One hundred and three specimens of juvenile and mature one-tentacle, relatively small-sized hydromedusae of unclear systematic position were found in the zooplankton collections from the Arctic Ocean. Examination of the mature specimens led to the conclusion that their morphology fits with the last diagnoses of the genus Rhabdoon (Anthomedusae) and that they belong to the species named here as R. reesi. The study of the morphology and cnidome composition of immature medusae suggested them to be young stages of the same species. The comparative study of these juvenile forms with Yakovia polinae, the medusae described previously from the Arctic as a new genus and new species, suggested Y. polinae to be a junior synonym of R. reesi. The distribution of R. reesi in the Arctic Ocean is mapped according to recent and historical records.  相似文献

Abundance and reproductive biology (gonad maturation and egg production) of the Arctic copepod Calanus glacialis were studied in the Laptev Sea and adjacent Arctic Ocean in September 1993 and from July to September 1995. Both abundance and reproductive activity were subject to strong spatial and seasonal variability, which was related to the ice cover, feeding conditions and circulation pattern. Maximum abundance of the C. glacialis population was generally confined to the outer shelf and slope with depths between 50 and 1000 m. During both cruises, highest egg production rates and largest number of young copepodite stages were observed in the eastern Laptev Sea, where the development of the C. glacialis population seems to follow the opening of the “Siberian Polynya”. In the western part, which is usually covered by pack ice, females were all immature, and no young stages were found. However, females responded quickly to a temporary opening of the ice there in 1995 and spawned. Starvation experiments showed that food-independent reproduction fuelled by internal energy resources was at least partly responsible for relatively high egg production rates at low ambient food concentrations. Zooplankton composition and distribution were investigated on the Laptev Sea shelf, over the continental slope and in the adjacent deep Nansen Basin during the joint German-Russian expedition “Arctic 93” with RV Polarstern and Ivan Kireyev in August/September 1993. In the shelf area biomass decreased from west to east with the lowest values in the area influenced by the Lena river runoff. A gradual increase of biomass from the shallow to the deep area correlated with water depth. Total biomass ranged between 0.1 and 1.5 g m−2 on the shelf and 4.7 and 7.9 g m−2 in the adjacent Nansen Basin. On the shelf Calanus glacialis/finmarchicus dominated overall. The contribution of brackish-water taxa was low in the west, where high salinity and southward currents from the Arctic Basin supported a marine neritic community, but on the southern and eastern Laptev shelf, in the areas of freshwater influence, brackish-water taxa contributed up to 27% of the total biomass. On the slope and in deep areas a few large Arctic copepod species, Calanus glacialis, C. hyperboreus and Metridia longa, composed the bulk of biomass and determined the pattern of its vertical distribution. The export of Calanus species from the Nansen Basin onto the Laptev shelf appears to be of great importance for the shelf communities. In turn, the eastern outer shelf and slope area of the Laptev Sea are thought to have a pronounced effect on the deep basin, modifying the populations entering the central Arctic. A method was developed for the production and purification of biologically active recombinant human interferon α-2b (rhIFN α-2b) synthesized by expression in Nicotiana benthamiana plants. A gene construct containing a modified hIFN α-2b gene was cloned in two vectors based on tobacco mosaic virus driven by an actin promoter from Arabidopsis thaliana (pA-IFN-A) and cauliflower mosaic virus driven by a 35S promoter (pA-IFN-S). The expression vectors were introduced into the plant cells by agroinfiltration. The maximum rates of synthesis achieved in the case of pA-IFN-A and pA-IFN-S 5 days after agroinfiltration were determined to be 200 and 20 mg per 1 kg of fresh leaves, respectively. The recombinant hIFN α-2b synthesized in the plant showed high antiviral and antitumor activity comparable with that of commercial drug.  相似文献

Therapeutic monoclonal antibodies (TMA) provide an important means for treating diseases that were previously considered untreatable. Currently more than 40 full-size TMAs created primarily based on immunoglobulin G1 are widely used for treating various illnesses. Glycosylation of TMA is among other numerous factors that affect their biological activity, effector functions, immunogenicity, and half-life in the patient’s serum. The importance of carbohydrate residues for activity of human serum immunoglobulin and TMA produced in animal cells is considered in this review, with emphasis given to N-glycosylation of the Fc fragment of the antibody.  相似文献

