families Elapidae, Viperidae, and Colubridae were considered in order to study the conservative and variable regions location. The PLA2 sequences were divided into two groups (taxons) according to the phylogenetic tree reconstructed from the pair similarity matrix. Results of the intergroup comparison were plotted to facilitate the identification of significant conservative and variable regions. It was shown that the results of the comparison between two phylogenetic groups of snake PLA2 did not much depend on the number of each group representatives and did not markedly change if one of the groups was represented by the single sequence. The knowledge of the number and location of conservative and variable regions and their dependence on the phylogenetic relations between compared taxa may be used to predict a synthetic peptide structure to obtain specific antibodies against PLA2 of one of these taxons. Such prediction is possible if there is a specific region conservative for one taxon but variable for two of them.  相似文献

Major glycolipids [monogalactosyldiacylglycerol (MGDG), digalactosyldiacylglycerol (DGDG), sulfoquinovosyldiacylglycerol (SQDG)) and phospholipids (phosphatidylcholine (PC), phosphatidylethanolamine (PE) and phosphatidylglycerol (PG)] as well as betaine lipid 1,2-diacylglycero-O-4'-(N,N,N-tri-methyl)-homoserine (DGTS) were isolated from Anfeltia tobuchiensis (Rhodophyta), Laminaria japonica, Sargassum pallidum (Phaeophyta), Ulva fenestrata (Chlorophyta) and Zostera marina (Embriophyta), harvested in the Sea of Japan. GC analysis of their fatty acid (FA) composition revealed that the n-6 polyunsaturated FAs (PUFAs) shared the most part of the sum of n-6 and n-3 PUFAs in PC and PE compared with glycolipids and PG. In algae, it was related to the prevalence of 20:4n-6 over 20:5n-3 in non-photosynthetic lipids. Percentage of n-6 PUFAs as well as the sum of n-3 and n-6 PUFAs decreased in the following sequence: PC-->PE-->PG. The saturation increased in the lines of MGDG-->DGDG-->SQDG and PC-->PE-->PG. PG was close to SQDG by the level of saturation. Distribution of C(18) and C(20) PUFAs in polar lipids depended on taxonomic position of macrophytes. Balance between C(18) and C(20) PUFAs was preferably shifted to the side of C(20) PUFAs in PC and PE that was observed in contrast to glycolipids and PG from L. japonica containing both series of FAs. The set of major FAs of polar lipid classes can essentially differ from each other and from total lipids of macrophytes. For example, MGDG was found to accumulate characteristic fatty acids 16:4n-3, 16:3n-3, 18:3n-6 and 18:4n-3, 20:3n-6 in U. fenestrata, Z. marina, L. japonica and S. pallidum, respectively.  相似文献

The ability of holothurian triterpene glycosides (cucumarioside A2-2 from Cucumaria japonica, cucumarioside G1 from C. fraudatrix, frondoside A from C. frondosa, and holotoxin A1 from Apostichopus japonicus) to form supramolecular lipid-saponin complexes was studied. TEM demonstrated that all the studied compounds form supramolecular cholesterol-saponin complexes (nanoparticles) in aqueous medium. The complexes formed by cucumarioside A2-2, holotoxin A1, and frondoside A had a tubular structure and fundamentally differed in the structure from the particles produced by cucumarioside G1. The morphology of the nanoparticles formed by cucumarioside A2-2, holotoxin A1, and cucumarioside G1 changed depending on the fraction of cholesterol in the lipid-saponin system; however, this pattern was not observed for frondoside A. At the same molar fraction of cholesterol in the lipid-saponin system, cucumarioside A2-2 formed the particles with the most pronounced tubular structure; the cholesterol-saponin complexes of holotoxin A1 had a less pronounced tubular structure, whereas the structure of frondoside A particles was extremely heterogeneous. Comparative analysis of the morphology of the described supramolecular complexes and specific structural features of the glycosides demonstrated that the structure of the corresponding nanoparticles depended on the degree of branching of the carbohydrate moiety in the glycoside molecule and the complexation with cholesterol was determined by the specific features of aglycone structure. Thus, the feasibility of producing new generation antigen carriers using the complexes in question was proved.  相似文献

The phospholipid (PL) and fatty acid (FA) composition of major membrane lipid constituents, phosphatidylcholine (PC) and phosphatidylethanolamine (PE), as well as the cholesterol/phospholipid (CL/PL) ratio were assayed in the muscles, gills and liver of the black plaice Pleuronectes (Liopsetta) obscura at different ambient temperatures (18, 9 and 0°C). PL and CL were shown to be actively involved in adaptation of the fish to changes in the seawater temperature. As temperature declines, the monounsaturated FA (MUFA) level increases while the polyunsaturated FA (PUFA) fraction in gills and liver PC and PE, on the contrary, decreases, resulting in diminished functional activity of the fish. However, in muscles this correlation is lacking. The PC and PE composition was shown to be organ- and ambient temperature-dependent. Major PC forms are saturated FA (SFA)/PUFA and MUFA/PUFA composed of a relatively small number of major molecular species. A temperature drop results in an increased SFA/PUFA level and decreased MUFA/PUFA and PUFA/PUFA levels in muscles and gills, and this may promote a drop in the viscosity of the outer lipid monolayer of membranes and in their functional activity. In contrast to PC, the PE composition in all organs tested is characterized by a decrease in the SFA/ PUFA level and an increase in MUFA/PUFA and PUFA/PUFA levels. Such changes promote the retention of functional activity of the inner lipid monolayer of membranes and are not synchronized with rearrangements in their outer monolayer. Due to intermolecular transfer of acyl radicals at a constancy of their composition, functional rearrangement of the lipid matrix appears to be achieved through changes in the membrane viscosity. Our data support the idea that different adaptation strategies in fish are driven by certain sets of PL molecular species.  相似文献

