Purification of recombinant proteins with an example of tumor necrosis factor thymosin-α1

Hybrid protein, cancer necrosis factor thymosin-α1 (TNF-T), when synthesizing in strain-producer of Escherichia coli SG200-50 with plasmid pThy315, was a part of “inclusion bodies” mostly in the form of a high-molecular complex with other proteins due to the S-S bonds formation. An approach of purification of TNF-T has been proposed, which is based on the destruction of the complex in the presence of sodium dodecylsulfate (DDS-Na) and dithiotreitol (DDT) followed by gel-filtration on Sephadex G-100 and renaturation by ultrafiltration on hollow fibers. The method allows the isolation of electrophoretically homogeneous TNF-T containing no DDS-Na and having high cytotoxic activity against cancer cells of mouse adenocarcinome L-929. The yield of TNF-T achieved 80% relative its content in biomass and 30% relative the total protein.     

Oxygen binding of myoglobins of diving animals]

When studying oxygen binding by myoglobins of diving animals it is shown that half-saturation of myoglobins obtained from the muscles of animals who can stop the external respiration for a long period of time occurs at oxygen strength of 0.62-0.67 mm Hg. Such indices of oxygenation of respiratory hemoprotein of muscles are characteristic of most mammals. Temperature being decreased from 37 to 15 degrees C, myoglobin affinity to oxygen considerably increases.

Morbidity and prophylaxis of hemorrhagic fever with renal syndrome in the Republic of Bashkortostan

The brief retrospective analysis of morbidity, epizootological data and preventive measures with respect to haemorrhagic fever with renal syndrome (HFRS) during the whole period of its registration on the territory of the Republic of Bashkortostan along with the structure of HFRS morbidity during epidemic seasons of 1998 and 1999 is presented. The main causes of decreased HFRS morbidity in 1998, its rise in 1999 as well as prognosis for the year 2000 were detected. The level of HFRS morbidity among the population was found to be directly related to the number of rodents and their infection rate. The basic strategy of epidemic control interventions has been proposed.     

The dependence of the antibacterial effect of the polycationic peptide warnerin on the energy state of target cells

The bactericidal effect of the polycationic peptide warnerin, produced by Staphylococcus warneri IEGM KL-1, was found to depend on the energy state of susceptible Staphylococcus epidermidis cells. The pre-treatment of these cells with compounds that diminish the proton-motive force of plasma membranes enhanced cell tolerance to warnerin. The components deltapsi and deltapH of the membrane proton potential influenced the antibacterial activity of warnerin in different ways. In particular, the antibacterial activity of warnerin decreased when the electric component of the proton-motive force of target membranes declined.     

Raoultella planticola, a new strain degrading 2,4,5-trichlorophenoxyacetic acid

A new strain that degrades the herbicide 2,4,5-trichlorophenoxyacetic acid (2,4,5-T) was isolated from soil, which was exposed to factors related to the petrochemical industry. According to its physiological, biochemical, cultural, and morphological traits, together with the sequence of the 16S rRNA gene, the strain was identified as Raoultella planticola 33-4ch. The strain could consume 2,4,5-T as a sole source of carbon and energy. The amount of 2,4,5-T in the culture medium decreased by 51% after five days of incubation. Raoultella planticola 33-4ch consumes 2,4,5-T to produce 4-chlorophenoxyacetic, phenoxyacetic, and 3-methyl-2,6-dioxo-4-hexenoic acids.     

Electroneuromyographic control of the efficacy of treatment of vertebral syndromes in aged patients

We studied the electroneuromyographic (ENMG) indices in aged patients with cervical vertebral syndromes. The main group of 116 patients was treated with manipulation physical therapy, while the second group of 62 patients was subjected to traditional curative procedures. In both patient groups, modifications of the EMG pattern of the forelimb muscles, a decrease in the conduction velocity via then. medianus, and a drop in the amplitude of M responses of themm. tenar et hypotenar were observed on the injured side. After a course of manipulation therapy, a clear trend toward normalization of the above indices was observed in the patients with a reflex pattern of the syndrome. This trend was less intensive in the subgroup with the root syndrome pattern. In patients treated according to the traditional schemes, positive shifts in the ENMG indices were rather small, in spite of some improvement of their clinical state. We emphasize that electrophysiological control of the efficacy of treatment in the cases of vertebral syndromes is extremely desirable, especially in groups of aged patients.