Reproduction of the dominant Antarctic copepod Calanus propinquus was studied in February–April, 1989 aboard the R.V Dmitry Mendeleev during cruise N° 43 to the Weddell Sea. Single females were kept at 0 °C in the laboratory for 56 days with abundant food concentration (above 300 µg C l–1 of Platymonas viridis). Females released clutches at night at 2–3 day intervals. Most clutches contained from 10 to 40 eggs, mean 37.3 eggs female–1. Average carbon content of an egg was 0.37 ± 0.05 µg C. The maximum daily egg production rate of 30–50 eggs female–1 d–1 was observed for the first 3 days of the laboratory incubation, corresponding to 3.7–6.2% body C. The state of gonadal development of females showed the decline of the reproductive season in late February. The data suggest that egg laying in the region under study starts in December and lasts until March. The state of ovarian maturation, changes in vertical distribution and biochemical body composition of females suggest the possibility of two-year life cycle in C. propinquus in the southern Weddell Sea.  相似文献

Plant biosimilars of anticancer therapeutic antibodies are of interest not only because of the prospects of their practical use, but also as an instrument and object for study of plant protein glycosylation. In this work, we first designed a pertuzumab plant biosimilar (PPB) and investigated the composition of its Asn297-linked glycan in comparison with trastuzumab plant biosimilar (TPB). Both biosimilars were produced in wild-type (WT) Nicotiana benthamiana plant (PPBWT and TPB-WT) and transgenic ΔXTFT N. benthamiana plant with XT and FT genes knockout (PPB-ΔXTFT and TPBΔXTFT). Western blot analysis with anti-α1,3-fucose and anti-xylose antibodies, as well as a test with peptide-N-glycosidase F, confirmed the absence of α1,3-fucose and xylose in the Asn297-linked glycan of PPB-ΔXTFT and TPB-ΔXTFT. Peptide analysis followed by the identification of glycomodified peptides using MALDI-TOF/TOF showed that PPB-WT and TPB-WT Asn297-linked glycans are mainly of complex type GnGnXF. The core of PPB-WT and TPB-WT Asn297linked GnGn-type glycan contains α1,3-fucose and β1,2-xylose, which, along with the absence of terminal galactose and sialic acid, distinguishes these plant biosimilars from human IgG. Analysis of TPB-ΔXTFT total carbohydrate content indicates the possibility of changing the composition of the carbohydrate profile not only of the Fc, but also of the Fab portion of an antibody produced in transgenic ΔXTFT N. benthamiana plants. Nevertheless, study of the antigen-binding capacity of the biosimilars showed that absence of xylose and fucose residues in the Asn297-linked glycans does not affect the ability of the glycomodified antibodies to interact with HER2/neu positive cancer cells.  相似文献

The size structure of the population, morphology of the reproductive system, and gonad maturation of the chaetognath Eukrohnia hamata (Möbius, 1875) (Chaetognatha) were studied for the first time from zooplankton samples collected in Fram Strait and the deep Eurasian basin of the Arctic Ocean north of the Kara and Laptev seas in the summers of 1995 and 1997. We describe the gonad morphology and the progress of gonad maturation in male and female specimens and propose a system for classification of gonad maturity stages. During the Arctic summer, the population was represented by individuals ranging in size from 2 to 37 mm. The newly hatched young (2.0–3.5 mm long) and juveniles (4.0–12.5 mm long) dominated numerically (30 and 50% of the population, respectively). Our results indicate that E. hamata successfully matures and reproduces in the Arctic basin, but mature individuals make up less than 1% of the population in the summer against the background of juvenile abundance.  相似文献

This paper describes design of the new ultra sensitive sensor system for fluorescence detection applications. System comprises two units: optical spectra separation unit and detection unit. Optical unit of the sensor performs spatial spectra separation of signal from the laser excited fluorescence, and resulting spectra is collected in the detection part of the system. Optical part is built using diffraction grating as spectra separation element. Detection part comprises 32-channel photomultiplier tube working in single photon counting mode with our 32-channel amplifier. Using single photon detection technique and specific signal processing algorithms for collected data, the proposed system allows to achieve unique combination of characteristics--high sensitivity, high detection speed and wide linearity dynamic range comparing to existing commercial instruments. DNA sequencing experiments with new sensor as detection device, and using two types of lasers (Ar-ion and Nd-YAG) were carried out, yielding sequencing traces which have quality factor of 20 for read lengths as long as 650 base pairs.  相似文献

Virtually all recombinant proteins are now prepared using fusion domains also known as “tags”. The use of tags helps to solve some serious problems: to simplify procedures of protein isolation, to increase expression and solubility of the desired protein, to simplify protein refolding and increase its efficiency, and to prevent proteolysis. In this review, advantages and disadvantages of such fusion tags are analyzed and data on both well-known and new tags are generalized. The authors own data are also presented.  相似文献