Major glyco- and phospholipids as well as betaine lipid 1,2-diacylglycero-O-4'-(N,N,N-tri-methyl)-homoserine (DGTS) were isolated from five species of marine macrophytes harvested in the Sea of Japan in summer and winter at seawater temperatures of 20-23 and 3 degrees C, respectively. GC and DSC analysis of lipids revealed a common increase of ratio between n-3 and n-6 polyunsaturated fatty acids (PUFAs) of polar lipids from summer to winter despite their chemotaxonomically different fatty acid (FA) composition. Especially, high level of different n-3 PUFAs was observed in galactolipids in winter. However, the rise in FA unsaturation did not result in the lowering of peak maximum temperature of phase transition of photosynthetic lipids (glycolipids and phosphatidylglycerol (PG)) in contrast to non-photosynthetic ones [phosphatidylcholine (PC) and phosphatidylethanolamine (PE)]. Different thermotropic behavior of these lipid groups was accompanied by higher content of n-6 PUFAs from the sum of n-6 and n-3 PUFAs in PC and PE compared with glycolipids and PG in both seasons. Seasonal changes of DSC transitions and FA composition of DGTS studied for the first time were similar to PC and PE. Thermograms of all polar lipids were characterized by complex profiles and located in a wide temperature range between -130 and 80 degrees C, while the most evident phase separation occurred in PGs in both seasons. Polarizing microscopy combined with DSC has shown that the liquid crystal - isotropic melt transitions of polar lipids from marine macrophytes began from 10 to 30 degrees C mostly, which can cause the thermal sensitivity of plants to superoptimal temperatures in their environment.  相似文献

This study examines the fatty radical (FR) composition and heat-induced crystalline to liquidcrystalline phase transitions of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) from the gills, hepatopancreas, gonads, and muscle of the tanner crab Chionoecetes bairdi, which was collected in the summer at a near-bottom water temperature of 2.8°C. The location of the PC and PE thermograms below 2.8°C indicates the functionally optimal liquid crystalline state of the membrane lipid matrix. The proximity of the thermogram profiles of PC and PE from the different organs and tissues of C. bairdi and significant overlapping of the temperature areas of transitions (symbatic behavior) correlate with a similar composition of major FR and their total parameters in PC and PE. The obtained data point to the effective adaptation of the bairdi crab to low water temperatures and to the need for adaptive changes in the FR composition or change of habitat with increasing temperature. The thermotropic behavior of muscle PC, in which the greater part of the thermogram is in the temperature range from 2.8 to 32°C, suggests a potential for the tanner crab to adapt to increased temperatures.  相似文献

The protein actinoxanthin (molecular weight 10,300) crystallizes in space group P212121, with cell dimensions a = 30.9 Å, b = 48.8 Å, c = 64.1 Å, and z = 4. The three-dimensional structure of actinoxanthin at 4-Å resolution was determined by x-ray methods on the basis of experimental data from the native protein and five isomorphous derivatives. At the stage of solving the phase problem, the heavy atoms in the derivatives were located using direct methods. The actinoxanthin molecule can be described as an oblate ellipsoid with approximate dimensions 20 × 30 × 40 Å and consists of two different sizes of folded units separated by a well-defined cleft. The larger unit, including the N- and C-terminals of the protein chain, is characterized by a significant content of β-sheet structure. The smaller unit, containing two deca- and hexapeptide cycles closed by disulfide bonds, has a mainly irregular structure.  相似文献

The development of cryopreservation methods for embryonic cells and larvae of sea animals offers a great potential for marine biotechnology. Larval cells of bivalves and sea urchins were frozen to −196 °C using traditional cryoprotectants (Me2SO and trehalose) and the cryoprotective mixture developed by us. In addition to Me2SO and trehalose, this mixture contained an exogenous lipid extract from mussel tissues and antioxidants. A positive effect of antioxidants (α-tocopherol acetate, ascorbic acid or echinochrome, the quinoid pigment of sea urchins) on cell viability became significant only in the presence of exogenous lipids. Antioxidants added to cryoprotective mixtures did not reveal visible cryoprotective activity when used separately. To better understand the mechanism of the protective effect of exogenous lipids on cell membranes of sea animals, a comparative analysis of the fatty acid (FA) composition of total lipids in larval cells before and after freezing was carried out using a gas–liquid chromatography. The results indicate that freezing–thawing has direct effects on the FA composition of major lipid classes in marine invertebrate cells, and these effects can vary depending on the provenance of the cells. We have found that (I) both cell viability and the FA profile of cell lipids after cryopreservation depend on the cryoprotectants used; (II) an amount of saturated, monoenic and polyenic FAs changes significantly after cryopreservation. We assume that the addition of the exogenous lipid extract in form of liposomes could promote a renewal of disturbance areas and prevent from membrane damages during freezing–thawing.  相似文献

Some physicochemical properties of glycoglycerolipids (monogalactosyldiacylglycerol, digalactosyldiacylglycerol and sulfoquinovosyldiacylglycerol) from the sea algae Laminaria japonica, as well as their ability to become incorporate into immunostimulating complexes (ISCOMs), used as a delivery system of microbial and tumor antigens in vesicular form, were studied. These glycolipids were found to differ essentially in fatty acid composition, unsaturation index and thermotropic behavior. The possibility of ISCOM modification by embedding the glycolipids studied instead of a phospholipid component in vesicles was shown. A preliminary research of the immunogenicity of the pore-forming protein from Yersinia pseudotuberculosis in modified (by monogalactosyldiacylglycerol) and typical (egg phosphatidylcholine) ISCOMs did not reveal a significant enhancement of immune response in comparison with that of isolated protein.  相似文献
