The anaphylatoxin, complement 5a (C5a), plays a key role in mediating various inflammatory reactions following complement activation. Several investigators have reported that C5a receptor (C5aR) is expressed in non-myeloid cells under certain conditions or in different cell lines. In our study, the abundance of C5aR-positive myeloid cells in rats depended on the organs examined. C5aR was usually expressed at the site of exposure to pathogens, such as in salivary gland or lung, and was up-regulated in liver in the inflammatory state induced by lipopolysaccharide (LPS) administration. Furthermore, the increased expression of C5aR antigen was not accompanied by an increase in C5aR mRNA in Kupffer cells following LPS challenge. 相似文献

Somatic mutations of the fibroblast growth factor receptor 3 (FGFR3) gene were detected by peptide nucleic acid (PNA)-mediated real-time PCR clamping. Mutation was detected in negative control containing only wild-type DNA due to a misincorporation of dNTPs to PNA binding sites when the amount of template DNA was decreased to 1 ng. Thus, the amount of template DNA was critical determinant of the assay sensitivity in PNA-mediated PCR clamping. Assay conditions were optimized to detect FGFR3 mutations in exons 7, 10, and 15, at a concentration of more than 1% mutated DNA using 50 ng of genomic DNA as the template. Mutations were detected in 12 of 13 (92.3%) tumor tissues and 11 of 13 (84.6%) urine samples from patients with superficial bladder cancer, while no mutations were detected in tissues and/or urine samples from patients with muscle-invasive bladder cancer or chronic cystitis. 相似文献

The levels of indole-acetic acid (IAA), gibberellic acid1 (GA1), trans-zeatin (Z) and trans-zeatin riboside (ZR) in seedless fruits of parthenocarpic tomato (Lycopersicon esculentum Mill. cv. Rarkuna First) were analysed using 13C6-IAA, 2H2-GA1, 2H5-Z and 2H5-ZR, as internal standards by liquid chromatography–mass spectrometry. Fruits were sampled at 6 cm in diameter (referred to as 6-cm-fruit) and 8 cm (8-cm-fruit, mature green stage) and separated into pericarps, partitions and locule tissues. The pericarps and partitions were centrifuged for the collection of apoplast (AP) solution (sap outside a cell) and symplast (SP) solution (sap within a cell). IAA concentrations of the pericarps and partitions were higher in 8-cm-fruit than in 6-cm-fruit. In the partitions, IAA concentrations of SP solution were higher than those of AP solution in both 6- and 8-cm-fruit. The SP solution of the partitions in 6-cm-fruit had the highest concentration of Z (4.6 pmol/g fresh weight) and was 2.7 times than the AP solution, while in the pericarps Z concentrations were the same level in AP and SP solution. The ZR concentration in locule tissues in 6-cm-fruit (55 pmol/g fresh weight ) was the highest of all parts. The results suggest that the sites of synthesis may be the SP of partitions for IAA and Z, and locules for ZR. 相似文献

Performance status (PS) frequently improves occurs in cancer patients who have been infused with their own lymphokine‐activated killer T cells (LAK‐T). In the present study, a culture supernatant of LAK‐T (LAK‐T sup) administered to 8‐week‐old rats caused neurogenesis as evidenced by increased 5‐ethynyl‐2′‐deoxyuridine staining of brain tissues. Intravenous injection of granulocyte‐macrophage colony stimulating factor (GM‐CSF), a major cytokine in LAK‐T sup, had a similar effect. Furthermore, LAK‐T sup induced Ca++ increase in rat hippocampal brain slices that was detected in neuronal cells by emission of Fluo‐8 NW at 520 nm. The same effect was observed with an rGM‐CSF solution. GM‐CSF may activate neuronal cells by stimulating the glial cells that surround and attach to them. If so, GM‐CSF and LAK‐T sup may improve the motor neurons of patients with amyotrophic lateral sclerosis. The neurogenerative effect of GM‐CSF in LAK‐T sup may also help improve brain function in aged adults including those with dementia such as Alzheimer's disease. 相似文献

Glycogen granules were isolated from the photosynthesizing cellsof purple sulfur bacterium, Chromatium strain D, and the chemicalstructure was studied by enzymic hydrolysis using -and ß-amylasesand pullulanase. The end-group assay of the bacterial glycogenby periodate oxidation gave the following analytical data: (average chain length), 11; ICL (interiorchain length), 3; and ECL (exterior chain length), 7. We concludethat the Chromatium glycogen is a glycogen- and amylopectin-type-glucan. Intracellular accumulation of glycogen granules inthe bacterial cells harvested at different growth stages wasexamined by electron microscope observation.

Salivary gland stem/progenitor cells are thought to be present in intercalated ductal cells, but the fact is unclear. In this study, we sought to clarify if stem/progenitor cells are present in submandibular glands using colony assay, which is one of the stem cell assay methods. Using a low-density culture of submandibular gland cells of neonatal rats, we developed a novel culture system that promotes single cell colony formation. Average doubling time for the colony-forming cells was 24.7 (SD=+/-7.02)h, indicating high proliferative potency. When epidermal growth factor (EGF) and hepatocyte growth factor (HGF) were added to the medium, the number of clonal colonies increased greater than those cultured without growth factors (13.2+/-4.18 vs. 4.5+/-1.73). The RT-PCR and immunostaining demonstrated expressing acinar, ductal, and myoepithelial cell lineage markers. This study demonstrated the presence of the salivary gland stem/progenitor cells that are highly proliferative and multipotent in salivary glands. 相似文献

Cell electrophoretic mobility of cultured melanoma cells or rat erythrocytes decreased with time after X-irradiation. Addition of tetravalent concanavalin A or divalent succinyl-concanavalin A before (not after) irradiation, completely blocked the mobility reduction in greater concentrations than 5 μg/l.At 5 μg/1 only 3.7 · 103 concanavalin A molecules bound to receptors per cell, while 4.18 · 107 molecules/cell bound at saturating concentrations. Preincubation with concanavalin A at 37°C was effective even when the cells were treated with α-methylmannoside immediately after irradiation. At low temperature, however, concanavalin A was not effective despite a sufficient amount of bound 125I-labelled concanavalin A. Treatment with α-methylmannoside following the binding of concanavalin A at 37°C before irradiation inhibited the concanavalin A effect depending on temperature. The residual amount of bound lectin could not account for the temperature dependence. The amount of sialic acid (the main charged substance) was not altered by X-irradiation with or without the lectin. Divalent succinyl-concanavalin A was also effective in blocking the radiation effect on electrophoretic mobility. These results seem to suggest that binding of a very small amount of concanavalin A without causing cell agglutination or clustering of its receptors, induces some alteration in the conformation of receptor glycoprotein, which blocks the internalization of acidic sugar residues by subsequent irradiation. 相似文献

Lectin activity in immature pods from 30 strains of winged bean was investigated. Most of the lectin activity occurred in green shells and a small portion in immature seeds. Hemagglutinating activity of green shells was classified into four groups, according to the agglutination of trypsinized human type A, B, and O erythrocytes. Extracts from green shells of four representative strains which showed different hemagglutination patterns gave different elution profiles of lectin activity from ion exchange columns. Four types of lectin activity with different blood group specificities were apparently found in green shells. 相似文献
